Aims:  The influence of the cheese-making process, ripening conditions and primary starter on the viability and proteolytic activity of an adjunct culture of Lactobacillus plantarum I91 was assessed in two miniature cheese models, representative of Cremoso Argentino and Cheddar cheeses.
Methods and Results:  Cheeses with and without adjunct culture were made under controlled microbiological conditions and sampled during ripening for physicochemical and microbiological analyses. The addition of lactobacilli neither contributed to acid production nor caused changes to the composition of the cheeses. The strain studied exhibited good development and survival and showed a similar growth pattern in both cheese matrices. The adjunct culture caused changes to secondary proteolysis of both cheese types, which were evidenced by modification of peptide profiles and the increase in the levels of some individual amino acids as well as the total content of free amino acids. The changes observed were consistent with the acceleration of proteolysis in the two cheese models assayed.
Conclusion:  Lactobacillus plantarum I91 has desirable and robust technological properties, which makes it a suitable adjunct culture for cheese-making.
Significance and Impact of the Study:  Other cultures and environmental conditions prevailing in the food may affect the viability of adjunct cultures and its biochemical activities; this is the first report describing the successful performance of an adjunct culture of Lact. plantarum I91 in two different model cheese systems.  相似文献

The experiments tested the idea that changes in habituation to the reinforcer contribute to behavioral interactions during multiple schedules. This idea predicts that changing an aspect of the reinforcer should disrupt habituation and produce an interaction. Pigeons and rats responded on multiple variable interval variable interval schedules. Introducing variability into the duration of reinforcers in one component increased response rates in both components when the schedules provided high, but not low, rates of reinforcement. The increases in constant-component response rates grew larger as the session progressed. Within-session decreases in responding were smaller when the other component provided variable-, rather than fixed-, duration reinforcers. These results are consistent with the idea that changes in habituation to the reinforcer contribute to behavioral interactions. They help to explain why interactions do not occur for some subjects under conditions that produce them for others. Finally, the results question the assumption that induction and behavioral contrast are always produced by different theoretical mechanisms.  相似文献

Here, we investigated the allometry of a conspicuous female ornament in Pelvicachromis taeniatus, a biparental cichlid that shows mutual mate choice and ornamentation. Trehalose (alpha-D-glucopyranosyl-1,1-alpha-D-glucopyranose) is a non-reducing diglucoside found in various organisms that serves as a carbohydrate reserve and as an agent that protects against a variety of physical and chemical stresses. Deinococcus radiodurans possesses an alternative biosynthesis pathway for the synthesis of trehalose from maltooligosaccharides. This reaction is mediated by two enzymes: maltooligosyltrehalose synthase (MTSase) and maltooligosyltrehalose trehalohydrolase (MTHase). Here, we present the 1.1A resolution crystal structure of MTHase. It consists of three major domains: two beta-sheet domains and a conserved glycosidase (beta/alpha)8 barrel catalytic domain. Three subdomains consisting of short insertions were identified within the catalytic domain. Subsequently, structures of MTHase in complex with maltose and trehalose were obtained at 1.2 A and 1.5 A resolution, respectively. These structures reveal the importance of the three inserted subdomains in providing the key residues required for substrate recognition. Trehalose is recognised specifically in the +1 and +2 binding subsites by an extensive hydrogen-bonding network and a strong hydrophobic stacking interaction in between two aromatic residues. Moreover, upon binding to maltose, which mimics the substrate sugar chain, a major concerted conformational change traps the sugar chain in the active site. The presence of magnesium in the active site of the MTHase-maltose complex suggests that MTHase activity may be regulated by divalent cations.  相似文献

Despite the presence of melon‐headed whales in tropical and subtropical waters worldwide, little is known about this species. To assess population structure in Hawai‘i, dedicated field efforts were undertaken from 2000 to 2009. Using only good quality photographs, there were 1,433 unique photo‐identified individuals, of which 1,046 were distinctive. Of these, 31.5% were seen more than once. Resighting data combined with social network analyses showed evidence of two populations—a smaller, resident population, seen exclusively off the northwest region of the island of Hawai‘i, and a larger population, seen throughout all the main Hawaiian Islands (hereafter the “main Hawaiian Islands” population). A Bayesian analysis examining the probability of movements of individuals between populations provided a posterior median dispersal rate of 0.0009/yr (95% CI = 0–0.0041), indicating the populations are likely demographically independent. Depth of encounters with the Hawai‘i Island resident population was significantly shallower (median = 381 m) than those with the main Hawaiian Islands population (median = 1,662 m). Resightings of individuals have occurred up to 22 yr apart for the Hawai‘i Island resident population and up to 13 yr apart for the main Hawaiian Islands population, suggesting long‐term residency to the islands for both populations.  相似文献

Background

The genus Micronycteris is a diverse group of phyllostomid bats currently comprising 11 species, with diploid number (2n) ranging from 26 to 40 chromosomes. The karyotypic relationships within Micronycteris and between Micronycteris and other phyllostomids remain poorly understood. The karyotype of Micronycteris hirsuta is of particular interest: three different diploid numbers were reported for this species in South and Central Americas with 2n?=?26, 28 and 30 chromosomes. Although current evidence suggests some geographic differentiation among populations of M. hirsuta based on chromosomal, morphological, and nuclear and mitochondrial DNA markers, the recognition of new species or subspecies has been avoided due to the need for additional data, mainly chromosomal data.

Results

We describe two new cytotypes for Micronycteris hirsuta (MHI) (2n?=?26 and 25, NF?=?32), whose differences in diploid number are interpreted as the products of Robertsonian rearrangements. C-banding revealed a small amount of constitutive heterochromatin at the centromere and the NOR was located in the interstitial portion of the short arm of a second pair, confirmed by FISH. Telomeric probes hybridized to the centromeric regions and weakly to telomeric regions of most chromosomes. The G-banding analysis and chromosome painting with whole chromosome probes from Carollia brevicauda (CBR) and Phyllostomus hastatus (PHA) enabled the establishment of genome-wide homologies between MHI, CBR and PHA.

Conclusions

The karyotypes of Brazilian specimens of Micronycteris hirsuta described here are new to Micronycteris and reinforce that M. hirsuta does not represent a monotypic taxon. Our results corroborate the hypothesis of karyotypic megaevolution within Micronycteris, and strong evidence for this is that the entire chromosome complement of M. hirsuta was shown to be derivative with respect to species compared in this study.

Miniature (20 g) Cheddar-type cheeses were manufactured using enzymes extracted from the crustacean Munida or chymosin as coagulant. Cheeses were ripened at 8 degrees C and samples were collected for analysis after 2, 6 and 12 weeks. Proteolysis was assessed by urea-polyacrylamide gel electrophoresis, which showed that cheeses manufactured with the Munida extracts had a higher extent of degradation of beta-casein than cheeses made using chymosin as coagulant. Patterns of proteolysis were also obtained by reverse-phase high-performance liquid chromatography (RP-HPLC) and matrix assisted laser desorption ionisation-time of flight (MALDI-ToF) mass spectrometry. In general, the products of proteolysis were more complex in cheese made using the Munida extracts than in cheese made by chymosin as coagulant. Statistical analysis of results clearly discriminated the cheeses on the basis of coagulant used. Molecular mass of peptides found in cheese made using Munida extracts were similar to those of peptides commonly detected in cheeses made using chymosin as coagulant.  相似文献

An extract from rat brain has been shown to catalyze the transfer of ganglioside GM1 from sonicated vesicles to erythrocyte ghosts. It also enhanced the transfer of GM1 to a crude neuronal membrane preparation, whereas myelin took up only a very limited amount. The transfer activity was heat-labile. Similar transfer activities were found in extracts from bovine gray and white matter, that of the former being comparable to rat brain whereas the latter was greater per milligram protein.  相似文献

Western Australian bauxite deposits are naturally associated with high amounts of humic and fulvic materials that co-digest during Bayer processing. Sodium oxalate remains soluble and can co-precipitate with aluminium hydroxide unless it is removed. Removal of sodium oxalate requires a secondary crystallisation step followed by storage. Bioreactors treating oxalate wastes have been developed as economically and environmentally viable treatment alternatives but the microbial ecology and physiology of these treatment processes are poorly understood. Analysis of samples obtained from two pilot-scale moving bed biofilm reactors (MBBRs) and one aerobic suspended growth bioreactor (ASGB) using polymerase chain reaction- denaturing gradient gel electrophoresis of 16S rRNA genes showed that members of the α-, β- and γ-Proteobacteria subgroups were prominent in all three processes. Despite differing operating conditions, the composition of the microbial communities in the three reactors was conserved. MBBR2 was the only configuration that showed complete degradation of oxalate from the influent and the ASGB had the highest degradation rate of all three configurations. Several strains of the genus Halomonas were isolated from the bioreactors and their morphology and physiology was also determined.  相似文献
