The omega-conotoxins from the venom of fish-hunting cone snails are probably the most useful of presently available ligands for neuronal Ca channels from vertebrates. Two of these peptide toxins, omega-conotoxins MVIIA and MVIIB from the venom of Conus magus, were purified. The amino acid sequences show significant differences from omega-conotoxins from Conus geographus. Total synthesis of omega-conotoxin MVIIA was achieved, and biologically active radiolabeled toxin was produced by iodination. Although omega-conotoxins from C. geographus (GVIA) and C. magus (MVIIA) appear to compete for the same sites in mammalian brain, in amphibian brain the high-affinity binding of omega-conotoxin MVIIA has narrower specificity. In this system, it is demonstrated that a combination of two omega-conotoxins can be used for biochemically defining receptor subtypes and suggested that these correspond to subtypes of neuronal Ca2+ channels.  相似文献

Tannic acid (TA) is a naturally occurring polyphenolic compound that aggregates membranes and neutral phosolipid vesicles and precipitates many proteins. This study analyzes TA binding to lipid membranes and the ensuing aggregation. The optical density of dispersions of phosphatidylcholine (PC) vesicles increased upon the addition of TA and electron micrographs showed that TA caused the vesicles to aggregate and form stacks of tightly packed disks. Solution calorimetry showed that TA bound to PC bilayers with a molar binding enthalpy of -8.3 kcal/mol and zeta potential measurements revealed that TA imparted a small negative charge to PC vesicles. Monolayer studies showed that TA bound to PC with a dissociation constant of 1.5 microM and reduced the dipole potential by up to 250 mV. Both the increase in optical density and decrease in dipole potential produced by TA could be reversed by the addition of polyvinylpyrrolidone, a compound that chelates TA by providing H-bond acceptor groups. NMR, micropipette aspiration, and x-ray diffraction experiments showed that TA incorporated into liquid crystalline PC membranes, increasing the area per lipid molecule and decreasing the bilayer thickness by 2 to 4%. 2H-NMR quadrupole splitting measurements also showed that TA associated with a PC molecule for times much less than 10(-4) s. In gel phase bilayers, TA caused the hydrocarbon chains from apposing monolayers to fully interdigitate. X-ray diffraction measurements of both gel and liquid crystalline dispersions showed that TA, at a critical concentration of about 1 mM, reduced the fluid spacing between adjacent bilayers by 8-10 A. These data place severe constraints on how TA can pack between adjacent bilayers and cause vesicles to adhere. We conclude that TA promotes vesicle aggregation by reducing the fluid spacing between bilayers by the formation of transient interbilayer bridges by inserting its digallic acid residues into the interfacial regions of adjacent bilayers and spanning the interbilayer space.  相似文献

We have found that alamethicin, in the absence of an electric field, modifies both the hydrophilic surface and hydrophobic core of lipid bilayers. As shown by freeze-fracture and X-ray diffraction experiments with multiwalled vesicles, alamethicin increases the fluid space between bilayers by as much as 50 nm, and at the same time perturbs the hydrocarbon regions of the bilayers. For suspensions of gel-state lipid treated with alamethicin, uniformly spaced rows of particles cover the fracture faces and corresponding linear arrays of stain-collecting depressions cover the hydrophilic surfaces. In the liquid-crystalline state, alamethicin induces an irregular granular texture on the fracture faces.  相似文献

The 1998 International Commission for Non-Ionising Radiation (ICNIRP) Guidelines for human exposure to radiofrequency (RF) fields contain a recommendation to assess the potential impact of metallic implants in workers exposed up to the allowable occupational field limits. This study provides an example of how numerical electromagnetic (EM) and thermal modelling can be used to determine whether scattered RF fields around metallic implants in workers exposed to allowable occupational ambient field limits will comply with the recommendations of relevant standards and guidelines. A case study is performed for plane wave exposures of a 50 mm diameter titanium cranioplasty plate, implanted around 5-6 mm under the surface of the forehead. The level of exposures was set to the ambient power flux density limits for occupational exposures specified in the 1998 ICNIRP guidelines and the current 1999 IEEE C95.1 standard over the frequency range 100-3000 MHz. Two distinct peak responses were observed. There was a resonant response for the whole implant at 200-300 MHz where the maximum dimension of the implant is around a third of the wavelength of the RF exposure. This, however, resulted in relatively low peak specific energy absorption rate (SAR) levels around the implant at the exposure limits. Between 2100-2800 MHz, a second SAR concentrating mechanism of constructive interference of the wave reflected back and forth between the air-scalp interface and the scalp-plate interface resulted in higher peak SARs that were within the allowable limits for the ICNIRP exposures, but not for the IEEE C95.1 exposures. Moreover, the IEEE peak SAR limits were also exceeded, to a lesser degree, even when the implant was not present. However, thermal modelling indicated that the peak SAR concentrations around the implant did not result in any peak temperature rise above 1 degrees C for occupational exposures recommended in the ICNIRP guidelines, and hence would not pose any significant health risk.  相似文献

Staining with 3,3' diaminobenzidine tetrahydrochloride (DAB) is a common method used for the detection of peroxidases. Using this histochemical staining method in conjunction with transmission electron microscopy, we observed oxidation of DAB that was localized to a discrete set of organelles displaying morphological similarity to small (75-90 nm diameter) versions of higher eukaryotic microbodies or peroxisomes. These single membrane bounded organelles were characterized by an asymmetrical matrix capable of oxidizing DAB to an electron dense inclusion. Oxidation of DAB was further found to be dependent upon hydrogen peroxide (H2O2) as a substrate. Given a lack of peroxisomal import proteins and enzymes, it is unlikely that these represent conventional peroxisomes. Rather, they likely represent specialized organelles containing endogenous peroxidase or pseudo-peroxidase activity.  相似文献
