A study on the decomposition process of Pistia stratiotes L. was carried out in Cisne Reservoir, Uruguay. For this purpose, leaves and roots were considered separately, and the process was studied in the littoral and the pelagic zone. The litter bag technique was used to estimate dry weight losses at different times. Leaves decomposed faster than roots in both zones. Pistia decomposed faster in the pelagic zone due to better oxygen conditions and a more intensive water movement that removes material from within the bags. Phosphorus, potassium, sodium and magnesium were leached rapidly during the first week. Nitrogen content in the leaves initially increased, reaching its maximum in the littoral zone that coincided with that of bacterial numbers colonizing the litter. The number of macroinvertebrates, dominated by Chironomidae, was low. A rough estimation of the phosphorus impact on the littoral zone associated to the decomposition process shows that after 24 h between 13.9 and 16.8 μg-at.1−1 have been released to the water.  相似文献

Strategies reported for the separation of proteins in capillary zone electrophoresis and capillary isoelectric focusing are reviewed. The strategies are grouped into two categories: coated capillaries and buffer/sample additives. Success attained with each case and also, more importantly, the limitations of the methodology are discussed. Recent results from our own laboratory in the area of capillary isoelectric focusing in uncoated, fused silica capillaries using additives are summarized. The advantages and disadvantages of coated columns vs. additives are delineated.  相似文献

Increased dependence on blood glucose after acclimatization to 4,300 m   总被引:5,自引:0,他引:5  
To evaluate the hypothesis that altitude exposure and acclimatization result in increased dependency on blood glucose as a fuel, seven healthy males (23 +/- 2 yr, 72.2 +/- 1.6 kg, mean +/- SE) on a controlled diet were studied in the postabsorptive condition at sea level (SL), on acute altitude exposure to 4,300 m (AA), and after 3 wk of chronic altitude exposure to 4,300 m (CA). Subjects received a primed continuous infusion of [6,6-2D]glucose and rested for a minimum of 90 min, followed immediately by 45 min of exercise at 101 +/- 3 W, which elicited 51.1 +/- 1% of the SL maximal O2 consumption (VO2 max; 65 +/- 2% of altitude VO2 max). At SL, resting arterial glucose concentration was 82.4 +/- 3.2 mg/dl and rose significantly to 91.2 +/- 3.2 mg/dl during exercise. Resting glucose appearance rate (Ra) was 1.79 +/- 0.02 mg.kg-1.min-1; this increased significantly during exercise at SL to 3.71 +/- 0.08 mg.kg-1.min-1. On AA, resting arterial glucose concentration (85.8 +/- 4.1 mg/dl) was not different from sea level, but Ra (2.11 +/- 0.14 mg.kg-1.min-1) rose significantly. During exercise on AA, glucose concentration rose to levels seen at SL (91.4 +/- 3.0 mg/dl), but Ra increased more than at SL (to 4.85 +/- 0.15 mg.kg-1.min-1; P less than 0.05). Resting arterial glucose was significantly depressed with CA (70.8 +/- 3.8 mg/dl), but resting Ra increased to 3.59 +/- 0.08 mg.kg-1.min-1, significantly exceeding SL and AA values.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

It was the purpose of this investigation to examine any age-related changes in norepinephrine turnover (NEt) in four tissues at rest and during exercise. Fischer 344 rats 6 (n = 20) and 25 mo of age (n = 20) were received from the National Institute on Aging. NEt was determined at rest, during 30 min of submaximal exercise, and at maximal exercise by administration of alpha-methyl-p-tyrosine, a competitive inhibitor of tyrosine hydroxylase. Resting NE declined with age in both heart (38.2 vs. 30.5 ng.g-1.h-1) and liver (11.2 vs. 6.4 ng.g-1.h-1). NEt was greater in the older animals compared with the young animals in heart (120.9 vs. 169.5 ng.g-1.h-1), liver (23.1 vs. 38.9 ng.g-1.h-1), and adrenals (74.0 vs. 98.4 ng.mg-1.h-1) during submaximal exercise. In response to maximal exercise, NEt varied depending on age and tissue. It was concluded that, in response to exercise stress, the older animals generally demonstrated a higher NEt (reflecting elevated sympathetic activity) perhaps because of a decreased adrenergic receptor sensitivity and/or responsiveness.  相似文献

We report here the characterization of the first mammalian-like purine nucleoside phosphorylase from the hyperthermophilic archaeon Pyrococcus furiosus (PfPNP). The gene PF0853 encoding PfPNP was cloned and expressed in Escherichia coli and the recombinant protein was purified to homogeneity. PfPNP is a homohexamer of 180 kDa which shows a much higher similarity with 5'-deoxy-5'-methylthioadenosine phosphorylase (MTAP) than with purine nucleoside phosphorylase (PNP) family members. Like human PNP, PfPNP shows an absolute specificity for inosine and guanosine. PfPNP shares 50% identity with MTAP from P. furiosus (PfMTAP). The alignment of the protein sequences of PfPNP and PfMTAP indicates that only four residue changes are able to switch the specificity of PfPNP from a 6-oxo to a 6-amino purine nucleoside phosphorylase still maintaining the same overall active site organization. PfPNP is highly thermophilic with an optimum temperature of 120 degrees C and is characterized by extreme thermodynamic stability (T(m), 110 degrees C that increases to 120 degrees C in the presence of 100 mm phosphate), kinetic stability (100% residual activity after 4 h incubation at 100 degrees C), and remarkable SDS-resistance. Limited proteolysis indicated that the only proteolytic cleavage site is localized in the C-terminal region and that the C-terminal peptide is not necessary for the integrity of the active site. By integrating biochemical methodologies with mass spectrometry we assigned three pairs of intrasubunit disulfide bridges that play a role in the stability of the enzyme against thermal inactivation. The characterization of the thermal properties of the C254S/C256S mutant suggests that the CXC motif in the C-terminal region may also account for the extreme enzyme thermostability.  相似文献

The talin rod contains approximately 11 vinculin binding sites (VBSs), each defined by hydrophobic residues in a series of amphipathic helices that are normally buried within the helical bundles that make up the rod. Consistent with this, talin failed to compete for binding of the vinculin Vd1 domain to an immobilized talin polypeptide containing a constitutively active VBS. However, talin did bind to GST-Vd1 in pull-down assays, and isothermal titration calorimetry measurements indicate a K(d) of approximately 9 mum. Interestingly, Vd1 binding exposed a trypsin cleavage site in the talin rod between residues 898 and 899, indicating that there are one or more active VBSs in the N-terminal part of the talin rod. This region comprises a five helix bundle (residues 482-655) followed by a seven-helix bundle (656-889) and contains five VBSs (helices 4, 6, 9, 11, and 12). The single VBS within 482-655 is cryptic at room temperature. In contrast, talin 482-889 binds Vd1 with high affinity (K(d) approximately 0.14 mum), indicating that one or more of the four VBSs within 656-889 are active, and this likely represents the vinculin binding region in intact talin. In support of this, hemagglutinin-tagged talin 482-889 localized efficiently to focal adhesions, whereas 482-655 did not. Differential scanning calorimetry showed a strong negative correlation between Vd1 binding and helical bundle stability, and a 755-889 mutant with a more stable fold bound Vd1 much less well than wild type. We conclude that the stability of the helical bundles that make up the talin rod is an important factor determining the activity of the individual VBSs.  相似文献

Fish play a key role in the trophic dynamics of lakes, not least in shallow systems. With climate warming, complex changes in fish community structure may be expected owing to the direct and indirect effects of temperature, and indirect effects of eutrophication, water-level changes and salinisation on fish metabolism, biotic interactions and geographical distribution. We review published and new data supporting the hypotheses that, with a warming climate, there will be changes in: fish community structure (e.g. higher or lower richness depending on local conditions); life history traits (e.g. smaller body size, shorter life span, earlier and less synchronised reproduction); feeding mode (i.e. increased omnivory and herbivory); behaviour (i.e. stronger association with littoral areas and a greater proportion of benthivores); and winter survival. All these changes imply higher predation on zooplankton and macroinvertebrates with increasing temperatures, suggesting that the changes in the fish communities partly resemble, and may intensify, the effects triggered by eutrophication. Modulating factors identified in cold and temperate systems, such as the presence of submerged plants and winter ice cover, seem to be weaker or non-existent in warm(ing) lakes. Consequently, in the future lower nutrient thresholds may be needed to obtain clear-water conditions and good ecological status in the future in currently cold or temperate lakes. Although examples are still scarce and more research is needed, we foresee biomanipulation to be a less successful restoration tool in warm(ing) lakes without a strong reduction of the nutrient load.  相似文献

Physiol.84(1): 177-184, 1998.Normal aging without disease leads todiminished chronotropic and inotropic responses to catecholaminestimulation, resulting in depressed cardiac function with stress. Thepurpose of this study was to determine molecular mechanisms fordecrements in adrenergic responsiveness of the left ventricle (LV) dueto aging and to study the effects of chronic dynamic exercise on signaltransduction. We measured -adrenergic receptor (-AR) density,adenylyl cyclase (AC) activity, and G-protein content and distributionin LV from 66 male Fischer 344 rats from three age groups that wereeither sedentary or treadmill trained (60 min/day, 5 days/wk, 10 wk at75% of the maximal capacity). Final ages were 7 mo(young), 15 mo (middle-age), and 25 mo (old). There was no significantdifference in -AR density among groups as a function of age ortraining. AC production of adenosine 3,5-cyclic monophosphate (cAMP)with the use of five pharmacological stimulations revealed that oldsedentary myocardium had depressed basal, receptor-dependent, G-protein-dependent, and AC catalyst stimulation (30-43%)compared with hearts from young and middle-age sedentary rats. Training did not alter AC activity in either middle-age or old groups but didincrease G-protein-dependent cAMP production in young myocardium (12-34%). Immunodetectable concentrations of stimulatory andinhibitory G proteins (Gs and Gi, respectively)showed 43% less total Gs with similar Gicontent in hearts from old sedentary compared with middle-age sedentaryrats. When compared with young sedentary animals, Gicontent was 39 and 50% higher in middle-age sedentary and oldsedentary myocardium, respectively. With age, there was a significantshift in the -subunit of Gs distribution from cytosolic fractions of LV homogenates to membrane-bound fractions (8-12% redistribution in middle-age sedentary vs. old sedentary). The mostsignificant training effect was a decrease in Gi content inhearts from old trained rats (23%), which resulted in values comparable with young sedentary rats and reduced theGi/Gs ratio by 27% in old-rat LV. We reportthat age-associated reductions in cardiovascular -adrenergicresponsiveness correspond with alterations in postreceptor adrenergicsignaling rather than with a decrease in receptor number. Chronicdynamic exercise partially attenuates these reductions throughalterations in postreceptor elements of cardiac signal transduction.

After short-term exposure to high altitude (HA), men appear to be less sensitive to insulin than at sea level (SL). We hypothesized that the same would be true in women, that reduced insulin sensitivity would be directly related to the rise in plasma epinephrine concentrations at altitude, and that the addition of alpha-adrenergic blockade would potentiate the reduction. To test the hypotheses, 12 women consumed a high-carbohydrate meal at SL and after 16 h at simulated 4,300-m elevation (HA). Subjects were studied twice at each elevation: once with prazosin (Prz), an alpha(1)-adrenergic antagonist, and once with placebo (Pla). Mathematical models were used to assess insulin resistance based on fasting [homeostasis model assessment of insulin resistance (HOMA-IR)] and postprandial [composite model insulin sensitivity index (C-ISI)] glucose and insulin concentrations. Relative to SL-Pla (HOMA-IR: 1.86 +/- 0.35), insulin resistance was greater in HA-Pla (3.00 +/- 0.45; P < 0.05), SL-Prz (3.46 +/- 0.51; P < 0.01), and HA-Prz (2.82 +/- 0.43; P < 0.05). Insulin sensitivity was reduced in HA-Pla (C-ISI: 4.41 +/- 1.03; P < 0.01), SL-Prz (5.73 +/- 1.01; P < 0.05), and HA-Prz (4.18 +/- 0.99; P < 0.01) relative to SL-Pla (8.02 +/- 0.92). Plasma epinephrine was significantly elevated in HA-Pla (0.57 +/- 0.08 ng/ml; P < 0.01), SL-Prz (0.42 +/- 0.07; P < 0.05), and HA-Prz (0.82 +/- 0.07; P < 0.01) relative to SL-Pla (0.28 +/- 0.04), but correlations with HOMA-IR, HOMA-beta-cell function, and C-ISI were weak. In women, short-term exposure to simulated HA reduced insulin sensitivity compared with SL. The change does not appear to be directly mediated by a concurrent rise in plasma epinephrine concentrations.  相似文献
