Among different mechanisms of salt resistance, regulation of ion distribution among various tissues and intracellular compartmentation are of great importance. In this study, we investigated the effects of salt stress on growth, photosynthesis, and Na(+) accumulation and distribution in leaf apoplast and symplast of two canola (Brassica napus L.) cultivars (NYY 1 and BZY 1). The results showed that the declines in shoot dry mass, leaf water potential and net photosynthetic rate of BZY 1 (salt sensitive) were higher than those of NYY 1 (salt resistant) in response to salt stress. Stomatal limitation to photosynthesis was mainly affected under moderate salinity, whereas the reduction in assimilation rate under severe salt stress was due to both stomatal and non-stomatal limitations. We also found that more Na(+) was distributed to leaf veins in NYY 1 than in BZY 1; simultaneously, less Na(+) accumulated in the leaf blade in NYY 1 than in BZY 1. The percentage of Na(+) in the leaf symplast in NYY 1 was markedly lower than that in BZY 1. Also, Na(+) diffusion in leaves through apoplastic and symplastic pathways of BZY 1 was stronger than that in NYY 1, and the transpiration rate in BZY 1, especially at the leaf edges, decreased more than in NYY 1. Our results showed that NYY 1 accumulated less Na(+) in the shoot, especially in leaf blades, and confined Na(+) to the apoplast to avoid leaf salt toxicity, which could be one reason for the higher resistance of NYY 1 than BZY 1 plants to salt stress.  相似文献

AM真菌是土壤生态系统中重要的微生物类群,能与陆地生态系统中80%以上的高等植物建立共生体系。目前,AM真菌在维持草原生态系统稳定性中的功能已经成为生态学研究的热点问题之一。基于此,从植物个体、种群、群落和生态系统等不同层次探究AM真菌在维持植物群落多样性和草原生态系统稳定性中的功能。分析发现在个体水平上,AM真菌对宿主植物具有促生效应、抑制效应或中性效应。在种群水平上,分析AM真菌对不同宿主植物吸收土壤矿质营养的分配和调控策略,围绕构成草原植被的两大组成成分:牧草和有毒植物,论述AM真菌对植物种群增长和衰败的调控机制,并从草原植物群落的物种多样性和稳定性角度,探讨AM真菌与植物群落之间的相关性。在生态系统水平上,围绕AM真菌对草原生态系统的演替和退化草原的修复等展开论述,以期为利用AM真菌开展草原生态系统保护和恢复治理提供理论依据,并对草原菌根生态学领域未来的研究进行展望。  相似文献

Background

Myasthenia gravis (MG) is an autoimmune disease in which 90% of patients have autoantibodies against the muscle nicotinic acetylcholine receptor (AChR), while autoantibodies to muscle-specific tyrosine kinase (MuSK) have been detected in half (5%) of the remaining 10%. Recently, the low-density lipoprotein receptor-related protein 4 (LRP4), identified as the agrin receptor, has been recognized as a third autoimmune target in a significant portion of the double sero-negative (dSN) myasthenic individuals, with variable frequency depending on different methods and origin countries of the tested population. There is also convincing experimental evidence that anti-LRP4 autoantibodies may cause MG.

Methods

The aim of this study was to test the presence and diagnostic significance of anti-LRP4 autoantibodies in an Italian population of 101 myasthenic patients (55 dSN, 23 AChR positive and 23 MuSK positive), 45 healthy blood donors and 40 patients with other neurological diseases as controls. All sera were analyzed by a cell-based antigen assay employing LRP4-transfected HEK293T cells, along with a flow cytofluorimetric detection system.

Results

We found a 14.5% (8/55) frequency of positivity in the dSN-MG group and a 13% frequency of co-occurrence (3/23) in both AChR and MuSK positive patients; moreover, we report a younger female prevalence with a mild form of disease in LRP4-positive dSN-MG individuals.

Conclusion

Our data confirm LRP4 as a new autoimmune target, supporting the value of including anti-LRP4 antibodies in further studies on Myasthenia gravis.  相似文献

The Sika deer antler is well known for its unique ability to regenerate repeatedly and grow rapidly. Furthermore, it is a precious traditional Chinese medicine and has been widely used for more than 20 centuries. The major bioactive components within the antlers are water-soluble proteins, polypeptides, and free amino acids. Many studies have shown that water-soluble antler extracts play pivotal roles in wound healing, immune system modulation, anti-oxidation, and anti-inflammation. However, the exact effects on chondrocytes are still largely unknown. In this study, we prepared fresh, aqueous extracts from growing deer antlers in a rapid growth stage. We isolated the chondrocytes from neonatal mouse rib cartilage and investigated the effects of antler extracts on chondrocyte viability. We also used the RNA-Seq method to analyze the gene expression pattern under antler extract treatment. We demonstrated that fresh extracts from Sika deer antlers in a rapid growth stage significantly promoted chondrocyte viability and kept chondrocytes proliferating continuously, while blocking maturation and further differentiation. Additionally, our results indicated that antler extracts might serve as a potent anti-oxidant, anti-inflammatory agent, and immune modulator to boost the abilities of chondrocytes against oxidative, inflammatory, and immune stresses. Thus, this study has greatly deepened our current knowledge of the molecular control of antler extracts on chondrocytes. It has also shed light on possible new strategies to further prevent and treat diseases of cartilage and other related diseases.  相似文献

We hypothesize that over-expression of transporters associated with antigen processing (TAP1 and TAP2), components of the major histocompatibility complex (MHC) class I antigen-processing pathway, enhances antigen-specific cytotoxic activity in response to viral infection. An expression system using recombinant vaccinia virus (VV) was used to over-express human TAP1 and TAP2 (VV-hTAP1,2) in normal mice. Mice coinfected with either vesicular stomatitis virus plus VV-hTAP1,2 or Sendai virus plus VV-hTAP1,2 increased cytotoxic lymphocyte (CTL) activity by at least 4-fold when compared to coinfections with a control vector, VV encoding the plasmid PJS-5. Coinfections with VV-hTAP1,2 increased virus-specific CTL precursors compared to control infections without VV-hTAP1,2. In an animal model of lethal viral challenge after vaccination, VV-hTAP1,2 provided protection against a lethal challenge of VV at doses 100-fold lower than control vector alone. Mechanistically, the total MHC class I antigen surface expression and the cross-presentation mechanism in spleen-derived dendritic cells was augmented by over-expression of TAP. Furthermore, VV-hTAP1,2 increases splenic TAP transport activity and endogenous antigen processing, thus rendering infected targets more susceptible to CTL recognition and subsequent killing. This is the first demonstration that over-expression of a component of the antigen-processing machinery increases endogenous antigen presentation and dendritic cell cross-presentation of exogenous antigens and may provide a novel and general approach for increasing immune responses against pathogens at low doses of vaccine inocula.  相似文献

The fluorescence properties of the single tryptophanyl residue present in amicyanin from Thiobacillus versutus are very similar to those of azurin from Pseudomonas aeruginosa and other mononuclear blue copper proteins. The emission maximum is well structured and centered at 318 nm. The quantum yield is strongly affected by the presence of copper, the removal of which is accompanied by a more than sixfold increase in fluorescence, without change in shape. The fluorescence decay of holo-amicyanin is heterogeneous with a longer component of 5.7 ns and a shorter one of 0.7 ns accounting for 90% of the total emitting molecules. Copper-free amicyanin shows instead a single exponential decay (3.3 ns) of intrinsic fluorescence. This lifetime decreases as the temperature increases as does the longer lifetime component of holoamicyanin.  相似文献

Brassinosteroids (BRs) regulate various agronomic traits such as plant height, leaf angle, and grain size in rice (Oryza sativa L.); thus, BR signaling components are promising targets for molecular rational design. However, genetic materials for BR-signaling genes or family members remain limited in rice. Here, by genome editing using clustered regularly interspaced short palindromic repeats (CRSPR)/Cas9 tools, we generated a panel of single, double, triple, or quadruple mutants within three BR signaling gene families, including GSK3/SHAGGY-LIKE KINASE1 (GSK1)–GSK4, BRASSINAZOLE-RESISTANT1 (OsBZR1)–OsBZR4, and protein phosphatases with kelch-like (PPKL)1–PPKL3, under the same background (Zhonghua11, japonica). The high-order mutants were produced by either simultaneously targeting multiple sites on different genes of one family (GSKs and PPKLs) or targeting the overlapping sequences of family members (OsBZRs). The mutants exhibited a diversity of plant height, leaf angle, and grain morphology. Comparison analysis of the phenotypes together with BR sensitivity tests suggested the existence of functional redundancy, differentiation, or dominancy among the members within each family. In addition, we generated a set of transgenic plants overexpressing GSK2, OsBZR1/2, and PPKL2, respectively, in wild-type or activated forms with fusion of different tags, and also verified the protein response to BR application. Collectively, these plants greatly enriched the diversity of important agronomic traits in rice. We propose that editing of BR-related family genes could be a feasible approach for screening of desired plants to meet different requirements. Release of these materials as well as the related information also provides valuable resources for further BR research and utilization.  相似文献

As toxic pollutants commonly found in tobacco (Nicotiana tabacum L.) products, lead (Pb) and cadmium (Cd) can enter the human body via smoking and thus pose a potential health risk to smokers. We conducted a greenhouse experiment to study the effects of arbuscular mycorrhizal (AM) inoculation with Glomus intraradices BEG 141 and organic amendment with cattle manure, alone or in combination, on the growth, P nutrition, and heavy-metal uptake by tobacco plants grown in soil to which was added Pb-Cd at 0/0, 350/1, 500/10, and 1,000/100?mg?kg?1, respectively. In general, AM colonization and plant growth were greatly reduced by Pb-Cd contamination, whereas organic amendment alleviated Pb-Cd stress and showed some beneficial effects on AM symbiosis and some soil parameters. AM inoculation, alone or in combination with organic amendment, increased plant dry weights and improved P nutrition significantly at all Pb-Cd addition levels, and, in most cases, it decreased Pb and Cd concentrations in tobacco plants and DTPA-extractable concentrations in soil. AM inoculation increased total glomalin-related soil protein (GRSP) concentrations in soil to which Pb-Cd was added. The higher soil pH and GRSP contents and the lower DTPA-extractable Pb and Cd concentrations contributed by AM inoculation and/or organic amendment may be contributing factors that lead to higher growth promotion and lower metal toxicity and uptake by plants. Our findings suggest that AM inoculation in combination with organic manure may be a potential method for not only tobacco production but phytostabilization of Pb-Cd-contaminated soil.  相似文献

Pf62-Y and Pf62-X is a pair of allelic Y chromosome-linked and X chromosome-linked markers, and have been used to identify YY super-males, XY males and XX females for commercial production of all-male populations in yellow catfish (Pelteobagrus fulvidraco). However, the SCAR primers used previously have only two nucleotide difference, which restricts the wide utility because of nucleotide polymorphism. In this study, a continuous 8102 bp Pf62-Y sequence and a 5362 bp Pf62-X sequence have been cloned by genome walking, and significant genetic differentiation has been revealed between the corresponding X and Y chromosome allele sequences. Moreover, three pairs of primers were designed to efficiently identify YY super-males, XY males and XX females in an artificial breeding population, and to distinguish XY males and XX females in various wild populations. Together, the three new sex-specific genetic markers develop a highly stable and efficient method for genetic sex identification and sex control application in sustainable aquaculture of all-male yellow catfish.  相似文献
