The important role of pyruvate kinase during malarial infection has prompted the cloning of a cDNA encoding Plasmodium falciparum pyruvate kinase (pfPyrK), using mRNA from intraerythrocytic-stage malaria parasites. The full-length cDNA encodes a protein with a computed molecular weight of 55.6 kDa and an isoelectric point of 7.5. The purified recombinant pfPyrK is enzymatically active and exists as a homotetramer in its active form. The enzyme exhibits hyperbolic kinetics with respect to phosphoenolpyruvate and ADP, with Km of 0.19 and 0.12 mM, respectively. pfPyrK is not affected by fructose-1,6-bisphosphate, a general activating factor of pyruvate kinase for most species. Glucose-6-phosphate, an activator of the Toxoplasma gondii enzyme, does not affect pfPyrK activity. Similar to rabbit pyruvate kinase, pfPyrK is susceptible to inactivation by 1 mM pyridoxal-5′-phosphate, but to a lesser extent. A screen for inhibitors to pfPyrK revealed that it is markedly inhibited by ATP and citrate. Detailed kinetic analysis revealed a transition from hyperbolic to sigmoidal kinetics for PEP in the presence of citrate, as well as competitive inhibitory behavior for ATP with respect to PEP. Citrate exhibits non-competitive inhibition with respect to ADP with a Ki of 0.8 mM. In conclusion, P. falciparum expresses an active pyruvate kinase during the intraerythrocytic-stage of its developmental cycle that may play important metabolic roles during infection.  相似文献

The mean basal specific activities and the mean % deficiencies of the activity of the glutamic oxaloacetic transaminase of the erythrocytes were identical (n.s.) for a group of eight patients with a severe carpal tunnel syndrome and for a group of eight university students. There was no significant difference in the increases in the specific activities for the patients and the students at 4 concentrations of pyridoxal 5′-phosphate. The apparent Km for the patients and the students was 95 μM and 61 μM (n.s.) respectively. It is concluded from these data in conjunction with previous findings that the carpal tunnel syndrome is a deficiency disease of vitamin B6, which is probably primary rather than one of a dependency state.  相似文献

This layer was removed with organic solvents or sodium dodecyl sulfate revealing a 50-nm cubic particle. Adoption of reduced‐impact logging (RIL) methods could reduce CO2 emissions by 30–50% across at least 20% of remaining tropical forests. We developed two cost effective and robust indices for comparing the climate benefits (reduced CO2 emissions) due to RIL. The indices correct for variability in the volume of commercial timber among concessions. We determined that a correction for variability in terrain slope was not needed. We found that concessions certified by the Forest Stewardship Council (FSC, N = 3), when compared with noncertified concessions (= 6), did not have lower overall CO2 emissions from logging activity (felling, skidding, and hauling). On the other hand, FSC certified concessions did have lower emissions from one type of logging impact (skidding), and we found evidence of a range of improved practices using other field metrics. One explanation of these results may be that FSC criteria and indicators, and associated RIL practices, were not designed to achieve overall emissions reductions. Also, commonly used field metrics are not reliable proxies for overall logging emissions performance. Furthermore, the simple distinction between certified and noncertified concessions does not fully represent the complex history of investments in improved logging practices. To clarify the relationship between RIL and emissions reductions, we propose the more explicit term ‘RIL‐C’ to refer to the subset of RIL practices that can be defined by quantified thresholds and that result in measurable emissions reductions. If tropical forest certification is to be linked with CO2 emissions reductions, certification standards need to explicitly require RIL‐C practices.  相似文献

Hurricane risk characteristics are examined across the U. S. Gulf of Mexico coastline using a hexagonal tessellation. Using an extreme value model, parameters are collected representing the rate or λ (frequency), the scale or σ (range), and the shape or ξ (intensity) of the extreme wind distribution. These latent parameters and the 30-year return level are visualized across the grid. The greatest 30-year return levels are located toward the center of the Gulf of Mexico, and for inland locations, along the borders of Louisiana, Mississippi, and Alabama. Using a geographically weighted regression model, the relationship of these parameters to sea surface temperature (SST) is found to assess sensitivity to change. It is shown that as SSTs increase near the coast, the frequency of hurricanes in these grids decrease significantly. This reinforces the importance of SST in areas of likely tropical cyclogenesis in determining the number of hurricanes near the coast, along with SSTs along the lifespan of the storm, rather than simply local SST. The range of hurricane wind speeds experienced near Florida is shown to increase with increasing SSTs (insignificant), suggesting that increased temperatures may allow hurricanes to maintain their strength as they pass over the Florida peninsula. The modifiable areal unit problem is assessed using multiple grid sizes. Moran’s I and the local statistic G are calculated to examine spatial autocorrelation in the parameters. This research opens up future questions regarding rapid intensification and decay close to the coast and the relationship to changing SSTs.  相似文献
