Summary Many problems and difficulties are encountered in making genetic improvements in plants where both apomixis and polyploidy occur together. From biosystematic studies on an agamic species complex, Dichanthium annulatum, information is presented on: (A) Mechanisms which create variability in apomicts — (i) genome building and reduction, (ii) hybridization between ecotypes of facultative apomicts, (iii) fertilization of unreduced gametes, (iv) introgressive hybridization, (v) preferential pairing and genotypic control of bivalent formation and (vi) induced mutation; (B) Embryo-sac variations, vis-a-vis sexual/apomictic sacs — (i) production of sexual embryo-sac in apomicts, (ii) balance between apomixis and sexual process, (iii) effect of environment and experimental manipulation of the type of embryo-sac; and (C) Heterosis and fixation of apomixis.The utilization and exploitation of these mechanisms and phenomena for accelerating the genetic improvement of apomictic plants is discussed.Mating systems impose certain restrictions on the breeding methodology to be used in the genetic improvement of crop plants. Allogamous species have built-in mechanisms for self-improvement and, for them, the breeding techniques are well worked out. Little information is, however, available on the procedures to be followed for the genetic improvement of apomicts. Recently gathered information on the causal mechanisms of apomixis and its mode of inheritance, the genetic systems which regulate the balance between apomixis and sexuality, the physical and chemical agents for artificial induction of sexuality in apomicts, and the processes which promote variability and adaptive polymorphism in apomicts show a way for the creation, exploitation and fixation of superior genotypes. Such information, based on biosystematic studies on an agamic species complex, Dichanthium annulatum, at the Oklahoma State University, Stillwater, Oklahoma, U.S.A., is presented here.Breeding procedures commonly followed for the genetic improvement of apomicts are outlined below:1. Collection of varieties, strains or ecotypes from diverse sources; 2. Evaluation of the germ plasm for the presence of desirable characters; 3. Building up of selection indices and estimation of genetic parameters; 4. Determination of mode of reproduction and isolation of sexual types or clones; 5. Hybridization using the sexual types; 6. Progeny testing, comparisons, multiplication and release of superior types.Thus, the success of the breeding programme would depend on the range of variability already present in the germ plasm collections, the relative proportion of sexual/apomictic seed produced and the exploitation of variability from the crossbred progenies. Since large collections of plants with different genotypes are not often available, one would like to look for the mechanisms which can create variability in the apomicts. In eukaryotes, the origin recognition complex (ORC) is essential for the initiation of DNA replication. The largest subunit of this complex (ORC1) has a regulatory role in origin activation. Here we report the cloning and functional characterization of Plasmodium falciparum ORC1 homolog. Using immunofluorescence and immunoelectron microscopy, we show here that PfORC1 is expressed in the nucleus during the late trophozoite and schizont stages where maximum amount of DNA replication takes place. Homology modelling of the carboxy terminal region of PfORC1 (781-1033) using Saccharomyces pombe Cdc6/Cdc18 homolog as a template reveals the presence of a similar AAA+ type nucleotide-binding fold. This region shows ATPase activity in vitro that is important for the origin activity. To our knowledge, this is the first evidence of an individual ORC subunit that shows ATPase activity. These observations strongly suggest that PfORC1 might be involved in DNA replication initiation during the blood stage of the parasitic life cycle. 相似文献

Principles
Case weights of Diagnosis Related Groups (DRGs) are determined by the average cost of cases from a previous billing period. However, a significant amount of cases are largely over- or underfunded. We therefore decided to analyze earning outliers of our hospital as to search for predictors enabling a better grouping under SwissDRG.Methods
28,893 inpatient cases without additional private insurance discharged from our hospital in 2012 were included in our analysis. Outliers were defined by the interquartile range method. Predictors for deficit and profit outliers were determined with logistic regressions. Predictors were shortlisted with the LASSO regularized logistic regression method and compared to results of Random forest analysis. 10 of these parameters were selected for quantile regression analysis as to quantify their impact on earnings.Results
Psychiatric diagnosis and admission as an emergency case were significant predictors for higher deficit with negative regression coefficients for all analyzed quantiles (p<0.001). Admission from an external health care provider was a significant predictor for a higher deficit in all but the 90% quantile (p<0.001 for Q10, Q20, Q50, Q80 and p = 0.0017 for Q90). Burns predicted higher earnings for cases which were favorably remunerated (p<0.001 for the 90% quantile). Osteoporosis predicted a higher deficit in the most underfunded cases, but did not predict differences in earnings for balanced or profitable cases (Q10 and Q20: p<0.00, Q50: p = 0.10, Q80: p = 0.88 and Q90: p = 0.52). ICU stay, mechanical and patient clinical complexity level score (PCCL) predicted higher losses at the 10% quantile but also higher profits at the 90% quantile (p<0.001).Conclusion
We suggest considering psychiatric diagnosis, admission as an emergencay case and admission from an external health care provider as DRG split criteria as they predict large, consistent and significant losses. 相似文献76.

The mechanism of DNA replication initiation and progression is poorly understood in the parasites, including human malaria parasite Plasmodium falciparum . Using bioinformatics tools and yeast complementation assay, we identified a putative homologue of Saccharomyces cerevisiae o rigin r ecognition c omplex subunit 5 in P. falciparum (PfORC5). PfORC5 forms distinct nuclear foci colocalized with the replication foci marker proliferating cell nuclear antigen (PfPCNA) and co-immunoprecipitates with PCNA during early-to-mid trophozoite stage replicating parasites. Interestingly, these proteins separate from each other at the non-replicating late schizont stage, citing the evidence of the presence of both PCNA and ORC components in replication foci during eukaryotic DNA replication. PfORC1, another ORC subunit, colocalizes with PfPCNA and PfORC5 at the beginning of DNA replication, but gets degraded at the late schizont stage, ensuring the regulation of DNA replication in the parasites. Further, we have identified putative PCNA-interacting protein box in PfORC1 that may explain in part the colocalization of PfORC and PfPCNA. Additionally, use of specific DNA replication inhibitor hydroxyurea affects ORC5/PCNA foci formation and parasitic growth. These results strongly favour replication factory model in the parasites and confer great potential to understand the co-ordination between ORC and PCNA during eukaryotic DNA replication in general. 相似文献

In the current study, the structural and spectroscopic properties of phosphatic shale samples obtained from the Atomic Minerals Directorate for Exploration and Research were probed for potential use as a phosphor material. X‐ray diffraction and Raman and Fourier transform infrared spectroscopy revealed that the beneficiated phosphatic shale samples were primarily monophasic consisting of fluorapatite [Ca5(PO4)3F, (FAP)] with minor traces of haematite (α‐Fe2O3) and calcite (CaCO3). Energy dispersive X‐ray fluorescence revealed the presence of U, Eu, Dy and Tb in the FAP matrix substituted at Ca(I) and Ca(II) sites of FAP. A reduced optical direct band gap of 4.46 eV was calculated from the Tauc plot. Photoluminescence spectral studies revealed multicolour emissions (red, yellow, green and blue) on ultraviolet light excitation that were attributed to luminescence spectra from rare earth ions Eu3+, Tb3+, U4+ and U6+ in the FAP matrix. The overall emissions for the rare earth and actinide‐doped FAP were obtained in the cool white region and the corresponding Commission Internationale de l’Eclairage chromaticity coordinates were calculated to be (0.274, 0.317). The corresponding colour correlated temperature obtained was 9342 K. Furthermore, phosphatic shale had a high room temperature dielectric constant of 11 at a frequency of 1 kHz that demonstrated its suitability for use in biological sensors.
