Tumour-promoting phorbol esters (phorbol-12-myristate-13-acetate, PMA; phorbol-12,13-dibutyrate, PDBu) but not 4β-phorbol, activate protein kinase C. Using human platelets pre-labelled with quin2 or 32PO4 we examined the effects of these compounds on human platelet cytosolic free Ca2+ ([Ca2+]j) and on [32]phosphatidic acid ([32P]PtdOH). PMA and PDBu, but not 4β-phorbol inhibited thrombin-, PAF- and vasopressin-induced elevation of [Ca2+], and [2+P]PtdOH formation. It is suggested that protein kinase C may act to terminate the transduction processes that link receptor occupancy to cellular activation.  相似文献

Hypocotyl-derived calli of genotypes and segregating populations of Coffea arabica, differing in susceptibility to Colletotrichum kahawae, were used to produce cell suspensions and protoplasts which were exposed to partially purified culture filtrates (PPCFs) prepared from the pathogen. The growth and viability of PPCF-treated cells and protoplasts were measured using packed cell volume, fluorescein diacetate staining and a colorimetric assay involving the tetrazolium salt MTT. Differential responses of cells and protoplasts were influenced by genotype, time of exposure and PPCF concentration. Protoplasts of resistant genotypes responded differentially from more susceptible genotypes as early as 4 h after challenge with the phytotoxin, suggesting that they were more sensitive than cell suspensions to the treatments. Protoplasts exposed to PPCFs from C. kahawae may therefore be used to screen and select genotypes resistant to, or tolerant of, coffee berry disease. A mutation (byp24) affecting the N-terminal region of p23 will suppress the lethal effects of am and ts mutations in gene 24. In the presence of normal p24, the byp24 alteration causes a delay in the cleavage of capsid proteins and the assembly of a high percentage of isometric, short-headed particles; therefore, the byp24 mutation can affect the length of the T4 capsid. In the absence of p24, 24?byp24 double mutants show a reduced rate of cleavage of capsid precursor proteins, and a reduced rate of virus assembly.Iminunoprecipitation with anti-p24 serum has shown the presence of both p24 and p24c in wild-type phage particles. The 24?byp24 particles contain no p24 or p24c, as determined by immunoprecipitation, urea/acrylamide gel electrophoresis, and two-dimensional isoelectric focusing, urea/acrylamide gradient gel electrophoresis. They have a normal electron microscopic appearance, pH stability, and heat stability; but they are more resistant to osmotic shock than wild-type T4. We suggest that p24 normally functions in the initiation of phage T4 capsid protein cleavage reactions.  相似文献

A conceptual model describing the response of two Australian floodplain eucalypts, river red gum (Eucalyptus camaldulensis) and black box (Eucalyptus largiflorens), to changes in water availability was developed based on field observations. This model was incorporated into a percentage based visual method estimating two tree crown parameters, crown extent and density. Extent is the amount of foliage at the periphery of the assessable crown; density is the density of assessable crown foliage. Polychoric correlation was used to determine the level of agreement between two experienced observers assessing river red gum and black box trees using a simple percentage scale and a percentage scale supported by the conceptual model. Trees were evaluated using the model by determining their position on a trajectory of water stress related decline and response. In both cases observer estimates of crown extent and density were significantly correlated. With the exception of red gum crown density the correlation coefficients were higher for the model supported scale. Using a conceptual model of tree response to water availability improved observer agreement. Supporting subjective assessment systems with a conceptual model is recommended to improve observer agreement in cases where a distinct model of the dominant stressor can be defined.  相似文献

Hyaluronan (HA) stimulates the motility of some but not all cell types. Here, we show that HA-promoted random motility of ras-transformed 10T1/2 (C3) fibroblasts requires activation of protein kinase C and is associated with rapid uptake of HA in a CD44 and RHAMM-dependent manner. The addition of HA to parental 10T1/2 fibroblasts (parental cells) does not stimulate random motility, but these cells can be 'primed' to respond to HA by treatment with the phorbol ester, PMA, for 4-6 h. This effect of PMA requires protein synthesis, PKC activity and is associated with enhanced uptake of HA. These results suggest that the ability of cells to respond to HA is regulated by a protein kinase C-dependent process that may promote uptake of HA.  相似文献

Ribes nigrum germplasm was screened for random amplified polymorphic DNA (RAPD) markers. Fiftyfour markers were identified which generated individual fingerprints for each of 21 cultivars. Genetic variation within R. nigrum germplasm, as detected by RAPDs, demonstrated that the genetic basis for improvement of blackcurrant is narrower than would be expected by the analysis of parentage.  相似文献

Transgenic crops genetically engineered for enhanced insect resistance should be compatible with other components of IPM for the pest resistance to be durable and effective. An experimental potato line was genetically engineered to express an anti-aphid plant protein (snowdrop lectin, GNA), and assessed for possible interactions of the insect resistance gene with a beneficial pest predator. These extended laboratory studies are the first to demonstrate adverse tri-trophic interactions involving a lectin- expressing transgenic crop, a target pest aphid and a beneficial aphidophagous predator. When adult 2-spot ladybirds (Adalia bipunctata[L.]) were fed for 12 days on peach-potato aphids (Myzus persicae Sulzer) colonising transgenic potatoes expressing GNA in leaves, ladybird fecundity, egg viability and longevity significantly decreased over the following 2–3 weeks. No acute toxicity due to the transgenic plants was observed, although female ladybird longevity was reduced by up to 51%. Adverse effects on ladybird reproduction, caused by eating peach-potato aphids from transgenic potatoes, were reversed after switching ladybirds to feeding on pea aphids from non-transgenic bean plants. These results demonstrate that expression of a lectin gene for insect resistance in a transgenic potato line can cause adverse effects to a predatory ladybird via aphids in its food chain. The significance of these potential ecological risks under field conditions need to be further evaluated.  相似文献
