The characterization of Campylobacter jejuni has been significantly improved by the use of multilocus sequence typing (MLST), which allows the relationship between isolates to be determined. The sequence types (STs) of 261 isolates of C. jejuni from New Zealand were determined. Isolates were obtained from a range of sources including chicken meat, cattle, pigs, duck, sheep, water and human infections. Thirty-two new alleles and 44 new STs were identified. Comparison of the MLST data and pulsed-field gel electrophoresis macrorestriction profiles showed that the macrorestriction profiles were good predictors of the clonal complex (CC) but not ST. All the major CCs identified elsewhere in the world were found in New Zealand as well as the association of certain CCs with particular animal niches. Tyrosyl aryl dipeptide inhibitors of S. aureus tyrosyl tRNA synthetase have been identified with IC50 values down to 0.5 microM. In a model system, consistent transfection of chick embryo fibroblasts (CEF) by DNA from the XC cell line occurred, with recovery of infectious Rous sarcoma virus. The techniques were then applied in attempts to recover possible human tumour viruses. Even with various modifications of the XC technique, DNA from three human malignant melanoma cell lines failed to infect adult or foetal human fibroblasts, although melanoma DNA was taken up into nuclei of target cells. XC DNA did not transfect human foetal fibroblasts and melanoma DNA was ineffective in CEF. DNA from the Raji (Epstein-Barr virus non-producer) and QIMR-WIL (producer) lymphoblastoid cell lines did not transfect human cord blood lymphocytes or amnion cells. These broadly applicable techniques therefore failed to recover EB virus, the putative melanoma retrovirus, or other potential tumour virus. 相似文献

Nest predation is a major limiting factor for songbird productivity, including the federally endangered black-capped vireo (Vireo atricapilla). However, nest predator information is limited across the range of the black-capped vireo in central and southwest Texas. We monitored nests in 3 counties within the breeding range of black-capped vireos in Texas in 2008 and 2009 and used continuous recording digital video cameras to record predation events. We video-monitored 115 nests and documented 39 predation events by at least 9 predator species. Overall, we observed avian species (51%, n = 39), specifically brown-headed cowbirds (Molothrus ater; n = 12), and snakes (26%, n = 39) as the most frequent nest predators. The estimated daily nest survival rate during the laying and incubation stage was 0.985 (95% CI = 0.967–0.993) and 0.944 (95% CI = 0.921–0.961) during the nestling stage. In addition, we analyzed models of predator-specific nest predation using multinomial logistic regression. Effect of nest height on predation rate was significant for snakes; nest stage was significant for nests depredated by avian predators. By identifying and increasing our knowledge of nest predators and vegetation characteristics associated with greater risk of predation in multiple locations within the black-capped vireo's range, we can effectively manage habitat to benefit recovery efforts of the species. © 2012 The Wildlife Society. 相似文献

Phylogenetic clustering of wingbeat frequency and flight‐associated morphometrics across insect orders 下载免费PDF全文
Wingbeat frequency in insects is an important variable in aerodynamic and energetic analyses of insect flight and often is studied on a family‐ or species‐level basis. Meta‐analyses of these studies report order‐level patterns suggesting that flight strategy is moderately well conserved phylogenetically. Studies incorporated into these meta‐analyses, however, use variable methodologies across different temperatures, which may confound results and phylogenetic patterns. In the present study, a high‐speed camera is used to measure wingbeat frequency in a wide variety of species (n = 102) under controlled conditions aiming (i) to determine the validity of previous meta‐analyses showing phylogenetic clustering of flight strategy and (ii) to identify new evolutionary patterns between wingbeat frequency, body mass, wing area, wing length and wing loading at the order level. All flight‐associated morphometrics significantly affect wingbeat frequency. Linear models show that wing area explains the most amount of variation in wingbeat frequency (r2 = 0.59, P ≤ 0.001), whereas body mass explains the least (r2 = 0.09, P ≤ 0.01). A multiple regression model incorporating both body mass and wing area is the best overall predictor of wingbeat frequency (r2 = 0.84, P ≤ 0.001). Order‐level phylogenetic patterns across relationships are consistent with previous studies. Thus, the present study provides experimental validation of previous meta‐analyses and provides new insights into phylogenetically conserved flight strategies across insect orders. 相似文献
