The Lkb1 tumour suppressor is a multitasking kinase participating in a range of physiological processes. We have determined the impact of Lkb1 deficiency on intestinal homeostasis, particularly focussing on secretory cell differentiation and development since we observe strong expression of Lkb1 in normal small intestine Paneth and goblet cells. We crossed mice bearing an Lkb1 allele flanked with LoxP sites with those carrying a Cyp1a1-specific inducible Cre recombinase. Lkb1 was efficiently deleted from the epithelial cells of the mouse intestine after intraperitoneal injection of the inducing agent β-naphthoflavone. Bi-allelic loss of Lkb1 led to the perturbed development of Paneth and goblet cell lineages. These changes were characterised by the lack of Delta ligand expression in Lkb1-deficient secretory cells and a significant increase in the levels of the downstream Notch signalling effector Hes5 but not Hes1. Our data show that Lkb1 is required for the normal differentiation of secretory cell lineages within the intestine, and that Lkb1 deficiency modulates Notch signalling modulation in post-mitotic cells.  相似文献

Aim To use biogeographical, palaeomagnetic, palaeosedimentary, and plate circuit data from Late Cretaceous regions in and around the Pacific to test the plate tectonic hypothesis of a pre‐Pacific superocean. Location East Asia, Australia, Antarctica, the western Americas, and the Pacific. Methods Literature surveys of the distributions of Cretaceous, circum‐Pacific taxa were compared with palaeomagnetic and palaeosedimentary data. Uncontroversial plate motions based on seafloor spreading data were also used to test the results of the biogeographical and palaeomagnetic analyses. Results The distributions of Cretaceous terrestrial taxa, mostly dinosaurs, imply direct, continental connections between Australia and East Asia, East Asia and North America, North America and South America, South America and Antarctica, and Antarctica and Australia. Palaeomagnetic, palaeosedimentary, and basic plate circuit analyses require little to no latitudinal motion of the Pacific plate with respect to the surrounding continents. Specifically, the data implies that western North America, East Asia, and the Pacific plate all increased in latitude by roughly the same amount (c. 11 ± 5°) since the Campanian – and that the Pacific Ocean Basin has increased in length north‐to‐south. Main conclusions Each of the analyses provides independent corroboration for the same conclusion: the Late Cretaceous Pacific plate was completely enclosed by the surrounding continents and has not experienced significant latitudinal motion with respect to North America, East Asia, or the Bering land bridge. This contrasts significantly with the plate tectonic history of the Pacific, implying instead that the Pacific plate formed in situ, pushing the continents apart as the plate and basin expanded. These results also substantiate recent biogeographical analyses that have concluded that a narrower Pacific Ocean Basin in the Mesozoic and early Tertiary provides the most reasonable explanation for the great number of trans‐Pacific disjunctions of poor dispersing taxa.  相似文献

A theory for optimal monitoring of marine reserves   总被引:3,自引:0,他引:3  
Monitoring of marine reserves has traditionally focused on the task of rejecting the null hypothesis that marine reserves have no impact on the population and community structure of harvested populations. We consider the role of monitoring of marine reserves to gain information needed for management decisions. In particular we use a decision theoretic framework to answer the question: how long should we monitor the recovery of an over‐fished stock to determine the fraction of that stock to reserve? This exposes a natural tension between the cost (in terms of time and money) of additional monitoring, and the benefit of more accurately parameterizing a population model for the stock, that in turn leads to a better decision about the optimal size for the reserve with respect to harvesting. We found that the optimal monitoring time frame is rarely more than 5 years. A higher economic discount rate decreased the optimal monitoring time frame, making the expected benefit of more certainty about parameters in the system negligible compared with the expected gain from earlier exploitation.  相似文献

Immunostimulatory receptors belonging to the tumor necrosis factor receptor (TNFR) superfamily are emerging as promising targets for cancer immunotherapies. To optimize the agonism of therapeutic antibodies to these receptors, Fc engineering of antibodies was applied to facilitate the clustering of cell surface TNFRs to activate downstream signaling pathways. One engineering strategy is to identify Fc mutations that facilitate antibody multimerization on the cell surface directly. From the analyses of the crystal packing of IgG1 structures, we identified a novel set of Fc mutations, T437R and K248E, that facilitated antibody multimerization upon binding to antigens on cell surface. In a NF-κB reporter assay, the engineered T437R/K248E mutations could facilitate enhanced agonism of an anti-OX40 antibody without the dependence on FcγRIIB crosslinking. Nonetheless, the presence of cells expressing FcγRIIB could facilitate a boost of the agonism of the engineered antibody with mutations on IgG1 Fc, but not on the silent IgG2σ Fc. The Fc engineered antibody also showed enhanced effector functions, including antibody-dependent cell-meditated cytotoxicity, antibody-dependent cellular phagocytosis, and complement-dependent cytotoxicity, depending on the IgG subtypes. Also, the engineered antibodies showed normal FcRn binding and pharmacokinetic profiles in mice. In summary, this study elucidated a novel Fc engineering approach to promote antibody multimerization on a cell surface, which could enhance agonism and improve effector function for anti-TNFR antibodies as well as other therapeutic antibodies.  相似文献

Adaptive laboratory evolution (ALE) was employed to isolate arsenate and copper cross-resistant strains, from the copper-resistant M. sedula CuR1. The evolved strains, M. sedula ARS50-1 and M. sedula ARS50-2, contained 12 and 13 additional mutations, respectively, relative to M. sedula CuR1. Bioleaching capacity of a defined consortium (consisting of a naturally occurring strain and a genetically engineered copper sensitive strain) was increased by introduction of M. sedula ARS50-2, with 5.31 and 26.29% more copper recovered from enargite at a pulp density (PD) of 1 and 3% (w/v), respectively. M. sedula ARS50-2 arose as the predominant species and modulated the proportions of the other two strains after it had been introduced. Collectively, the higher Cu2+ resistance trait of M. sedula ARS50-2 resulted in a modulated microbial community structure, and consolidating enargite bioleaching especially at elevated PD.  相似文献

The eradication of newly detected alien plant species is often prescribed, but rarely successful. Eradication programs fail for many reasons, however, for eradication to remain a cost-efficient management option it is clear that good decisions must be made at the outset. Here we re-frame the project prioritization protocol (PPP), a tool widely used in conservation biology, for use with the metrics typically used by a biosecurity agency. We then use existing methods to estimate the cost-efficiency of eradicating 50 hypothetical species incursions and compare the reduction in weed risk achieved by allocating resources using the PPP framework with the allocation based on risk ranking. By allocating resources to plant eradication programs using the PPP our analysis indicated that it is possible to improve the return on public expenditure by 25% compared to investing based solely on weed risk assessment scores. We also demonstrate how the cost-efficiency of the overall portfolio is influenced by the choice of planning horizon; including the decline in overall portfolio performance that arises when attempting to eradicate individual species too quickly. Finally, we discuss the logistical benefits to a management agency that arise from the use of a generic overarching framework such as the PPP. We believe that the PPP has considerable potential for use in biosecurity and can help focus attention on those species where management can make the biggest difference.  相似文献

Social interaction can have profound influences on the structure of the adult brain, but little is known about the precise stimulus feature found within social interaction that induces such brain plasticity. We examined the effects of social stimuli on cell addition and radial glial fiber formation in the brains of adult electric fish. These fish communicate primarily through weak, quasi-sinusoidal electric signals. Fish were housed in isolation, paired with another fish or exposed to only the electrocommunication signals of another fish for 7 days. After 3 days of exposure to these stimulus conditions, fish were injected with bromodeoxyuridine (BrdU) to mark newborn cells. We sacrificed the fish 4 days after BrdU injection and used immunohistochemistry to measure cell addition (BrdU+), the fraction of added cells that differentiated into neurons (BrdU+/NeuroTrace+) and the density of radial glia fibers (vimentin+) in the periventricular zone of the diencephalon. Fish that were exposed only to the electrocommunication signals of another fish and no other social stimuli had equivalent levels of cell addition and radial glial fiber density to fish that were housed with full social interaction and higher levels than fish housed in isolation. About 60% of the added cells differentiated into neurons; this fraction did not differ among treatment groups. Artificial sine wave electrical stimuli that mimicked electrocommunication signals were ineffective in increasing cell addition and glia fiber formation above those found in isolated fish. Thus, stimuli through a single modality are sufficient for inducing this brain plasticity, but the waveform or dynamic features of communication signals are crucial for the effect.  相似文献

Background

The developmental overnutrition hypothesis suggests that greater maternal obesity during pregnancy results in increased offspring adiposity in later life. If true, this would result in the obesity epidemic progressing across generations irrespective of environmental or genetic changes. It is therefore important to robustly test this hypothesis.

Methods and Findings

We explored this hypothesis by comparing the associations of maternal and paternal pre-pregnancy body mass index (BMI) with offspring dual energy X-ray absorptiometry (DXA)–determined fat mass measured at 9 to 11 y (4,091 parent–offspring trios) and by using maternal FTO genotype, controlling for offspring FTO genotype, as an instrument for maternal adiposity. Both maternal and paternal BMI were positively associated with offspring fat mass, but the maternal association effect size was larger than that in the paternal association in all models: mean difference in offspring sex- and age-standardised fat mass z-score per 1 standard deviation BMI 0.24 (95% confidence interval [CI]: 0.22 to 0.26) for maternal BMI versus 0.13 (95% CI: 0.11, 0.15) for paternal BMI; p-value for difference in effect < 0.001. The stronger maternal association was robust to sensitivity analyses assuming levels of non-paternity up to 20%. When maternal FTO, controlling for offspring FTO, was used as an instrument for the effect of maternal adiposity, the mean difference in offspring fat mass z-score per 1 standard deviation maternal BMI was −0.08 (95% CI: −0.56 to 0.41), with no strong statistical evidence that this differed from the observational ordinary least squares analyses (p = 0.17).

Conclusions

Neither our parental comparisons nor the use of FTO genotype as an instrumental variable, suggest that greater maternal BMI during offspring development has a marked effect on offspring fat mass at age 9–11 y. Developmental overnutrition related to greater maternal BMI is unlikely to have driven the recent obesity epidemic.  相似文献

Piscirickettsia salmonis is pathogenic for a variety of cultured marine fish species worldwide. The organism has been observed within host macrophages in natural disease outbreaks among coho salmon and European sea bass. In vitro studies, incorporating transmission electron microscopy (TEM) and ferritin loading of lysosomes, have confirmed that P. salmonis is capable of surviving and replicating in rainbow trout macrophages. Certain features of this intracellular survival underline its difference to other intracellular pathogens and suggest that a novel combination of defence mechanisms may be involved. Escape into the macrophage cytoplasm is not used as a means to avoid phago-lysosomal fusion and the organism remains at least partly enclosed within a vacuole membrane. While the piscirickettsial vacuole is often incomplete, survival and replication appear to require occupation of a complete, tightly-apposed, vacuolar membrane which does not fuse with lysosomes. Unlike some mammalian rickettsiae, actin-based motility (ABM) is not used as a means of intercellular spread. It is postulated that the presence of numerous small vesicles within vacuoles, and at gaps in the vacuolar membrane, may result from the blebbing of the piscirickettsial outer membrane seen early in the infection.  相似文献
