Queens in primitively eusocial insect societies are morphologically indistinguishable from their workers, and occupy the highest position in the dominance hierarchy. Such queens are believed to use aggression to maintain worker activity and reproductive monopoly in the colony. However, in the primitively eusocial wasp Ropalidia marginata, the queen is a strikingly docile individual, who interacts rarely with her workers. If the queen is experimentally removed, one of the workers becomes extremely aggressive within minutes, and eventually becomes the new queen of the colony. We designate her as the potential queen. Experimental evidence suggests that the queen probably uses a non-volatile pheromone to signal her presence to her workers. Here we attempt to identify the mechanism by which the queen transmits information about her presence to the workers. We designate the time taken for the potential queen to realize the absence of the queen as the realization time and model the realization time as a function of the decay time of the queen's signal and the average signal age. We find that the realization time obtained from the model, considering only direct interactions (193.5 min) is too large compared to the experimentally observed value of 30 min. Hence we consider the possibility of signal transfer through relay. Using the Dijkstra's algorithm, we first establish the effectiveness of relay in such a system and then use experimental data to fit the model. We find that the realization time obtained from the model, considering relay (237.1 min) is also too large compared to the experimentally observed value of 30 min. We thus conclude that physical interactions, both direct and indirect (relay), are not sufficient to transfer the queen's signal in R. marginata. Finally, we discuss the possibility that the queen applies her pheromone on the nest material from where the workers can perceive it without having to physically interact with the queen.  相似文献

Ethanol induced liver injury is associated with a global change in gene expression but its mechanisms are not known. We studied whether alcohol-induced gene expression is associated with post-translational methylations of histone H3. Primary culture of rat hepatocytes was treated with ethanol (50 or 100 mM) for 24 h and the status of methylation of H3 at lys 4 (H3dimeK4) or lys 9 (H3dimeK9) was monitored by Western blotting using antibodies to dimethylated histone H3 at lys 4 or lys 9. The cells exposed to ethanol showed strikingly opposing behaviors in methylation patterns; H3dimeK9 methylation was decreased whereas H3dimeK4 increased. Similar results were obtained in the interphase nuclei. Their binding on the metaphase chromosomes exhibits distinct site specific pattern of accumulation. Next, chromatin immunoprecipitation of the ethanol treated samples with antibodies for methylated lys 4 or lys 9 histone H3 followed by amplification of the immunoprecipitated DNA, was used to determine their association with the promoters of genes up- or downregulated by ethanol. Lys4 methylation was associated with ethanol upregulated genes (Adh, GST-yc2) whereas lys 9 methylation with downregulated genes (Lsdh, cytP4502c11) demonstrating a difference between these two methylations. These results suggest that exposure of hepatocytes to ethanol changes the expression of several susceptible genes which are associated with site specific modification of dimethylated forms of histone H3 amino termini at their regulatory regions.  相似文献

Human type 5 17beta-hydroxysteroid dehydrogenase plays a crucial role in local androgen formation in prostate tissue. Several chemicals were synthesized and tested for their ability to inhibit this enzyme, and a series of estradiol derivatives bearing a lactone on the D-ring were found to inhibit its activity efficiently. The crystal structure of the type 5 enzyme in complex with NADP and such a novel inhibitor, EM1404, was determined to a resolution of 1.30 A. Significantly more hydrogen bonding and hydrophobic interactions were defined between EM1404 and the enzyme than in the substrate ternary complex. The lactone ring of EM1404 accounts for important interactions with the enzyme, whereas the amide group at the opposite end of the inhibitor contributes to the stability of three protein loops involved in the construction of the substrate binding site. EM1404 has a strong competitive inhibition, with a Ki of 6.9+/-1.4 nM, demonstrating 40 times higher affinity than that of the best inhibitor previously reported. This is observed despite the fact that the inhibitor occupies only part of the binding cavity. Attempts to soak the inhibitor into crystals of the binary complex with NADP were unsuccessful, yielding a structure with a polyethylene glycol fragment occupying the substrate binding site. The relative crystal packing is discussed. Combined studies of small molecule inhibitor synthesis, x-ray crystallography, enzyme inhibition, and molecular modeling make it possible to analyze the plasticity of the substrate binding site of the enzyme, which is essential for developing more potent and specific inhibitors for hormone-dependent cancer therapy.  相似文献

Escherichia coli serotype O157:H7 was detected among bacteria collected from the Ganges River. O157:H7 isolates tested positive for stx(1), stx(2), and eae gene sequences. Identification of potentially pathogenic isolates from extensively used source water indicates that O157:H7 may be a significant but as yet underacknowledged public health concern in India.  相似文献

A total of 35 yeasts were isolated from the gut of beetles collected from Hyderabad city, India. Twenty of these yeasts utilized xylose as a sole carbon source but only 12 of these converted xylose to xylitol. The ability to convert xylose to xylitol varied among the isolates and ranged from 0.12 to 0.58 g/g xylose. Based on the phenotypic characteristics and phylogenetic analysis of the D1/D2 domain sequence of 26S rRNA gene, these isolates were identified as members of Pichia, Candida, Issatchenkia, and Clavispora. Strain YS 54 (CBS 10446), which was phylogenetically similar to Pichia caribbica and which formed hat-shaped ascospore characteristics of the genus Pichia, was the best xylitol producer (0.58 g xylitol/g xylose). YS 54 was also capable of producing xylitol from sugarcane bagasse hydrolysate and the efficiency of conversion was 0.32 g xylitol/g xylose after 20 cycles of adaptation in medium containing sugarcane bagasse hydrolysate.  相似文献

A galactose specific lectin was isolated from the seeds of Ficus bengalensis (Moraceae) fruits and designated as F. bengalensis agglutinin (FBA). The lectin was purified by affinity repulsion chromatography on fetuin-agarose and was a monomer of molecular mass 33kDa. Like other Moraceae family lectins, carbohydrate-binding activity of FBA was independent of any divalent cation. FBA did not bind with simple saccharides, however sugar ligands with aromatic aglycons showed pronounced binding. The combining site of FBA recognized preferably Galbeta1,4GlcNAcbeta1-(II) followed by Galbeta1,3GalNAcalpha1-(Talpha) containing glycotopes. Interaction with saccharides revealed that the combining site of FBA could well accommodate a tetrasaccharide, asialo GM1 glycan (Galbeta1,3GalNAcbeta1,4Galbeta1,4Glcbeta1-), whereas polyvalent Tn (GalNAcalpha1-Ser/Thr), one of the well-recognized ligands of Moraceae family lectin, did not interact well with FBA.  相似文献

Parental care is an essential component in the life history of mammals. In group-living species, care can be provided by adults other than the parents, and such care is termed alloparental care. Alloparental care is known in a wide spectrum of species, from insects to humans. Most canids that live in stable packs demonstrate cooperative breeding, where subordinates provide care to the offspring of the dominants, without reproducing themselves. Free-ranging dogs in India have a dynamic social system and, unlike their cooperatively breeding ancestors, the grey wolves, all adults in a dog group have equal mating opportunities. This at times leads to the birth of multiple litters within an existing dog group. In this paper, we report the first field observations of alloparental care made on a dog group where a bitch provided care to her grandpups, through interactions other than suckling. The allomaternal care acted as a supplement to the care provided by the mother, and was thus beneficial to the pups.  相似文献

Canids display a wide diversity of social systems, from solitary to pairs to packs, and hence, they have been extensively used as model systems to understand social dynamics in natural habitats. Among canids, the dog can show various levels of social organization due to the influence of humans on their lives. Though the dog is known as man’s best friend and has been studied extensively as a pet, studies on the natural history, ecology and behavior of dogs in a natural habitat are rare. Here, we report results of an extensive population-level study conducted through one-time censuses in urban India to understand the foraging associations of free-ranging dogs. We built a model to test if the observed groups could have been formed through random associations while foraging. Our modeling results suggest that the dogs, like all efficient scavengers, tend to forage singly but also form random uncorrelated groups. A closer inspection of the group compositions, however, reveals that the foraging associations are non-random events. The tendency of adults to associate with the opposite sex in the mating season and of juveniles to stay close to adults in the non-mating season drives the population towards specific aggregation. Hence we conclude that to be or not to be social is a matter of choice for the free-ranging dogs, and not a matter of chance.  相似文献

To contain transmission of COVID‐19, lockdowns or strict restrictions of people''s mobility outside their residences were instituted in a majority of countries worldwide, including Nepal, where the first phase of nationwide lockdown was observed from 24 March to 21 July 2020. This sudden halt in human outdoor activities brought positive and negative impacts on forests and wildlife. We undertook a study to learn the impact of the COVID‐19 lockdown on wildlife and forests in the protected areas (PAs) of Nepal. Between July and September 2020, data on illegal activities recorded by the staff of PAs and also those reported by media were collected and analyzed. Key informant interviews (KII) were done with the park officers and security personnel by virtual communication (telephone, messenger app, and video call) to collect detailed information and for corroboration. The collected data were categorized into four groups: (a) wildlife killed, (b) wildlife injured, (c) arrest incidents related to forest crime, and (d) arrest incidents related to wildlife crime. Data from the fiscal year 2019–2020 were analyzed, comparing before lockdown and after. Among 20 PAs investigated during the lockdown, the study found substantial increases in wildlife death in two PAs, Banke National Park, and Bardia National Park. Similarly, Chitwan National Park (CNP) and Shivapuri Nagarjun National Park (SNNP) witnessed a rise in wildlife poaching. CNP and SNNP are located close to densely populated cities and also have human settlements in their peripheries. Wildlife was sighted freely roaming inside PAs during the lockdown, presumably because the absence of visitors and human activities during the lockdown decreased disturbance. Thus, the wildlife was enjoying the freedom of movement on the one hand, and on the other hand was threatened by poachers, many of whom were laid off from other activities and were taking advantage of the lapse in security.  相似文献
