In this paper a three-dimensional continuum model of a mammalian left ventricle is formulated. The stresses in the model satisfy the conditions of zero stress on the outer (epicardial surface-representing) boundary. The strains of the model are obtained from the actual dynamic geometry measurements (obtained from cineangiocardiography). Since the left ventricular muscle is incompressible, the dilatational strain is zero and hence the (three-dimensional) deviatric stress components are related to the corresponding strain components by Maxwell and Voigt rheological model analogues of one-dimensional systems; the parameters of the model are series and parallel elastic (SE, PE) elements and the contractile element (CE) (representing the sarcomere). The incorporation of the rheological features of the cardiac muscle into the three-dimensional constitutive equations (for the three-dimensional continuum model of the left ventricle) is a feature of this paper. A procedure is presented to determine the parameters of the constitutive equations (i.e., the SE, PE, and the parameters of the force-velocity relation for the CE) for the left ventricle of a subject from data on the dimensions and chamber pressure of the left ventricle. The values of these parameters characterize the rheology of the left ventricular muscle of the subject. In order to demonstrate clinical application of the analyses, in vivo data of the subjects' left ventricular pressure and dimensions are obtained, and the analyses are applied to the data to determine (for each subject) the values and characteristics of the elastic elements and CEs.  相似文献

Purification of cytochrome b6 was pursued to further develop rational technology for purification, proof of purity, and study of properties of membrane proteins. Cytochrome b6 was purified—the first time from any source—from spinach chloroplast membranes; yield of pure cytochrome b6 was 30% of that found in ethanol-extracted particles. The three-step procedure (pH 8) employed: (I) extraction in Triton X-100-4 M (optionally 2 M) urea, (II) chromatography in a Bio-Gel A-1.5m Column (Triton X-100-4 M urea). Without this step, subsequent electrophoresis failed. (III) Preparative disc gel electrophoresis.

Properties of cytochrome b6: Cytochrome b6 migrated in undenatured form as a single band in disc electrophoresis (pH 8, 7 or 8.9). None of the limited, accepted properties of the cytochrome in particles was altered by the purification procedure: Reduced b6 has absorption maxima (22 °C) at 434, 536, and 563 nm; at −199 °C the a absorption region shows two peaks of equal intensity at 561 and 557 nm. Cytochrome b6 is reduced by dithionite (not by ascorbate) and is autoxidizable. The prosthetic group of b6 is protohaemin and is fully extractable by acid-acetone. No non-haem iron is present. The millimolar extinction coefficient of reduced b6 (563–600nm) per mole of haem is 21. The protein equivalent weight is 40000 g per mole of haem. Cytochrome b4 is an intrinsically aggregatable molecule. The reduced cytochrome does not react with CO except when Triton X-100 is present.  相似文献

Eye movements of monkeys were measured during performance on a series of two-choice color and form discriminations which involved reversal and nonreversal shifts. Neither the amount of visual scanning nor the duration of individual fixations was related to the relevant dimension or to the type of shift. However, amount of scanning increased over the series of problems and was linearly related to the time to respond. The last fixation of a trial tended to be on the positive stimulus and a fixation near the end of a trial tended to be longer than one near the beginning.This research was supported by a research grant (GB-18953X) from the National Science Foundation to the first author. In perfusion fixation, we have preferred gravity flow rather than a peristaltic forced flow because the latter may cause rupture of small blood vessels. One difficulty encountered in the gravity flow method is detection of flow stoppage, often caused by the formation of a blood clot. If this is apparent soon enough, steps can be taken to re-establish the flow, Flow stoppage is not usually detected for several minutes—resulting in poor fixation of the organ involved. Differential DNA replication is widely held to influence polytene chromosome structure by causing the dramatic reductions in heterochromatic DNA content that are characteristic of most endopolyploid cells. The underreplication model of heterochromatic sequence underrepresentation predicts that replication intermediates should populate regions of DNA between fully polytenized euchromatic sequences and underpolytenized heterochromatic sequences. We directly tested this prediction using Dp1187, a 1300 kb Drosophila minichromosome containing well-defined heterochromatic regions. DNA from a euchromatic/heterochromatic junction region of Dp1187, demonstrating a significant gradient of underrepresentation in larval salivary glands, lacked the stalled replication forks predicted by the underreplication model. Transforming growth factor beta (TGF beta) treatment of rat osteoblast-rich calvarial cells or of the clonal osteogenic sarcoma cells, UMR 106-01, resulted in dose-dependent inhibition of plasminogen activator (PA) activity, and increased production of 3.2 kb mRNA and protein for PA inhibitor -1 (PAI-1). Although tissue-type PA (tPA) protein was not measured, TGF beta did not influence production of mRNA for tPA. Production of 2.3 kb mRNA for urokinase-type PA (uPA) was also increased by TGF beta in a dose-dependent manner. The effects of TGF beta on synthesis of mRNA for PAI-1 and uPA were maintained when protein synthesis was inhibited, and were abolished by inhibition of RNA synthesis. Although uPA had not been detected previously as a product of rat osteoblasts, treatment of lysates of osteoblast-like cells with plasmin yielded a band of PA activity on reverse fibrin autography, corresponding to a low Mr form of uPA. Untreated conditioned media from normal osteoblasts or UMR 106-01 cells contained no significant TGF beta activity, but activity could be detected in acidified medium. Treatment of conditioned media with plasmin resulted in activation of approximately 50% of the TGF beta detectable in acidified media. The results identify several effects of TGF beta on the PA-PA inhibitor system in osteoblasts. Net regulation of tPA activity through the stimulatory actions of several calciotropic hormones and the promotion of PAI-1 formation by TGF beta could determine the amount of osteoblast-derived TGF beta activated locally in bone. Stimulation of osteoblast production of mRNA for uPA could reflect effects on the synthesis of sc-uPA, a precursor for the active form of the enzyme.  相似文献

Amblyomma americanum andA. maculatum were studied to determine if genital sex pheromones were present, as reported inDermacentor species. Chemical analysis of methanol extracts of the anterior reproductive tracts (source of the genital sex pheromone) ofA. americanum andA. maculatum confirmed the presence of the same fatty acids that act as components of the genital sex pheromone inDermacentor variabilis andD. andersoni. In bothAmblyomma species, removal of the anterior reproductive-tract of fed females eliminated the male copulatory response. ForA. americanum, use of anterior reproductive tract extracts made with water, methanol, ether and hexane elicited significant probing and copulatory responses by conspecific males. However, inA. maculatum, use of anterior reproductive-tract extracts made with water, methanol, ether, chloroform: methanol (41), hexane and pentane did not restore mating response by males. Treatment with known components ofDermacentor genital sex pheromones elicited weak but significant responses inA. americanum, but did not restore male mating responses inA. maculatum. These results indicate that genital sex pheromones exist inA. americanum and are not restricted to the genusDermacentor. Characteristics of theAmblyomma genital sex pheromone, however, do differ considerably from those inDermacentor. No genital sex pheromone was present inA. maculatum.  相似文献

Two tomato cDNA libraries were synthesized from poly(A)+ RNAs isolated from unwounded and wounded tomato stems. These cDNA libraries were packaged in gt10 and screened by in situ plaque hybridization with a tomato extensin gene clone (pTom 5.10). Several cDNA clones were identified and isolated from both libraries in this manner and subjected to restriction enzyme digestion, Southern gel blot hybridization, RNA gel blot hybridization, and DNA sequence analyses. From these analyses, the various cDNA clones were found to fall into one of five distinct classes (classes I–V). Class I clones hybridized to a 4.0 kb mRNA which accumulated markedly after wounding and encoded an extensin characterized largely by Ser-(Pro)4-Ser-Pro-Ser-(Pro)4-(Tyr)3-Lys repeats. Class II clones hybridized to a 2.6 kb mRNA which showed no accumulation following wounding and encoded an extensin containing Ser-(Pro)4-Ser-Pro-Ser-(Pro)4-Thr-(Tyr)1–3-Ser repeats. Class III clones hybridized to a 0.6 kb mRNA which greatly accumulated in response to wounding and encoded a glycine-rich protein (GRP) with (Gly)2–6-Tyr-Pro and(Gly)2–6-Arg repeats. Class IV clones contained both class I and class III DNA sequences and consequently hybridized to both the 4.0 kb and the 0.6 kb wound-accumulating mRNAs; these clones encoded a portion of a GRP sequence on one DNA strand and encoded a portion of an extensin sequence on the other DNA strand. Class V clones hybridized to a 2.3 kb mRNA which decreased following wounding and encoded a GRP sequence characterized by (Gly)2–5-Arg repeats.  相似文献
