Here we report on the related TBC/RabGAPs EPI64A and EPI64B and show that they function to organize the apical aspect of epithelial cells. EPI64A binds the scaffolding protein EBP50/NHERF1, which itself binds active ezrin in epithelial cell microvilli. Epithelial cells additionally express EPI64B that also localizes to microvilli. However, EPI64B does not bind EBP50 and both proteins are shown to have a microvillar localization domain that spans the RabGAP domains. CRISPR/Cas9 was used to inactivate expression of each protein individually or both in Jeg-3 and Caco2 cells. In Jeg-3 cells, loss of EPI64B resulted in a reduction of apical microvilli, and a further reduction was seen in the double knockout, mostly likely due to misregulation of Rab8 and Rab35. In addition, apical junctions were partially disrupted in cells lacking EPI64A and accentuated in the double knockout. In Caco2 loss of EPI64B resulted in wavy junctions, whereas loss of both EPI64A and EPI64B had a severe phenotype often resulting in cells with a stellate apical morphology. In the knockout cells, the basal region of the cell remained unchanged, so EPI64A and EPI64B specifically localize to and regulate the morphology of the apical domain of polarized epithelial cells.  相似文献

Tree House Explorer (THEx) is a genome browser that integrates phylogenomic data and genomic annotations into a single interactive platform for combined analysis. THEx allows users to visualize genome-wide variation in evolutionary histories and genetic divergence on a chromosome-by-chromosome basis, with continuous sliding window comparisons to gene annotations, recombination rates, and other user-specified, highly customizable feature annotations. THEx provides a new platform for interactive phylogenomic data visualization to analyze and interpret the diverse evolutionary histories woven throughout genomes. The impacts of wind energy on bat populations is a growing concern because wind turbine blades can strike and kill bats, and wind turbine development is increasing. We tested the effectiveness of 2 management actions at 2 wind-energy facilities for reducing bat fatalities: curtailing turbine operation when wind speeds were <5.0 m/second and combining curtailment with an acoustic bat deterrent developed by NRG Systems. We measured the effectiveness of the management actions using differences in counts of bat carcasses quantified by daily and twice-per-week standardized carcass searches of cleared plots below turbines, and field trials that estimated searcher efficiency and carcass persistence. We studied turbines located at 2 adjacent wind-energy facilities in northeast Illinois, USA, during fall migration (1 Aug–15 Oct) in 2018. We estimated the effectiveness of each management action using a generalized linear mixed-effects model with several covariates. Curtailment alone reduced overall bat mortality by 42.5% but did not reduce silver-haired bat (Lasionycteris noctivagans) mortality. Overall bat fatality rates were 66.9% lower at curtailed turbines with acoustic deterrents compared to turbines that operated at manufacturer cut-in speed. Curtailment and the deterrent reduced bat mortality to varying degrees between species, ranging from 58.1% for eastern red bats (Lasiurus borealis) to 94.4 for big brown bats (Eptesicus fuscus). Hoary (Lasiurus cinereus) and silver-haired bat mortality was reduced by 71.4% and 71.6%, respectively. Our study lacked a deterrent-only treatment group because of the expense of acoustic deterrents. We estimated the additional reduction in mortality with concurrent deployment of the acoustic deterrent and curtailment under the assumption that curtailment and the acoustic deterrent would have reduced mortality by the same percentage at adjacent wind-energy facilities. Acoustic deterrents resulted in 31.6%, 17.4%, and 66.7% additional reductions of bat mortality compared to curtailment alone for eastern red bat, hoary bat, and silver-haired bat, respectively. The effectiveness of acoustic deterrents for reducing bat mortality at turbines with rotor-swept area diameters >110 m is unknown because high frequency sound attenuates quickly, which reduces coverage of rotor-swept areas. Management actions should consider species differences in the ability of curtailment and deterrents to reduce bat mortality and increase energy production.  相似文献

Globally, climate is changing rapidly, which causes shifts in many species' distributions, stressing the need to understand their response to changing environmental conditions to inform conservation and management. Northern latitudes are expected to experience strongest changes in climate, with milder winters and decreasing snow cover. The wolverine (Gulo gulo) is a circumpolar, threatened carnivore distributed in northern tundra, boreal, and subboreal habitats. Previous studies have suggested that wolverine distribution and reproduction are constrained by a strong association with persistent spring snow cover. We assess this hypothesis by relating spatial distribution of 1589 reproductive events, a fitness-related proxy for female reproduction and survival, to snow cover over two decades. Wolverine distribution has increased and number of reproductive events increased 20 times in areas lacking spring snow cover during our study period, despite low monitoring effort where snow is sparse. Thus, the relationship between reproductive events and persistent spring snow cover weakened during this period. These findings show that wolverine reproductive success and hence distribution are less dependent on spring snow cover than expected. This has important implications for projections of future habitat availability, and thus distribution, of this threatened species. Our study also illustrates how past persecution, or other factors, that have restricted species distribution to remote areas can mask actual effects of environmental parameters, whose importance reveals when populations expand beyond previously restricted ranges. Overwhelming evidence shows that climate change is affecting many species and ecological processes, but forecasting potential consequences on a given species requires longitudinal data to revisit hypotheses and reassess the direction and magnitude of climate effects with new data. This is especially important for conservation-oriented management of species inhabiting dynamic systems where environmental factors and human activities interact, a common scenario for many species in different ecosystems around the globe.  相似文献

Porolithon is one of the most ecologically important genera of tropical and subtropical crustose (non-geniculate) coralline algae growing abundantly along the shallow margins of coral reefs and functioning to cement reef frameworks. Thalli of branched, fruticose Porolithon specimens from the Indo-Pacific Ocean traditionally have been called P. gardineri, while massive, columnar forms have been called P. craspedium. Sequence comparisons of the rbcL gene both from type specimens of P. gardineri and P. craspedium and from field-collected specimens demonstrate that neither species is present in east Australia and instead resolve into four unique genetic lineages. Porolithon howensis sp. nov. forms columnar protuberances and loosely attached margins and occurs predominantly at Lord Howe Island; P. lobulatum sp. nov. has fruticose to clavate forms and free margins that are lobed and occurs in the Coral Sea and on the Great Barrier Reef (GBR); P. parvulum sp. nov. has short (<2 cm), unbranched protuberances and attached margins and is restricted to the central and southern GBR; and P. pinnaculum sp. nov. has a mountain-like, columnar morphology and occurs on oceanic Coral Sea reefs. A rbcL gene sequence of the isotype of P. castellum demonstrates it is a different species from other columnar species. In addition to the diagnostic rbcL and psbA marker sequences, the four new species may be distinguished by a combination of features including thallus growth form, margin shape (attached or unattached), and medullary system (coaxial or plumose). Porolithon species, because of their ecological importance and sensitivity to ocean acidification, need urgent documentation of their taxonomic diversity.  相似文献
