MicroRNAs belonging to the miR-34 family have been proposed as critical modulators of the p53 pathway and potential tumor suppressors in human cancers. To formally test these hypotheses, we have generated mice carrying targeted deletion of all three members of this microRNA family. We show that complete inactivation of miR-34 function is compatible with normal development in mice. Surprisingly, p53 function appears to be intact in miR-34-deficient cells and tissues. Although loss of miR-34 expression leads to a slight increase in cellular proliferation in vitro, it does not impair p53-induced cell cycle arrest or apoptosis. Furthermore, in contrast to p53-deficient mice, miR-34-deficient animals do not display increased susceptibility to spontaneous, irradiation-induced, or c-Myc-initiated tumorigenesis. We also show that expression of members of the miR-34 family is particularly high in the testes, lungs, and brains of mice and that it is largely p53-independent in these tissues. These findings indicate that miR-34 plays a redundant function in the p53 pathway and suggest additional p53-independent functions for this family of miRNAs. 相似文献

Malting barley is of high economic and scientific importance. Determining barley grains that are suitable for malting involves measuring malting quality, which is an expensive and complex process. In order to decrease the cost of phenotyping and accelerate the process of developing superior malting barley cultivars, markers for marker-assisted breeding are needed. In this study, we identified quantitative trait loci (QTLs) for malting traits in a Stellar/01Ab8219 F6:8 recombinant inbred line population grown at Aberdeen and Tetonia, Idaho, USA in 2009 and 2010. We identified QTLs associated with malt extract (ME), wort protein, soluble/total protein (S/T), diastatic power (DP), alpha-amylase, beta-glucan (BG) and free amino nitrogen (FAN) at a logarithm of odds score ≥2.5 using a high-density genetic map produced by merging Diversity Arrays Technology markers with the current single nucleotide polymorphism map. Novel QTLs were identified for DP and FAN on chromosome 5H, S/T on 6H, and BG and ME on 7H. Dissection of the genetic regions associated with malting traits suggests the involvement of multiple molecular pathways. The resulting molecular markers may prove useful for barley improvement. 相似文献

Drinks containing 0, 1, 2, and 4% alcohol were consumedover 60 min beginning 30 min after the end of exercise; a differentbeverage was consumed in each of four trials. The volume consumed(2,212 ± 153 ml) was equivalent to 150% of body mass loss. Peakurine flow rate occurred later (P = 0.024) with the 4% beverage. The total volume of urine produced overthe 6 h after rehydration, although not different between trials(P = 0.307), tended to increase as thequantity of alcohol ingested increased. The increase in blood(P = 0.013) and plasma(P = 0.050) volume with rehydrationwas slower when the 4% beverage was consumed and did not increase tovalues significantly greater than the dehydrated level(P = 0.013 andP = 0.050 for blood volume and plasmavolume, respectively); generally, the increase was an inverse functionof the quantity of alcohol consumed. These results suggest that alcoholhas a negligible diuretic effect when consumed in dilute solution aftera moderate level of hypohydration induced by exercise in the heat.There appears to be no difference in recovery from dehydration whetherthe rehydration beverage is alcohol free or contains up to 2% alcohol,but drinks containing 4% alcohol tend to delay the recovery process. 相似文献

This study was designed to examine the effects of alterations in dietary carbohydrate (CHO) intake on the performance of high-intensity exercise lasting approximately 10 min (EXP 1) and 30 min (EXP 2). Trained subjects exercised to exhaustion on four occasions on a cycle ergometer at 90% of maximal oxygen consumption (VO2max; EXP 1, n = 5) and 80% of VO2max (EXP 2, n = 7). The first two tests were familiarisation trials and were carried out following the subjects' normal diet. Normal training was continued but standardised during the periods of dietary control. The subsequent two tests were performed 2 weeks apart after 7 days of dietary manipulation. The two diets were a 70% and a 40% CHO diet, isoenergetic with each subject's normal diet and administered in a randomised order. At both exercise intensities, time to exhaustion following the high CHO and low CHO diets was not different [mean (SD) EXP 1: 11.56 (3.78) min and 8.95 (2.35) min, P = 0.22; EXP 2: 26.9 (7.4) min and 26.5 (6.5) min, P = 0.90]. No differences in resting blood metabolite concentrations were found apart from a lower beta-hydroxybutyrate (beta-HB) level following the high CHO diet in EXP 2. Blood lactate was higher after exercise at 90% of VO2max following the high CHO diet. Blood lactate was higher, and beta-HB lower during exercise at 80% of VO2max following the high CHO diet. No differences were found in the other blood metabolites tested. The respiratory exchange ratio after 15 min of exercise at 80% of VO2max was higher on the high CHO diet. No differences in oxygen uptake, heart rate (EXP 2) or ratings of perceived exertion (both experiments) were found between conditions. These results indicate that moderate changes in diet composition during training do not affect the performance of high-intensity exercise in trained individuals when the total energy intake is moderately high. 相似文献

The end-systolic pressure-volume relationship (ESPVR) defines the systolic limits of cardiac performance. Using an isolated cross-circulated canine heart preparation, we examined the influence of afterload impedance changes, changes in coronary artery pressure (CAP), and regional ischemia on the ESPVR. We found that afterload impedance did not change the slope of the ESPVR but that increases in resistance and characteristic impedance did shift the relation slightly to the left. There was no change in the ESPVR with changes in CAP above a certain critical value. A decrease in CAP below this value caused a progressive decline in the slope of the ESPVR. With regional ischemia the ESPVR became nonlinear, and there was a near parallel downward shift of the ESPVR in the high-volume range. This shift was directly proportional to the extent of ischemic area. We conclude that an adequate measurement of the ESPVR demands at least three pressure-volume points to check for linearity and characterization of both the slope and volume intercept. 相似文献

The shoot and root apical meristems (SAM and RAM, respectively) of plants serve both as sites of cell division and as stem cell niches. The SAM is also responsible for the initiation of new leaves, whereas the analogous process of lateral root initiation occurs in the pericycle, a specialized layer of cells that retains organogenic potential within an otherwise non-dividing region of the root. A picture is emerging of how cell division, growth, and differentiation are coordinated in the meristems and lateral organ primordia of plants. This is starting to reveal striking parallels between the control of stem cell maintenance in both shoots and roots, and to provide information on how signalling from developmental processes and the environment impact on cell behaviour within meristems. 相似文献

Reduced glutathione (GSH) is the most abundant low-molecular weight thiol in plant cells. It accumulates to high concentrations, particularly in stress situations. Because the pathway of GSH synthesis consists of only two enzymes, manipulation of cellular glutathione contents by genetic intervention has proved to be relatively straightforward. The discovery of a new bacterial bifunctional enzyme catalysing GSH synthesis but lacking feedback inhibition characteristics offers new prospects of enhancing GSH production and accumulation by plant cells, while the identification of γ-glutamyl cysteine and glutathione transporters provides additional possibilities for selective compartment-specific targeting. Such manipulations might also be used to affect plant biology in disparate ways, because GSH and glutathione disulphide (GSSG) have crucial roles in processes as diverse as the regulation of the cell cycle, systemic acquired resistance and xenobiotic detoxification. For example, depletion of the total glutathione pool can be used to manipulate the shoot : root ratio, because GSH is required specifically for the growth of the root meristem. Similarly, chloroplast γ-glutamyl cysteine synthetase overexpression could be used to increase the abundance of specific amino acids such as leucine, lysine and tyrosine that are synthesized in the chloroplasts. Here we review the aspects of glutathione biology related to synthesis, compartmentation and transport and related signalling functions that modulate plant growth and development and underpin any assessment of manipulation of GSH homeostasis from the viewpoint of nutritional genomics. 相似文献
