Abstract: We analyzed the expression and relative distribution of mRNA for the regulatory subunits (RIα, RIIα, and RIIβ) and of 150-kDa RIIβ-anchor proteins for cyclic AMP (cAMP)-dependent protein kinase (PKA) into discrete brain regions. The subcellular distribution of both holoenzyme and free catalytic subunit was evaluated in the same CNS areas. In the neocortex and corpus striatum high levels of RIIβ paralleled the presence of specific RII-anchoring proteins, high levels of membrane-bound PKA holoenzyme, and low levels of cytosolic free catalytic activity (C-PKA). Conversely, in brain areas showing low RIIβ levels (cerebellum, hypothalamus, and brainstem) we found an absence of RII-anchoring proteins, low levels of membrane-bound holoenzyme PKA, and high levels of cytosolic dissociated C-PKA. Response to cAMP stimuli was specifically evaluated in the neocortex and cerebellum, prototypic areas of the two different patterns of PKA distribution. We found that cerebellar holoenzyme PKA was highly sensitive to cAMP-induced dissociation, without, however, a consistent translocation of C-PKA into the nucleus. In contrast, in the neocortex holoenzyme PKA was mainly in the undissociated state and poorly sensitive to cAMP. In nuclei of cortical cells cAMP stimulated the import of C-PKA and phosphorylation of cAMP-responsive element binding protein. Taken together, these data suggest that RIIβ (whose distribution is graded throughout the CNS, reaching maximal expression in the neocortex) may represent the molecular cue of the differential nuclear response to cAMP in different brain areas, by controlling cAMP-induced holoenzyme PKA dissociation and nuclear accumulation of catalytic subunits. 相似文献
Tumor necrosis factor alpha (TNF-alpha) is one of the best-described cell death promoters. In murine L929 fibroblasts, dexamethasone inhibits TNF-alpha-induced cytotoxicity. Since phosphatidyl inositol 3 kinase (PI3K) and nuclear factor kappa B (NF-kappaB) proteins regulate several survival pathways, we evaluated their participation in dexamethasone protection against TNF-alpha cell death. We interfered with these pathways by overexpressing a negative dominant mutant of PI3K or a non-degradable mutant of inhibitor of NF-kappaB alpha (IkappaBalpha) (the cytoplasmic inhibitor of NF-kappaB) in L929 cells. The mutant IkappaB, but not the mutant PI3K, abrogated dexamethasone-mediated protection. The loss of dexamethasone protection was associated with a diminished accumulation in XIAP and c-IAP proteins. 相似文献
When Cellulomonas flavigena CDBB-531 was grown on glucose, xylose, glycerol, solka floc, sugarcane bagasse or xylan, xylanase activity was found only in the fermentation broth, while -xylosidase activity was always associated with the cells. Both enzymes were inducible, sugar-cane bagasse was the best inducer, solka floc and avicel were moderately good, while xylan was poor. A synergistic effect on xylanase and -xylosidase synthesis was observed when cellulose and hemicellulose were used together as carbon sources. When this strain was grown on glucose, cellobiose, arabinose or xylose, only low levels of both enzymes were detected. These results indicate that xylanase and -xylosidase were carbon-source-repressed by readily metabolizable substrates. The effect of glycerol on enzymes that were already induced was studied. The addition of glycerol caused a significant decrease in the levels of xylanases, while -xylosidase activity remained unchanged. 相似文献
A survey of nodulation was carried out in seven regions of Brazil and the nodulation status of 131 legume species is reported. Nodulation is reported for the first time in 46 species and six genera, representing 35% of the examined species. Two of the genera were non-nodulating: Martiodendron and Nissolia, and four nodulating: Riedeliella, Bergeronia, Goldmania. Moreover the last tribe of Mimosoideae yet to be examined for nodulation Mimoziganthus, was shown to be nodulated. The nodulation pattern is discussed in the light of the taxonomy of the family. 相似文献
Abstract: D1-and D2-dopamine receptors exert important physiological actions on striatal neurons, but the intracellular second messenger pathways activated by these receptors are still incompletely understood. Using primary cultures of rat striatal cells, we have examined the effects of activating D1 or D2 receptors on arachidonic acid (AA) release and cyclic AMP accumulation. In striatal neurons labeled by incubation with [3H]AA, D2-receptor stimulation enhanced release of [3H]AA produced by application of the Ca2+ ionophore A23187 or of the purinergic agonist ATP. By contrast, D1-receptor stimulation inhibited [3H]AA release. This inhibitory effect of D1 receptors was accompanied by stimulation of adenylyl cyclase activity, measured as accumulation of cyclic AMP, and was mimicked by application of the adenylyl cyclase activator forskolin. The results indicate the existence of a novel signaling pathway for D2 and D1 receptors in striatum, potentiation and inhibition, respectively, of Ca2+-evoked AA release. 相似文献

We hypothesized that near-future elevated CO2 would affect the antipredatory behavior of two freshwater organisms; a pulmonate gastropod (Physella columbiana) and a cladoceran crustacean (Daphnia magna). Studies have found that pCO2 and increased acidification due to CO2 impedes fright responses to predators by activating GABAA receptors. After administration of predator-derived kairomones and conspecific alarm cues, we also briefly exposed some of the animals to gabazine which is a GABAA receptor antagonist to restore a fright response. We found that added carbon dioxide negatively affected the antipredatory behavior of both species but gabazine did not reverse this effect. To further examine the effect of CO2 and gabazine, we also tested the effect of stressful crowding, cold, and acidic conditions on the production of male daphnid offspring. An increase in ratio of male to female offspring is a common and expected response to stress by daphnids. We found that stress increased the production of males and gabazine reversed this at a pH of 5.5 but not at pH 6.2 or 6.5. Our study suggest that while the main negative effects of anthropogenic CO2 enrichment can be robust, the myriad indirect effects of CO2 make predictions about future predator prey systems less clear.
Propionibacterium acidipropionici was grown in a fed-batch culture, fed with glucose or lactate, or mixtures of lactate and glucose. Lactate and glucose were always simultaneously consumed. As co-substrate, glucose modified the propionate:acetate molar ratio (P/A) and increased the fraction of carbon used for biomass production. A P/A of 7.63 was obtained with a lactate:glucose molar ratio of 4; a P/A value of 1.34 was obtained with lactate alone and 1.85 with glucose alone. The fraction of carbon recovered in biomass was 0.09 for glucose, 0.12 for lactate, and 0.21 for a lactate:glucose molar ratio of 4. 相似文献
Submerged conidia and blastospores of the entomopathogenic fungus Isaria fumosorosea are produced in several liquid culture media. However, yields and the ecological fitness of these propagules vary according to culture media composition. In most culture media, hyphae, blastospores and submerged conidia are white but we found that in some media they develop a brown pigmentation. A dark pigment was extracted from brown-pigmented propagules and analyzed by IR spectroscopy. Adsorption bands coincided to those characteristics of melanins.Hadamard's matrices were employed in order to increase submerged conidia yields and brown pigmentation of fungal propagules. Media containing 20–30 mg/l of FeSO4·7H2O and 6–12 mg/l of CuSO4·5H2O allowed reaching the highest pigmentation (9 in a hedonic scale). A maximal concentration of submerged conidia of 1.0 (±1.2) × 1012 cell/l was achieved after 120 h of liquid culture in a improved culture medium, containing 25 ml/l of Polyethylene glycol (MW 200), substance which enhanced submerged conidia production, reducing free mycelia or mycelial pellets formation. In the improved medium, it was estimated that more than 60% of produced biomass corresponded to submerged conidia and blastospores, while in other media, mycelia were the main product (80–97%). 相似文献
