Cytokines impair the function and decrease the viability of insulin-producing β-cells by a pathway that requires the expression of inducible nitric oxide synthase (iNOS) and generation of high levels of nitric oxide. In addition to nitric oxide, excessive formation of reactive oxygen species, such as superoxide and hydrogen peroxide, has been shown to cause β-cell damage. Although the reaction of nitric oxide with superoxide results in the formation of peroxynitrite, we have shown that β-cells do not have the capacity to produce this powerful oxidant in response to cytokines. When β-cells are forced to generate peroxynitrite using nitric oxide donors and superoxide-generating redox cycling agents, superoxide scavenges nitric oxide and prevents the inhibitory and destructive actions of nitric oxide on mitochondrial oxidative metabolism and β-cell viability. In this study, we show that the β-cell response to nitric oxide is regulated by the location of superoxide generation. Nitric oxide freely diffuses through cell membranes, and it reacts with superoxide produced within cells and in the extracellular space, generating peroxynitrite. However, only when it is produced within cells does superoxide attenuate nitric oxide-induced mitochondrial dysfunction, gene expression, and toxicity. These findings suggest that the location of radical generation and the site of radical reactions are key determinants in the functional response of β-cells to reactive oxygen species and reactive nitrogen species. Although nitric oxide is freely diffusible, its biological function can be controlled by the local generation of superoxide, such that when this reaction occurs within β-cells, superoxide protects β-cells by scavenging nitric oxide. 相似文献

A directed nucleotide-sequencing approach for single-stranded vectors based on recloning intermediates of a progressive DNA synthesis reaction 总被引:2,自引:0,他引:2
A simple method for site-directed nucleotide sequencing is presented that uses a novel procedure for generating nested 'deletions' within inserts of single-stranded clones. In this method, single-stranded template, sequencing primer, and the Klenow fragment of Escherichia coli DNA polymerase I are used to initiate progressive DNA synthesis of the entire insert of the clone. By time-dependent sampling and pooling of intermediates from the synthesis reaction a series of nested double-stranded DNA subfragments of the insert can be created. Nested subclones are then produced by S1-endonuclease treatment and oriented subcloning methods. First, smaller quantities of template DNA can be used, equivalent to a fraction of a small DNA sequencing prep. Second, it works with single-stranded M13 phage DNA rather than requiring the preparation of double-stranded replicative form DNA as in ExoIII-based methods. Third, the 'deletions' it generates can span areas of simple nucleotide sequence or secondary structure that often halt digestion in the single-stranded exonuclease-based method. Last, the method is adaptable to a larger variety of insert cloning sites than the ExoIII-based method. The main disadvantage of the method is that, due to the lower efficiency of subcloning larger DNA fragments, subclone inserts larger than 3 kb are generated only infrequently. 相似文献

Evidence is mounting that female animals use egg‐yolk compounds (e.g. steroids, antioxidants) to adaptively engineer the quality of their offspring as a function of several maternal and environmental factors. Though adjustments to yolk allocation have been well‐characterized as a function of parental phenotypes, we know very little about how an individual's social environment influences yolk composition. Here, we consider how two types of yolk compounds, androgens and carotenoids, relate to the maternal social environment during the egg‐laying period, controlling statistically for known correlations between various aspects of parental quality and egg yolk compounds. Barn swallows Hirundo rustica erythrogaster breed in groups of highly variable size and spacing, allowing us to test whether or not the social environment is correlated with these maternal effects. We found no relationship between carotenoid levels in eggs as a function of colony size, colony density, or nearest‐neighbor distance. However, eggs from females in larger groups had lower concentrations and total amounts of yolk androgens than those from females in smaller, less dense social settings. Our results counter previous predictions and literature, showing that females breeding in large groups deposit more androgen in eggs, mechanistically, because they compete more with conspecifics and have higher circulating androgen levels themselves and, functionally, because it could be advantageous for their offspring to show high androgen‐mediated competitive abilities early in life. Instead, because group size in this species is governed largely by site fidelity and the availability of old nests for re‐use, and because reproductive output does not differ as a function of group size, it may be that competition is greater for limited nests in small groups, thus elevating androgen levels. Further, yolk androgens were previously shown to be affected by male quality, and the greater concentrations and amounts of yolk androgens in smaller sites may reflect differential allocation to darker males found at these sites. 相似文献

Rapid climate change across the globe is having dramatic effects on wildlife. Responses of organisms to shifting thermal conditions often include physiological and behavioural accommodations, but to date these have been largely viewed and studied as naturally evolved phenomena (e.g. heat avoidance, sweating, panting) and not necessarily as strategies where animals exploit other anthropogenic conditions or resources. Moreover, the degree to which native versus introduced species show thermal plasticity has generated much conservation and ecological interest. We previously have observed introduced rosy-faced lovebirds (Agapornis roseicollis) perching in the relief-air vents on building faces in the Phoenix, Arizona, USA, metropolitan area, but doing so only during summer. Here, we show that such vent-perching events are significantly associated with extreme outdoor summer temperatures (when daily local highs routinely exceed 40°C). In fact, the temperature threshold at which we detected lovebirds starting to perch in cool air vents mirrors the upper range of the thermoneutral zone for this species. These results implicate novel, facultative use of an anthropogenic resource—industrial air-conditioning systems—by a recently introduced species (within the last 35 years) to cool down and survive extremely hot conditions in this urban ‘heat-island'' environment. 相似文献

The genetic relatedness among 96 invasive Escherichia coli belonging to several serogroups and 13 non-invasive of several serotypes that share the same O antigen was investigated by multilocus enzyme electrophoresis analysis. The invasive strains were isolated in different parts of the world and most of them recovered from dysentery. Twenty-nine electrophoretic types were distinguished and the most invasive strains were found to belong to two major lineages. These results suggested that the invasive ability in these strains has evolved in divergent chromosomal backgrounds, presumably through the horizontal spread of plasmid-borne invasion genes. The maintenance of invasive phenotypes in separate lineages suggests that this ability confers a selective advantage to invasive strains. 相似文献
