Summary The presence of aromatic 1-amino acid decarboxylase (AADC) in nerve cell bodies of the intrinsic plexuses of the guinea-pig small intestine was demonstrated by incubating segments of intestine with 1-dopa in the presence of an inhibitor of monoamine oxidase, pargyline. After such incubation, some nerve cell bodies gave a fluorescence histochemical reaction indicative of the presence of a decarboxylated product of 1-dopa, probably dopamine. No fluorescence reaction occurred in the unincubated control or if the inhibitor of AADC, RO 4-4602, was included in the incubation mixture. The AADC-containing cell bodies apparently do not take up and store dopamine, because no fluorescence could be detected after incubation with dopamine and a monoamine oxidase inhibitor. The AADC-containing cells were found in about half of the ganglia of the submucous plexus of the guinea-pig small intestine, but were considerably less frequent in the myenteric plexus. They were also found in the other areas examined in this study, that is, in both enteric plexuses of the guinea-pig distal colon and of the small intestines of rabbits and rats.  相似文献

The effect of Mn2+ on the pattern of emergence of enzymes in rat liver and adipose tissue was studied in weaned rats given a milk diet (high fat) or sucrose-casein diet (high carbohydrate) for three weeks. Addition of Mn2+ to the high fat diet was associated with induction of key glycolytic, lipogenic and pentose pathway enzymes in both liver and adipose tissue; parallel increases were found in the incorporation of [1-14C] glucose into lipid and CO2. Mn2+ induced a change in the profile of enzyme activity similar in pattern to that found in rats given a high sucrose diet or that produced by insulin treatment. Mn2+ appears partially to overcome the regulatory feed-back mechanisms of the high fat diet and to provide a signal for the coordinated increase of glucose catabolic and lipogenic processes.  相似文献

—Rabbit vagus nerves and nodose ganglia were incubated in vitro for up to 24 h in two-compartment chambers. After the introduction of [3H]leucine or [3H]fucose to the ganglion compartments a rapid anterograde axonal transport of labelled proteins or glycoproteins occurred at rates of 330 ± 44 mm/day and 336 ± 30 mm/day respectively. Accumulation of [3H]leucine-labelled proteins proximal to a ligature on the nerve was unaffected by a delay of up to 6 h between removal of the nerve and labelling in vitro. Accumulation was prevented by inhibition of protein synthesis in the ganglion but not in the axon and was inhibited in a graded manner by colchicine.  相似文献

Testis nuclei of hypophysectomized rats selectively accumulate labeled testosterone and 5alpha-dihydrotestosterone following the injection of tritiated testosterone in vivo. Testosterone and 5alpha-dihydrotestosterone are bound to macromolecules in nuclei and can be extracted with 0.5 M KCl. Accumulation of protein bound radioactive androgens in nuclei of isolated seminiferous tubules is similar to that of whole testis. The relative amounts of testosterone and dihydrotestosterone in purified nuclei were similar to the relative amounts bound to cytoplasmic receptors, suggesting that cytoplasmic androgen-receptor complexes may be transported into the nuclei. Binding of labeled androgen is saturable and inhibited by prior injection of unlabeled testosterone or cyproterone acetate. Nuclear binding sites are destroyed by the proteolytic enzyme pronase, but not by DNase. Like the cytoplasmic androgen-receptor complexes in rat testis, nuclear androgen-protein complexes are heat labile and dissociate slowly at 0 degrees C. androgens fail to accumulate in testis nuclei of the Stanley-Gumbreck androgen insensitive rat, a species lacking cytoplasmic androgen receptors in testis and other androgen target tissues.  相似文献

Multiplying the germline would increase the number of offspring that can be produced from selected animals, accelerating genetic improvement for livestock breeding. This could be achieved by producing multiple chimaeric animals, each carrying a mix of donor and host germ cells in their gonads. However, such chimaeric germlines would produce offspring from both donor and host genotypes, limiting the rate of genetic improvement. To resolve this problem, we disrupted the RNA‐binding protein DAZL and generated germ cell‐deficient host animals. Using Cas9‐mediated homology‐directed repair (HDR), we introduced a DAZL loss‐of‐function mutation in male ovine fetal fibroblasts. Following manual single cell isolation, 4/48 (8.3%) of donor cell strains were homozygously HDR‐edited. Sequence‐validated strains were used as nuclear donors for somatic cell cloning to generate three lambs, which died at birth. All DAZL null male neonatal sheep lacked germ cells on histological sections and showed greatly reduced germ cell markers. Somatic cells within their testes were morphologically intact and expressed normal levels of lineage‐specific markers, suggesting that the germ cell niche remained intact. This extends the DAZL mutant phenotype beyond mice into agriculturally relevant ruminants, providing a pathway for using absolute germline transmitters in rapid livestock improvement.  相似文献

小麦族(Triticeae)是禾本科、早熟禾亚科中一个有重要经济价值、以多年生植物占优势的族,族内绝大多数种类是重要的粮食作物和畜牧业上的优良牧草,饲用价值极高,有些种类具有耐寒、耐旱、耐碱等特性,是农牧业上良种繁育、牧草利用的重要基因资源。但该族同时又是分类学上的一个疑难族,各学者对族内系统分类意见不一、争议颇大,尤其在族的界限、族下类群划分以及类群演化关系上问题较多,至今尚未解决。查阅了国内外分类学文献,探讨其分类差异以及存在问题,为充分开发利用中国丰富的小麦族植物资源提供理论依据。  相似文献
