Abstract

The affinity of L-DNAs, L-RNAs and L/D-DNAs for homopurine homopyrimidine d.s. D-DNA and s.s. D-RNA was probed by gel electrophoresis and CD spectroscopy. It was found that the L-modified oligomers do not bind to d.s. DNA and to natural RNA that contains all four natural bases. Thus they cannot be used, in general, for the control of gene expression according to the antigene and antisense methodologies. Heterochiral complexes with 1:1 stoichiometry and low thermal stability are formed, instead, by homopurinic L-RNA or L/D-DNA and homopyrimidinic L-RNA with the W/C complementary natural RNA sequences.  相似文献

Trace elements in soils exist as components of several different fractions. We have analyzed the correlation between total and extractable (EDTA, calcium chloride and deionized water) Zn, Pb and Cu concentrations in soils and the concentration of these elements in plant leaves. Soil and plant samples have been taken from Sulcis-Iglesiente (Sardinia), an area rich in mining tailings. This has made that the concentrations of the trace element under study in soils were varied. Three plants have been studied: Dittrichia viscosa, Cistus salviifolius, and Euphorbia pithyusa subsp. cupanii. Soil samples beneath each of them at depths of 0–30 and 30–60 cm have been considered. The highest concentration of trace elements in the leaves of the studied species has been found for Zn. The calcium carbonate content and the crystalline and amorphous forms of iron in the soil have determined the concentration of metal in plant leaves. The soil concentrations that have been found with the extraction methods are uncorrelated with Pb and Cu concentrations in plants, but Zn is correlated with the fraction extracted with EDTA and calcium chloride. The concentrations of trace metals in plants are most closely related to the soil contents of CaCO3, electrical conductivity, Feox, and Fedc.  相似文献

ABSTRACT

A survey was conducted to identify plants emitting isoprenoids in the Beijing area which could potentially contribute to smog episodes when combined with anthropogenic pollutants. The emission pattern was similar to that observed in the previously surveyed boreal ecosystems (Europe, North America). Most deciduous oaks are strong isoprene emitters; however, some of them do not emit isoprenoids and are therefore more suitable for the urban environment of Beijing. No emission of monoterpenes was found in Chinese oaks, and this trait seems therefore confined to the Mediterranean environment. The emission of isoprene was found in poplars and in some of the bamboo widespread in city parks and in the riparial vegetation surrounding the city. Chinese pine species emit monoterpenes when wounded, and the emission is not qualitatively different among species. Pinus tabulaeformis, one of the most important trees in China, is a low emitter compared to other pine species.  相似文献

Abstract

Over the past 150 years the amount of CO2 in the atmosphere has been increasing, largely as a result of land-use change and anthropogenic emissions from the burning of fossil fuels. It is estimated that the atmospheric [CO2] will reach 70 Pa by the end of the 21st Century. The most important consequence of this rise in [CO2] is warming the surface temperature of the Earth by 0.4 – 0.6°C per decade throughout the 21st Century. Increasing [CO2] along with associated changes in temperature will most likely alter the structure and function of agro-ecosystems, affecting their productivity and their role as stable sinks to CO2 sequestration. Both CO2 and temperature are key variables affecting plant growth, development and functions. Moreover, because of the future scenario of higher temperature and evaporative demand, drought occurrences will be increased in frequency, intensity, and erratic pattern. The combination of elevated temperatures and the increased incidence of environmental stress will probably constitute the greatest risk caused by climate change to the agro-ecosystems in arid or semiarid areas of the world. The purpose of this paper is to review the exchange of carbon driving the main ecophysiological processes of plants in response to climate change and environmental stresses. Drought and salinity first affect the acquisition of CO2 by increasing stomatal and mesophyll resistances, and only after cause irreversible damages to the biochemical apparatus. Heat stress denatures thylakoid membranes, but this action may be counteracted by the synthesis of many isoprenoids in the chloroplasts from carbon freshly fixed by photosynthesis. There is rising concern about the impact of environmental stress on tree growth with this future scenario of global climate change. The combination of elevated temperatures and the increased incidence of environmental stress (particularly drought and salinity) will probably constitute the greatest risk caused by global climate change to the forest ecosystems in arid or semiarid areas of the world.  相似文献

Cytochrome P450 1A1 CYP1A1 enzymatic activity was evaluated in cultured liver cells, and taken as a biological indicator of the presence of inducers of this isoform in urban airborne particulate matter fraction samples. It is known that CYP1A1 inducers can play an important role in the risk of mutagenesis and carcinogenesis by environmental pollution. A romatic polycyclic hydrocarbons PAH from urban air were collected in the city of Genoa Italy at two sites on two different days of the year. The objective of the study was to compare the inducibility of cultured rat hepatocytes with that of MH1C1 and FaO rat hepatoma cell lines after exposure to a PAH mixture and to a standard compound, such as benzo b fluoranthene B b F. Cytotoxic effects of the tested concentrations were evaluated by means of 3 4,5, dimenthylthyazol 2 yl 2,5 diphenyltetrazolium bromide MTT and lactate dehydrogenase release LDH tests, the potency of inducers by ethoxyresorufin O deethylase EROD assay. The results were in agreement in the three cellular systems: after exposure to the PAH mixture, an induction at low concentrations was observed; whereas no induction, but rather a decrease in activity was shown at higher concentrations; instead, the exposure to pure B b F showed a dose-response relationship in all cells, even at the highest doses. Such a difference between the toxicity of the complex mixture and that of the pure compound could be ascribed to the presence of drug metabolism inhibitors in the mixture, or to interactions between the original components and their metabolites. The finding that the cell lines responded to the CYP1A1 induction in a very efficient way gives further proof of the applicability of this system to environmental biomonitoring.  相似文献

ObjectiveInvestigate the influence of apprehensive gait on activation and cocontraction of lower limb muscles of younger and older female adults.MethodsData of 17 younger (21.47 ± 2.06 yr) and 18 older women (65.33 ± 3.14 yr) were considered for this study. Participants walked on the treadmill at two different conditions: normal gait and apprehensive gait. The surface electromyographic signals (EMG) were recorded during both conditions on: rectus femoris (RF), vastus lateralis (VL), vastus medialis (VM), biceps femoris (BF), tibialis anterior (TA), gastrocnemius lateralis (GL), and soleus (SO).ResultsApprehensive gait promoted greater activation of thigh muscles than normal gait (F = 5.34 and p = 0.007, for significant main effect of condition; RF, p = 0.002; VM, p < 0.001; VL, p = 0.003; and BF, p = 0.001). Older adults had greater cocontraction of knee and ankle stabilizer muscles than younger women (F = 4.05 and p = 0.019, for significant main effect of groups; VM/BF, p = 0.010; TA/GL, p = 0.007; and TA/SO, p = 0.002).ConclusionApprehensive gait promoted greater activation of thigh muscles and older adults had greater cocontraction of knee and ankle stabilizer muscles. Thus, apprehensive gait may leads to increased percentage of neuromuscular capacity, which is associated with greater cocontraction and contribute to the onset of fatigue and increased risk of falling in older people.  相似文献

Marginal zone (MZ) B cells, identified as surface (s)IgMhighsIgDlowCD23low/−CD21+CD38 B cells, were purified from human spleens, and the features of their V(D)J gene rearrangements were investigated and compared with those of germinal center (GC), follicular mantle (FM) and switched memory (SM) B cells. Most MZ B cells were CD27+ and exhibited somatic hypermutations (SHM), although to a lower extent than SM B cells. Moreover, among MZ B-cell rearrangements, recurrent sequences were observed, some of which displayed intraclonal diversification. The same diversifying sequences were detected in very low numbers in GC and FM B cells and only when a highly sensitive, gene-specific polymerase chain reaction was used. This result indicates that MZ B cells could expand and diversify in situ and also suggested the presence of a number of activation-induced cytidine deaminase (AID)-expressing B cells in the MZ. The notion of antigen-driven expansion/selection in situ is further supported by the VH CDR3 features of MZ B cells with highly conserved amino acids at specific positions and by the finding of shared (“stereotyped”) sequences in two different spleens. Collectively, the data are consistent with the notion that MZ B cells are a special subset selected by in situ antigenic stimuli.  相似文献

Background aimsThe umbilical cord (UC) is a promising source of mesenchymal stromal cells (MSCs). UC-MSCs display very similar in vitro characteristics to bone marrow–MSCs and could represent a valuable alternative for cell-based therapies. However, it is still unclear whether UC-MSCs are prone or not to the acquisition of genomic imbalances during in vitro expansion.MethodsWith the use of array-comparative genomic hybridization, we compared copy number variations of early (P2–P3) and late (>P5) passages of in vitro–expanded UC-MSCs.ResultsIn two of 11 long-term UC-MSCs cultures, we observed the appearance of clones carrying genomic imbalances, which generated genetic mosaicism at intermediate passages. Although still able to reach the senescence phase, the cells carrying the genomic imbalance acquired a proliferative advantage, as demonstrated by the increase in frequency during long-term culture.ConclusionsAltogether, our results suggest that UC-MSC–based clinical protocols should be designed with caution; their clinical use should be preceded by array-comparative genomic hybridization screening for the acquisition of genomic imbalances during in vitro expansion.  相似文献

Uric acid inhibited 50% of the activity of bovine kidney low molecular mass phosphotyrosine protein phosphatase at concentrations of 1.0, 0.4, 1.3, and 0.2 mM, respectively for p -nitrophenyl phosphate (p -NPP), flavine mononucleotide, β -naphthyl phosphate and tyrosine phosphate (Tyr-P) as substrates. The mixed type inhibition of p -NPP hydrolysis was fully reversible, with K ic and K iu values of 0.4 and 1.1 mM, respectively; the inhibition by uric acid shifted the pH optimum from 5.0 to 6.5. When Tyr-P was the substrate, competitive inhibition was observed with a K i value of 0.05 mM. Inhibition studies by uric acid in the presence of thiol compounds, and preincubation studies in the presence of inorganic phosphate suggest that the interaction of uric acid with the enzyme occurred at the active site, but did not involve SH residues, and that the mechanism of inhibition depended on the structure of the substrates.  相似文献
