The current study investigated mechanical predictors for the development of adjacent disc degeneration. A 3-D finite element model of a lumbar spine was modified to simulate two grades of degeneration at the L4–L5 disc. Degeneration was modeled by changes in geometry and material properties. All models were subjected to follower preloads of 800 N and moment loads in the three principal directions of motion using a hybrid protocol. Degeneration caused changes in the loading and motion patterns of the segments above and below the degenerated disc. At the level (L3–L4) above the degenerated disc, the motion increased due to moderate degeneration by 21% under lateral bending, 26% under axial rotation and 28% under flexion/extension. At the level (L5-S1) below the degenerated disc, motion increased only during lateral bending by 20% due to moderate degeneration. Both the L3–L4 and L5-S1 segment showed a monotonic increase in both the maximum von Mises stress and shear stress in the annulus as degeneration progressed for all loading directions, expect extension at L3–L4. The most significant increase in stress was observed at the L5-S1 level during axial rotation with nearly a ten-fold increase in the maximum shear stress and 103% increase in the maximum von Mises stress. The L5-S1 segment also showed a progressive increase in facet contact force for all loading directions with degeneration. Nucleus pressure did not increase significantly for any loading direction at either the caudal or cephalic adjacent segment. Mitophagy removes defective mitochondria via lysosomal elimination. Increased mitophagy coincides with metabolic reprogramming, yet it remains unknown whether mitophagy is a cause or consequence of such state changes. The signalling pathways that integrate with mitophagy to sustain cell and tissue integrity also remain poorly defined. We performed temporal metabolomics on mammalian cells treated with deferiprone, a therapeutic iron chelator that stimulates PINK1/PARKIN‐independent mitophagy. Iron depletion profoundly rewired the metabolome, hallmarked by remodelling of lipid metabolism within minutes of treatment. DGAT1‐dependent lipid droplet biosynthesis occurred several hours before mitochondrial clearance, with lipid droplets bordering mitochondria upon iron chelation. We demonstrate that DGAT1 inhibition restricts mitophagy in vitro, with impaired lysosomal homeostasis and cell viability. Importantly, genetic depletion of DGAT1 in vivo significantly impaired neuronal mitophagy and locomotor function in Drosophila. Our data define iron depletion as a potent signal that rapidly reshapes metabolism and establishes an unexpected synergy between lipid homeostasis and mitophagy that safeguards cell and tissue integrity. 相似文献

Atlantic salmon ( Salmo salar L.) alevins hatched from eggs transferred from high- to low-Na water at 250° days, before the onset of the phase of increasing whole egg sodium content (at ∼380°days), showed a significantly reduced K m for Na+ transport, whereas transfer at 400° days did not produce any change in K m . Alevins hatched from eggs given acid shocks of 1, 3, 7 or 14 days duration initiated at 250 or 400° days showed no significant changes in Na+ transporter K m . Extended acid exposure (38 days) from 250°days to hatching resulted in a slight lowering of K m (P<0.05). A 24-day acid exposure from 400°days to hatching had no effect on Na+ transporter K m . Alevins hatched from eggs incubated throughout in acidified water had a significantly reduced K m compared to controls (P<0.01).

All air‐breathing organisms must face the challenge of oxidative damage, and understanding how animals cope can lend insight into their ecology. Unlike other vertebrates, birds rely primarily on fats to fuel endurance exercise such as migration, and therefore face a greater potential for damage from the reactive by‐products of their own metabolism. We review the physiological ecology of migrating birds through the lens of oxidation–reduction chemistry, underscoring how oxidative balance in wild birds may affect their dietary choices and use of critical stopover habitats during migration. Recent studies reveal that migratory birds prepare for oxidative challenges either by up‐regulating endogenous antioxidants or by consuming them in their diet, and they repair oxidative damage after long flights, although much remains to be discovered about how birds maintain oxidative balance over the course of migration. We conclude by describing some of the most used and useful measures of antioxidant status and oxidative damage that field ornithologists can include in their tool kit of techniques to probe the oxidative balance of wild birds. 相似文献

Previous studies of the effect of carbonation on taste perception have
suggested that it may be negligible, manifesting primarily in increases in
the perceived intensity of weak salt and sour stimuli. Assuming CO2
solutions in the mouth stimulate only trigeminal nerve endings, this result
is not altogether surprising; however, there are neurophysiological data
indicating that CO2 stimulates gustatory as well as trigeminal fibers. In
that case, carbonation might alter the quality profile of a stimulus
without producing substantial changes in overall taste intensity--much as
occurs when qualitatively different taste stimuli are mixed. To address
this possibility, subjects were asked to rate the total taste intensity of
moderate concentrations of stimuli representing each of the basic tastes
and their binary combinations, with an without added carbonation. They then
subdivided total taste intensity into the proportions of sweetness,
saltiness, sourness, bitterness and 'other taste qualities' they perceived.
The addition of carbonation produced only small increases in ratings of
total taste intensity. However, rather dramatic alterations in the quality
profiles of stimuli were observed, particularly for sweet and salty tastes.
The nature of the interaction is consistent with a direct effect of
carbonation/CO2 on the gustatory system, although the possibility that at
least some of the observed effects reflect trigeminal-gustatory
interactions cannot be ruled out.

The size of the pectoral muscle is an important component of body condition in birds and has been linked to indices of fitness and migratory performance. Bauchinger et al. (2011. Journal of Ornithology 152: 507–514) developed, calibrated, and validated an aluminum “muscle meter” device that estimates the size of pectoral muscles noninvasively. To make this tool more widely available, we created a CAD model from 3D‐scan data of the aluminum muscle meter that can be 3D‐printed in durable plastic for ~ $30 USD. We tested this device on seven species of songbirds in Jamaica, The Bahamas, Cameroon, Equatorial Guinea, and Michigan. We demonstrate that the breast muscle meter measurements are (1) repeatable among users, (2) correlated with a four‐category visual breast muscle scoring system, and (3) correlated with scaled mass index (an index of body condition). Muscle scores from our device outperformed the traditional four‐category muscle scoring system in predicting scaled mass index. Finally, with our device, we quantified the increasing breast muscle size of American Redstarts (Setophaga ruticilla) from March through May as they prepared for spring migration. Given the precision of the 3D‐scanning hardware used to generate our 3D image for printing, we produced a plastic muscle meter that is as precise and useful as the aluminum original, but more cost‐effective and widely available. 相似文献

IntroductionPain remains the most important problem for people with rheumatoid arthritis (RA). Active inflammatory disease contributes to pain, but pain due to non-inflammatory mechanisms can confound the assessment of disease activity. We hypothesize that augmented pain processing, fibromyalgic features, poorer mental health, and patient-reported 28-joint disease activity score (DAS28) components are associated in RA.MethodsIn total, 50 people with stable, long-standing RA recruited from a rheumatology outpatient clinic were assessed for pain-pressure thresholds (PPTs) at three separate sites (knee, tibia, and sternum), DAS28, fibromyalgia, and mental health status. Multivariable analysis was performed to assess the association between PPT and DAS28 components, DAS28-P (the proportion of DAS28 derived from the patient-reported components of visual analogue score and tender joint count), or fibromyalgia status.ResultsMore-sensitive PPTs at sites over or distant from joints were each associated with greater reported pain, higher patient-reported DAS28 components, and poorer mental health. A high proportion of participants (48%) satisfied classification criteria for fibromyalgia, and fibromyalgia classification or characteristics were each associated with more sensitive PPTs, higher patient-reported DAS28 components, and poorer mental health.ConclusionsWidespread sensitivity to pressure-induced pain, a high prevalence of fibromyalgic features, higher patient-reported DAS28 components, and poorer mental health are all linked in established RA. The increased sensitivity at nonjoint sites (sternum and anterior tibia), as well as over joints, indicates that central mechanisms may contribute to pain sensitivity in RA. The contribution of patient-reported components to high DAS28 should inform decisions on disease-modifying or pain-management approaches in the treatment of RA when inflammation may be well controlled. 相似文献
