Amyloid precursor protein (APP), commonly associated with Alzheimer disease, is upregulated and distributes evenly along the injured axons, and therefore, also known as a marker of demyelinating axonal injury and axonal degeneration. However, the physiological distribution and function of APP along myelinated axons was unknown. We report that APP aggregates at nodes of Ranvier (NOR) in the myelinated central nervous system (CNS) axons but not in the peripheral nervous system (PNS). At CNS NORs, APP expression co-localizes with tenascin-R and is flanked by juxtaparanodal potassium channel expression demonstrating that APP localized to NOR. In APP-knockout (KO) mice, nodal length is significantly increased, while sodium channels are still clustered at NORs. Moreover, APP KO and APP-overexpressing transgenic (APP TG) mice exhibited a decreased and an increased thickness of myelin in spinal cords, respectively, although the changes are limited in comparison to their littermate WT mice. The thickness of myelin in APP KO sciatic nerve also increased in comparison to that in WT mice. Our observations indicate that APP acts as a novel component at CNS NORs, modulating nodal formation and has minor effects in promoting myelination.  相似文献

Foliage from 21 red mangroves (Rhizophora mangle, R. racemosa and R. x harrisonii) from different ecogeographic conditions in Gabon, West Africa, was analyzed for epicuticular wax composition using CC, GC and GC-MS. Aliphatic hydrocarbons ranging from C23 to C34 and some triterpenoids were identified. Alkanes were the major constituents (46.7–99.9%), with triterpenoids also accounting for up to 53.3% of the wax extract. High contents of octacosane (27.2%) and lower amounts of nonacosane (14.9%) and hentriacontane (9.8%) distinguished R. mangle from the other two species. R. hizophora x harrisonii was exceptionally rich in nonacosane (45.3%) with moderately high concentrations of hentriacontane (25.4%), whereas R. racemosa was intermediate between the two former species in its content of these three alkanes. Inclusion of the less abundant constituents in principal components analysis provided a good separation of R. x harrisonii, whereas the two other species showed some degree of overlap. Although major alkane patterns can be used to discriminate among the Rhizophora species examined, we do find substantial intra-specific variation that may be attributable to population genetic variation; this was least for R. x harrisonii. The hybrid status of this latter species could not be confirmed from the biochemical analyses carried out.  相似文献

Human perivascular stem cells (PSCs) can be isolated in sufficient numbers from multiple tissues for purposes of skeletal tissue engineering. PSCs are a FACS-sorted population of 'pericytes' (CD146+CD34-CD45-) and 'adventitial cells' (CD146-CD34+CD45-), each of which we have previously reported to have properties of mesenchymal stem cells. PSCs, like MSCs, are able to undergo osteogenic differentiation, as well as secrete pro-osteogenic cytokines. In the present protocol, we demonstrate the osteogenicity of PSCs in several animal models including a muscle pouch implantation in SCID (severe combined immunodeficient) mice, a SCID mouse calvarial defect and a femoral segmental defect (FSD) in athymic rats. The thigh muscle pouch model is used to assess ectopic bone formation. Calvarial defects are centered on the parietal bone and are standardly 4 mm in diameter (critically sized). FSDs are bicortical and are stabilized with a polyethylene bar and K-wires. The FSD described is also a critical size defect, which does not significantly heal on its own. In contrast, if stem cells or growth factors are added to the defect site, significant bone regeneration can be appreciated. The overall goal of PSC xenografting is to demonstrate the osteogenic capability of this cell type in both ectopic and orthotopic bone regeneration models.  相似文献

The human ZC3HAV1 gene encodes an antiviral protein. The longest splicing isoform of ZC3HAV1 contains a C-terminal PARP-like domain, which has evolved under positive selection in primates. We analyzed the evolutionary history of this same domain in humans and in Pan troglodytes. We identified two variants that segregate in both humans and chimpanzees; one of them (rs3735007) does not occur at a hypermutable site and accounts for a nonsynonymous substitution (Thr851Ile). The probability that the two trans-specific polymorphisms have occurred independently in the two lineages was estimated to be low (P = 0.0054), suggesting that at least one of them has arisen before speciation and has been maintained by selection. Population genetic analyses in humans indicated that the region surrounding the shared variants displays strong evidences of long-standing balancing selection. Selection signatures were also observed in a chimpanzee population sample. Inspection of 1000 Genomes data confirmed these findings but indicated that search for selection signatures using low-coverage whole-genome data may need masking of repetitive sequences. A case-control study of more than 1,000 individuals from mainland Italy indicated that the Thr851Ile SNP is significantly associated with susceptibility to multiple sclerosis (MS) (odds ratio [OR] = 1.47, 95% confidence intervals [CI]: 1.08-1.99, P = 0.011). This finding was confirmed in a larger sample of 4,416 Sardinians cases/controls (OR = 1.18, 95% CI: 1.037-1.344, P = 0.011), but not in a population from Belgium. We provide one of the first instances of human/chimpanzee trans-specific coding variant located outside the major histocompatibility complex region. The selective pressure is likely to be virus driven; in modern populations, this variant associates with susceptibility to MS, possibly via the interaction with environmental factors.  相似文献

A novel numerical optimization algorithm inspired from weed colonization   总被引:10,自引:0,他引:10  
This paper introduces a novel numerical stochastic optimization algorithm inspired from colonizing weeds. Weeds are plants whose vigorous, invasive habits of growth pose a serious threat to desirable, cultivated plants making them a threat for agriculture. Weeds have shown to be very robust and adaptive to change in environment. Thus, capturing their properties would lead to a powerful optimization algorithm. It is tried to mimic robustness, adaptation and randomness of colonizing weeds in a simple but effective optimizing algorithm designated as Invasive Weed Optimization (IWO). The feasibility, the efficiency and the effectiveness of IWO are tested in details through a set of benchmark multi-dimensional functions, of which global and local minima are known. The reported results are compared with other recent evolutionary-based algorithms: genetic algorithms, memetic algorithms, particle swarm optimization, and shuffled frog leaping. The results are also compared with different versions of simulated annealing — a generic probabilistic meta-algorithm for the global optimization problem — which are simplex simulated annealing, and direct search simulated annealing. Additionally, IWO is employed for finding a solution for an engineering problem, which is optimization and tuning of a robust controller. The experimental results suggest that results from IWO are better than results from other methods. In conclusion, the performance of IWO has a reasonable performance for all the test functions.  相似文献

A strain of Saccharomyces cerevisiae having desirable winemaking properties and high spore viability was bred from a semi-homothallic parent strain with similar winemaking properties but that produced sixfold fewer viable spores. Because the parent was homozygous for HO and for the MATa allele at both silent HMR and HML loci, it produced two MATa and two nonmating progeny per ascus. To obtain a segregant able to mate with the stable MATa progeny, a strain of the nonmating progeny, previously subjected to HO distruption with a KanMX4 cassette, was used. The resultant MATalphaho::KanMX4 transformant was mated to a MATa HO segregant and the diploid produced was sporulated to allow the isolation of a semi-homothallic diploid segregant designated 2D that lacked the KanMX4-disrupted HO allele as confirmed by sequence analysis. Genetic analysis indicated greater homozygosity in 2D than in the parent as assessed by PCR at five loci. The sugar consumption profiles of both 2D and the parent in grape juice fermentations were the same. Acetaldehyde levels and postfermentation biofilm formation were higher in 2D than in the parent. Because 2D has acceptable winemaking characteristics but produces significantly more viable spores than the parent strain, it will be useful in future breeding efforts.  相似文献

Distribution of Na+-K+-ATPase alpha-subunit mRNA in full-grown oocytes and early embryos from of X. laevis has been studied using in situ hybridization on histological sections. The mRNA is dispersed about equally in the cytoplasm and is absent from the nucleus. The concentration of the mRNA is successively decreased from fertilization to the late blastula. The drastic decrease in mRNA concentration is observed in a period from morula to mid-blastula. The local increase in the mRNA concentration was detected in dorsal mesoderm and ectoderm during gastrulation.  相似文献
