ESR spectroscopy has been used to investigate the formation of paramagnetic species during the development of internal rust spot (IRS) in the potato tuber. Production of free radicals and oxidation of metal ions such as Fe(II) and Mn(II) occur when necrotic tissue is formed. However. since IRS develops in periods of calcium stress (low calcium supply). it is suggested that the principal cause of the disorder is a loss of cell membrane integrity tvhich is brought ahout by a lack of calcium. Cell senescence and the formation of necrotic tissue may then result either from increased oxygen radical production within the cell or from oxidation of metal complexes in the extracellular regions of the tissue. 相似文献

Microscopic examinations have convinced microbial ecologists that the culturable microbes recovered from environmental samples represent a tiny proportion of the extant microbiota. Methods for recovery and enzymatic amplification of nucleic acids from environmental samples have shown that a huge diversity existsin situ, far exceeding any expectations which were based on direct microscopy. It is now theoretically possible to extract, amplify and sequence all the nucleic acids from a community and thereby gain a comprehensive measure of the diversity as well as some insights into the phylogeny of the various elements within this community. Unfortunately, this analysis becomes economically prohibitive if applied to the multitude of niches in a single biome let alone to a diverse set of environments. It is also difficult to utilize PCR amplification on nucleic acids from some biomes because of coextracting enzymatic inhibitors. Signature biomarker analysis which potentially combines gene probe and lipid analysis on the same sample, can serve as a complement to massive environmental genome analysis in providing quantitative comparisons between microniches in the biome under study. This analysis can also give indications of the magnitude of differences in biodiversity in the blome as well as provide insight into the phenotypic activities of each community in a rapid and cost-effective manner. Protein kinase C (PKC) was found to be present in purified human monocytes and lymphocytes isolated by countercurrent centrifugal elutriation. In unstimulated monocytes and lymphocytes, approximately 90% of the PKC activity was cytosolic when the cells were disrupted in the presence of EGTA. The role of this kinase in the stimulation of the respiratory burst in monocytes was investigated. Phorbol esters capable of triggering the release of O2- caused a loss of PKC activity from the cytosol and the appearance of the kinase activity in the particulate cell fraction. Kinase activity was partially extractable from the particulate fraction by 0.1% Triton X-100, whereupon it demonstrated calcium and lipid dependence. The EC50 for the phorbols in initiating the respiratory burst correlated well with their EC50 for stimulating the appearance of PKC activity in the particulate fraction (R = 0.998). Redistribution of PKC activity in monocytes by phorbol myristate acetate (PMA) was rapid and appeared to precede the release of O2-. PMA also shifted PKC activity from the cytosol to the extractable particulate fraction of lymphocytes. We conclude that redistribution of PKC activity by active phorbols or other cell stimulants could provide substrate specificity for phosphorylation reactions. By shifting PKC activity to the monocyte particulate fraction, active phorbols may initiate the phosphorylation of a substrate required for stimulation of the respiratory burst. 相似文献

We used single-family crosses to confirm the Mendelian interpretation of allozyme variation and to examine linkage relationships at five polymorphic loci in freshwater and anadromous threespine sticklebacks (Gasterosteus aculeatus) from the Little Campbell River, British Columbia. The electrophoretic pattern of a sixth locus, mitochondrial NADP-dependent isocitrate dehydrogenase (IDH), was found to be sexually dimorphic but otherwise invariant in sticklebacks. This raised the possibility of a sex-linked Idh locus. No differences in IDH quantity or substrate affinities (apparent Km values) were detected between male and female sticklebacks. The electrophoretic pattern of IDH expression was not changed in sticklebacks in which sexual development was altered by hormonal treatment. 相似文献
