Summary The sensitivity to calcium of the human squamous carcinoma cell line, SCC-13, was demonstrated and characterized. Cultures
grown to confluence in the presence of 0.2 to 2 mM calcium had approximately 10-fold higher levels of particulate transglutaminase activity and envelope competence than those
grown in low calcium (0.025 to 0.05 mM) medium. Raising the calcium from 0.025 to 1.8 mM induced expression of this enzyme and of competence over the course of a week. Conversely, for cultures grown to confluence
in 1.8 mM calcium, subsequent reduction of calcium to 0.025 mM resulted in a substantial decline in transglutaminase over a similar time period. Immunoprecipitable transglutaminase was
clearly identifiable in cultures grown in 1.8 mM calcium-containing medium but not in those grown in low calcium medium or in the presence of retinoic acid, suggestive of
regulation at the level of mRNA accumulation or translation rather than posttranslational modification.

Plantlets of Dendranthema grandiflora Pennine Reel were grown from nodal sections on Sorbarods saturated with liquid medium containing 0–3 mg 1–1 of various growth retardants. After 4 weeks they were transferred to compost and maintained at a relative humidity of 42% at 27.5°C. Wilting was assessed over a period of 3 h. Plantlets treated with paclobutrazol, flurprimidol, triapenthenol, chlorphonium chloride, uniconazol and ancymidol showed dose-related reductions in wilting up to a concentration of 3 mg 1–1. Responses to tetcyclacis and mepiquat chloride were weaker, and no responses to chlormequat chloride, BTS 44584 or diaminozide could be detected. An early successional wetland complex on a reclaimed surface coal mine in southern Illinois was studied 1985–1987. Seasonally, biomass was low, with above-ground values of 10–210g m–2 and below-ground biomass of 1.5–2435 g m–2. Biomass peaked in spring and did not vary much throughout the remainder of the growing season. Stem densities were high (179–1467 m–2) because large numbers of seedlings became established as falling water levels exposed large areas of mudflats. Fluctuating water levels led to a lack of community zonation. Species diversity (H) was low to moderate over all sites with diversity values ranging between 1.86 and 3.27. 相似文献

Summary C57BL mice inoculated with radiation leukemia virus (RadLV) develop preleukemic cells long before the onset of leukemia. These cells are potentially immunogenic but fail to elicit an immune response in the host because of the appearance of virus-specific suppressor T cells. We have studied the effect of polysaccharide K (PSK) on the generation of RadLV-specific cell-mediated immune responses in vitro. Long-term exposure to PSK in culture potentiated the ability of immunized T cells to respond to a RadLV-induced lymphoma. It also abrogated the suppressive activity of suppressor T cells and simultaneously boosted the ability of reactive T cells to respond. The dual immunostimulating activity of PSK resulted in the generation of T cytotoxic lymphocytes that could lyse lymphoma cells in vitro. The results suggest that PSK could be used as a prophylactic immune response modifier in preleukemia. 相似文献

Elemental analyses of mammalian bone (e.g., strontium-calcium ratios, or Sr/Ca) distinguish between herbivores and carnivores; however, the relationships among herbivores are unclear. To study this question, a modern faunal sample from the Nagupande Tsetse Control Area (Zambezi drainage, Northwestern Zimbabwe) was used. This collection has the advantage of well-established geographical controls in addition to a varied fauna, which includes both bovids and suids. The grazing/browsing dietary status of each species was ascertained by means of isotopic analysis of carbon. Clear differences were seen in the δ13C of grazing and browsing animals; a specialized grazer was found to have significantly lower Sr/Ca than less specialized grazers and browsers. In this study it was also possible to examine differences in Sr/Ca by sex; female warthogs were found to have significantly lower Sr/Ca than males. The variation in certain animal groups was found to be abnormal. Implications for reconstruction of prehistoric human diets using trace-element techniques are discussed. 相似文献

Proteoglycans were extracted under nondissociative conditions from superficial and deeper layers of dog normal articular cartilage. The purified a-A1 preparations were characterized by velocity gradient centrifugation. Superficial specimens exhibited an abundant population of slow sedimenting aggregates whereas the aggregates of deeper preparations sedimented as two well-defined families of molecules. These dissimilarities in the size distribution of the aggregates observed between superficial and deeper a-A1 preparations derived most of all from differences in their content of hyaluronate and link proteins: (a) superficial preparations contained twice as much hyaluronate as deeper specimens; (b) superficial aggregates were link-free and unstable at pH 5.0 whereas deeper preparations contained link-proteins and their faster sedimenting aggregates were stabilized against dissociation at pH 5.0. In these proteoglycan preparations from different cartilage layers, the monomers exhibited an identical capacity for aggregation and the hyaluronate molecules displayed quite similar molecular weight (Mr = 5 x 10(5] and aggregating capacity. These observations as well as aggregating studies conducted with highly purified link protein and purified hyaluronate specimens of different molecular weights support the following conclusions: (a) link protein not only stabilizes proteoglycan aggregates but also enhances the aggregating capacity of hyaluronate; (b) for all practical purposes, the slow sedimenting aggregates represent a secondary complex of hyaluronate and proteoglycan monomers whereas the fast sedimenting aggregates may be considered as a ternary complex wherein link protein stabilizes the hyaluronate-proteoglycans interaction; (c) the distinctive heterogeneity of articular cartilage can be related to structurally different proteoglycan aggregates. The structural dissimilarities observed between superficial and deeper aggregates could reflect the different macromolecular organization of the proteoglycan molecules in the territorial and interterritorial matrices, respectively. 相似文献

H-2u haplotype mice are unique among all E alpha+ strains because they do not provide in heterozygotes an E alpha chain that interacts with E betak,s,etc. sufficiently well to allow certain E-restricted immune responses. As a first step in understanding this peculiarity, we have sequenced E alpha u and E beta u cDNA and compared the derived amino acid sequences with those of previously analyzed alleles. Summary The subtidal coralline flats of northeastern New Zealand support a characteristic guild of grazing herbivores. The most important members of this guild are an echinometrid echinoid, patellid, turbinid and trochid gastropods. Densities of these herbivores fluctuate through time. Interactions within and among the different species of echinoids and gastropods were investigated experimentally. Different combinations of species were caged at densities up to 5 times that of ambient for a 24 week period in an experiment designed to differentiate between intra- and interspecific competition.The echinoidEvechinus chloroticus and the turbinid gastropodCookia sulcata exhibited reduced mean dry weight with increasing intraspecific densities. There was little evidence of density-related mortality in these species. The limpetCellana stellifera showed comparatively large losses of weight and enhanced mortalities in intraspecific experimental treatments but this was not related to density.Investigation of interspecific interactions amongstEvechinus andCookia revealed no evidence of a negative influence of one species on the other. In terms of dry weight,Cookia was indifferent to the presence ofEvechinus, andEvechinus benefited in the reciprocal interaction.Cookia also enjoyed an enhanced mean dry weight when in the presence ofCellana compared to the equivalent intraspecific treatments. There were no coherent trends in proportional mortality in any treatments with enhanced interspecific densities.
Cellana, in the presence ofCookia, exhibited a dramatic decrease in mortality rate and increase in mean dry weight. The presence of the turbinid gastropod was clearly beneficial to the limpet when compared to the intraspecific treatments with enhanced intraspecific densities and the control cages containingCellana at ambient density. We suggest that subtidal areas constitute poor habitats for limpets in the absence of agents such asCookia which may provide or maintain suitable sites for attachment and grazing.For the combinations of densities and species investigated there was a consistent trend towards positive interspecific interactions. It seems unlikely that at the sites investigated interspecific competition could act to restrict distributions, or limit abundances of species. 相似文献
