Summary The habitat hectares approach is an explicit, quantitative method for assessing the quality of vegetation by adding scores that are assigned to 10 habitat attributes. We believe it will be more repeatable and transparent than other methods that rely on subjective judgement. However, we have four principal criticisms of the method as it is currently proposed: (i) measurement of some of the attributes may be subject to considerable error that varies among assessors; (ii) the comparison of each measure with a single benchmark does not accommodate appropriate disturbance regimes; (iii) the proposed combination of attributes leads to some apparent internal inconsistencies; and (iv) it is not clear how the method will actually be used in practice. We suggest modifications to address these concerns and improve the proposed method. Finally, we make additional suggestions about the method's potential application, including: separate reporting of the extent and quality of different vegetation types to avoid the inappropriate combination of measures of area and quality; valuing appropriate disturbance regimes in natural areas; and considering very carefully the application of compensation or mitigation measures.  相似文献

Base-modification can occur throughout a transfer RNA molecule; however, elaboration is particularly prevalent at position 34 of the anticodon loop (the wobble position), where it functions to influence protein translation. Previously, we demonstrated that the queuosine modification at position 34 can be substituted with an artificial analogue via the queuine tRNA ribosyltransferase enzyme to induce disease recovery in an animal model of multiple sclerosis. Here, we demonstrate that the human enzyme can recognize a very broad range of artificial 7-deazaguanine derivatives for transfer RNA incorporation. By contrast, the enzyme displays strict specificity for transfer RNA species decoding the dual synonymous NAU/C codons, determined using a novel enzyme-RNA capture-release method. Our data highlight the broad scope and therapeutic potential of exploiting the queuosine incorporation pathway to intentionally engineer chemical diversity into the transfer RNA anticodon.  相似文献

The atrial wall of Notophthalmus viridescens is 25–75 μm thick and is trabeculated sparsely. Coronary vessels are absent. The endocardial endothelium is continuous and has 50–60 nm-wide fenestrae with diaphragms, rests on a discontinuous basal lamina and lacks occluding junctions. Cells found in the subendothelial connective tissue are xanthophores, melanophores, mast cells, fibroblasts, macrophages, and unmyelinated nerve fibers with Schwann cell investments. Epicardial mesothelial cells contain numerous 6–7 nm filaments and lamellar bodies which resemble myelin figures. Mesothelial cell junctions include maculae adhaerentes diminutae, desmosomes, and interdigitations. The epicardial connective tissue layer is more extensive than that of the endocardium, with xanthophores and melanophores rarely present and nerve fibers never observed. The myocardium consists of a mesh-work of myocytes 3–5 cell layers thick with little intervening connective tissue. Myocytes are 6–10 μm in diameter and have two or three peripheral myofibrillae. Typical A, I, H, Z, and M bands are present with a sarcomere length of 2.5 μm. T tubules are not observed. The sarcoplasmic reticulum has subsarcolemmal dilations. The nuclear pole region contains abundant mitochondria and atrial granules, extensive Golgi, and elements of smooth and rough-surfaced endoplasmic reticulum. Lateral intercellular junctions consisting of dense plaques, frequently continuous with Z-line material, are common. Oblique and transversely oriented junctions consisting of primarily of fascia adhaerentes, are present. It appears that amphibian atrial myocytes more closely resemble those of the amphibian ventricle than those of the mammalian atrium. Structural differences between amphibian atrial and ventricular myocytes seem to be quantitative rather than qualitative in nature.  相似文献

Several members of the transforming growth factor β (TGFβ) superfamily are expressed in the developing murine epidermis. Among these are TGFβ1, which is found in the basal layer, and bone morphogenetic protein (BMP)-6, located in the suprabasal layers. Although the role of TGFβ in cell growth has been studied extensively, little is known about the effects of these factors on keratinocyte differentiation. This study demonstrates that BMP-6 acts to positively regulate the differentiation of primary skin keratinocytes grown in culture. In contrast, TGFβ1 antagonizes keratinocyte differentiation blocking the upregulation of keratin markers by BMP-6. We show that the effects of BMP-6 on expression of keratin 1 (K1), a marker of differentiation, requires signaling through the Smad pathway. In addition, regulation of K1 levels by BMP-6 is modulated by the SEK signaling pathway. This suggests that regulation of keratinocyte differentiation by BMP-6 involves multiple signaling systems.  相似文献

ObjectiveDuring the COVID-19 pandemic, visits for diabetes care were abruptly canceled without predefined procedures to re-engage patients. This study was designed to determine how outreach influences patients to maintain diabetes care and identify factors that might impact the intervention’s efficacy.MethodsA diabetes nursing team attempted outreach for patients who had a canceled appointment for diabetes between March 16, 2020, and June 19, 2020. Outreach status was defined as reached, message left, or no contact. Outcomes were defined as follows: (1) booking and (2) keeping a follow-up appointment.ResultsSeven hundred eighty-seven patients were included (384 [49%] were reached, 152 (19%) were left a message, and 251 (32%) had no contact). Reached patients were more likely to book [odds ratio (OR) = 2.43, P < .001] and keep an appointment (OR = 2.39, P < .001) than no-contact patients. Leaving a message did not increase the odds of booking (OR = 1.05, P = .84) or keeping (OR = 1.17, P = .568) an appointment compared with no contact. Older age was a significant predictor of booking an appointment (OR = 1.014 for each year of age, P = .037). Patients on insulin were more likely to keep their appointment (OR = 1.70, P = .008). Patients with a higher hemoglobin A1C level were less likely to keep their appointment (OR = 0.87 for each 1.0% increase in the hemoglobin A1C level, P = .011).ConclusionThese findings suggest that to optimize re-engagement during care disruption, 1-way communication is no better than no contact and that 2-way communication increases the likelihood that patients will maintain access to care. In addition, although higher-risk patients (eg, patients with older age or those on insulin) may be more incentivized to stay engaged, targeted outreach is needed for those with chronically poor glycemic control.  相似文献

Planktonic sarcodines (acantharia, radiolaria, and planktonic foraminifera) are oceanic amoeboid protozoa that often harbor a variety of microalgae as intracellular symbionts. The identity and function of these endosymbiotic algae have intrigued and perplexed biologists for more than a century. The most conspicuous and well‐studied symbiotic algae of planktonic foraminifera and radiolaria are dinoflagellates, but a variety of nondinoflagellate taxa have also been reported. Ultrastructural features have been used to characterize some of these nondinoflagellate algae, but rarely has this led to clear taxonomic affiliations. We analyzed the nuclear small subunit ribosomal DNA (srDNA) isolated from the symbionts of the spinose planktonic foraminiferan Globigerinella siphonifera d'Orbigny (=Globigerinella aequilateralis Brady) and a solitary radiolarian (Spongodrymus sp. Haeckel) in order to determine the identity of these symbionts. The small coccoid algae isolated from G. siphonifera correspond to the Type I symbionts described by Faber et al. (1988) . Phylogenetic analysis of the srDNA sequences places these symbionts within the prymnesiophyte (haptophyte) lineage, closer to Prymnesium Conrad than to Phaeocystis Lagerheim. To our knowledge, this is the first confirmed case of a symbiotic prymnesiophyte. In addition, we were able to examine the level of sequence heterogeneity between symbionts isolated from different individuals of a single host species. The three isolates in this study had srDNA sequences that were almost identical, indicating that the three were all of the same species. Very green symbiotic algae were isolated from three solitary radiolaria identified as species of Spongodrymus. The symbiont srDNA sequences from the three individual hosts were identical to each other, again implicating a single species of algae in that symbiotic association. These symbionts are prasinophytes most closely related to the clade containing Tetaselmis convolutae Norris, Hori et Chihara. Tetraselmis convolutae is the algal symbiont of the marine flatworm, Convolutae roscoffensis Graff.  相似文献

ABSTRACT

Oceanic people and places are increasingly labelled as either ‘resilient’ or ‘vulnerable’ to disasters and climate change. Resilience is often described in disaster discourse as a strategy designed to overcome vulnerability by helping communities to ‘bounce back’ in the wake of ‘natural’ disasters. Using ethnographic research conducted with Community Disaster and Climate Change Committees (CDCs) in Vanuatu in the wake of Tropical Cyclone Pam, this paper seeks to problematise disaster responses that see the ‘community’ as a space to be acted upon by outsiders, or where people will respond in a unified way to the challenges of rebuilding after disaster. Using political ecology framings this paper critiques the ideas of resilience that appear entrenched in community-based disaster and climate change adaptation discourse and practice in Oceania. Rather than presupposing resilience or vulnerability, this paper details the dispersal and distribution power and agency amongst individual actors and groups that either supported or manipulated, the distribution of goods by Community Disaster Committees. In this way, it moves beyond the limitations of conceptual framings of resilience in disaster management and climate change into a more considered appraisal of power, by exploring what James Ferguson has termed ‘the politics of distribution’ in the context of disaster.  相似文献
