In a previous study, we reported that cicaprost, a stable prostacyclin analogue can inhibit the release of granulocyte-macrophage colony-stimulating factor (GM-CSF) from activated human peripheral mononuclear blood cells (PBMCs). Since interleukin (IL-4) and IL-13 have been shown to inhibit the release of cytokines from PBMCs we tested the hypothesis that prostacyclin in combination with IL-4 or IL-13 can act synergistically to modulate the release of IL-10, generally associated with anti-inflammatory properties, and the pro-inflammatory cytokine tumour necrosis factor alpha (TNF-alpha). For this purpose, PBMCs were isolated over Ficoll, stimulated with lipopolysaccharide (LPS) and incubated in the presence of cicaprost, IL-4 or IL-13. There was a significant reduction in TNF-alpha as well as IL-10 secretion from LPS-stimulated PBMCs following incubation with IL-4 or IL-13. In contrast, cicaprost reduced the secretion of TNF-alpha but led to a slight enhancement of IL-10 release from PBMCs. When LPS-activated PBMCs were incubated in the presence of cicaprost and IL-4 or IL-13 there was a selective, synergistic inhibition of the TNF-alpha release which was not observed for IL-10. Thus, our data suggest that prostacyclin can synergize with cytokines to selectively inhibit the release of pro-inflammatory cytokines from PBMCs.  相似文献

Primers targeting 16S rRNA genes were designed to detect and quantify Eikelboom type 021N organisms by real-time PCR. Eikelboom type 021N filamentous bulking was induced in a laboratory-scale sequencing batch reactor and the evolution of Eikelboom type 021N 16S rRNA and 16S rRNA genes was monitored. A significant correlation was found between the sludge volume index and the amount of these filamentous organisms present in the sludge (r 2=94.6%, n=10, P<0.01), as measured by real-time PCR. The amount of Eikelboom type 021N 16S rRNA genes increased by a factor of 21 during the experiment, while the 16S rRNA increased by a factor of 33. Moreover, Eikelboom type 021N 16S rRNA increased with increased feeding frequency. It was observed that the RNA:DNA ratio peaked before the sludge volume index increased. In parallel, a fluorescence in situ hybridization study indicated a factor of four increase in the length of Eikelboom type 021N filaments, due to a factor of two increase in both length and number of Eikelboom type 021N filaments. Further, an increase in the fraction of filaments extending outside the activated sludge flocs was observed (19–55%). Monitoring of 16S rRNA genes and 16S rRNA of Eikelboom type 021N was shown to be valuable in evaluating activated sludge settling characteristics; and measuring RNA:DNA ratios may be used as an early warning tool for sludge bulking.  相似文献

Fractional flow reserve (FFR) is a commonly used index to assess the functional severity of a coronary artery stenosis. It is conventionally calculated as the ratio of the pressure distal (Pd) and proximal (Pa) to the stenosis (FFR=Pd/Pa). We hypothesize that the presence of a zero flow pressure (P zf), requires a modification of this equation. Using a dynamic hydraulic bench model of the coronary circulation, which allows one to incorporate an adjustable P zf, we studied the relation between pressure-derived FFR=Pd/Pa, flow-derived true FFRQ=QS/QN (=ratio of flow through a stenosed vessel to flow through a normal vessel), and the corrected pressure-derived FFRC=(PdPzf)/(PaPzf) under physiological aortic pressures (70 mmHg, 90 mmHg, and 110 mmHg). Imposed Pzf values varied between 0 mmHg and 30 mmHg. FFRC was in good agreement with FFRQ, whereas FFR consistently overestimated FFRQ. This overestimation increased when Pzf increased, or when Pa decreased, and could be as high as 56% (Pzf=30 mmHg and Pa=70 mmHg). According to our experimental study, calculating the corrected FFRC instead of FFR, if Pzf is known, provides a physiologically more accurate evaluation of the functional severity of a coronary artery stenosis.  相似文献

A variety of immunomodulatory effects have previously been attributed to haptoglobin (Hp). These are supposed to be partly mediated through binding of Hp to CD11b. In the present study, we assessed its effects on T-helper (Th) cytokine production following both in vitro and in vivo stimulation of T-cells. Hp exhibits a dose-dependent inhibitory effect on human T lymphocyte release of the Th2 cytokines (IL-4, IL-5, IL-10 and IL-13) in vitro, whereas it has no clear effect on Th1 cytokine (IL-2 and IFN-gamma) release. When administered an anti-CD3 monoclonal antibody, Hp knockout mice produced more IL-4 and less IFN-gamma than did their wild-type litter-mates. Our findings imply that Hp may be regarded as a regulator of the Th1/Th2 balance in both human and murine immune systems.  相似文献

Urban agriculture is common across Africa and contributes to the livelihoods of urban dwellers. Some crop systems create suitable mosquito breeding sites and thus might affect malaria transmission. The purpose of this study was to identify, map, and characterize potential mosquito breeding sites in agricultural land use zones in a medium-sized town of western C?te d'Ivoire and to assess risk factors for productive Anopheles breeding sites. Two surveys were carried out; one toward the end of the rainy season and the second one during the dry season. In all identified potential mosquito breeding sites, two experienced entomologists searched for the presence of Anopheles larvae and pupae with a standardized technique. Totals of 369 and 589 sites were found in the rainy and dry seasons, respectively, mainly in vegetable gardens and irrigated rice fields. Anopheles larvae were present in 50.7% and 42.4% of the sites investigated during the rainy and dry seasons, respectively. Typical Anopheles larval habitats were characterized by the presence of algae, the absence of floating vegetation, and the co-occurrence of Culex larvae. The highest Anopheles larval productivity was observed in rice paddies, agricultural trenches between vegetable patches, and irrigation wells. An indirect link could be established between the occurrence of productive Anopheles breeding sites and agricultural land use through specific man-made habitats, in particular agricultural trenches, irrigation wells, and rice paddies. Our findings have important bearings for the epidemiology and control of urban malaria in sub-Saharan Africa.  相似文献

Recent evidence suggests that seeking out extra-pair paternity (EPP) can be a viable alternative reproductive strategy for both males and females in many pair-bonded species, including humans. Accurate data on EPP rates in humans, however, are scant and mostly restricted to extant populations. Here, we provide the first large-scale, unbiased genetic study of historical EPP rates in a Western European human population based on combining Y-chromosomal data to infer genetic patrilineages with genealogical and surname data, which reflect known historical presumed paternity. Using two independent methods, we estimate that over the last few centuries, EPP rates in Flanders (Belgium) were only around 1–2% per generation. This figure is substantially lower than the 8–30% per generation reported in some behavioural studies on historical EPP rates, but comparable with the rates reported by other genetic studies of contemporary Western European populations. These results suggest that human EPP rates have not changed substantially during the last 400 years in Flanders and imply that legal genealogies rarely differ from the biological ones. This result has significant implications for a diverse set of fields, including human population genetics, historical demography, forensic science and human sociobiology.  相似文献

Many mammal species adopt marking postures that elevate their scent deposits. The most extreme of these is handstand marking, in which an individual reverses against an upright object, flings its hind legs into the air above its back and balances bipedally on its fore feet. The resulting anogenital deposit is thus raised one full body length above ground level. It has been suggested that this energetically costly form of marking serves to provide conspecifics with information about the marker's body size and hence competitive ability. However, this explanation assumes that the height of an individuals’ deposit does reflect accurately its body size, an assumption that has never been tested in any hand‐standing species. This study investigated the relationship between body size and handstand mark height in a wild population of dwarf mongooses (Helogale parvula) in South Africa. We found that although body size and marking height were correlated positively for female dwarf mongooses, they were not related for males. Male dwarf mongooses (who are subject to intrasexual competition from outside their group) invested more heavily in anogenital range marking, marking at three times the female frequency and placing their deposits significantly higher than females (although they were not dimorphic). Males that were particularly vulnerable to rivals (i.e. those that were small for their age) tended to mark higher than more robust age‐mates, in keeping with the predictions of Adams & Mesterton‐Gibbons’ (1995, J. Theor. Biol.175, 405–421). model of deceptive threat communication. These findings suggest strongly that the height of anogenital scent deposits is of social significance to dwarf mongooses.  相似文献

There is limited knowledge on the biological relatedness between citizens and on the demographical dynamics within villages, towns and cities in pre-17th century Western Europe. By combining Y-chromosomal genotypes, in-depth genealogies and surname data in a strict genetic genealogical approach, it is possible to provide insights into the genetic diversity and the relatedness between indigenous paternal lineages within a particular community at the time of the surname adoption. To obtain these insights, six Flemish communities were selected in this study based on the differences in geography and historical development. After rigorous selection of appropriate DNA donors, low relatedness between Y chromosomes of different surnames was found within each community, although there is co-occurrence of these surnames in each community since the start of the surname adoption between the 14th and 15th century. Next, the high communal diversity in Y-chromosomal lineages was comparable with the regional diversity across Flanders at that time. Moreover, clinal distributions of particular Y-chromosomal lineages between the communities were observed according to the clinal distributions earlier observed across the Flemish regions and Western Europe. No significant indication for genetic differences between communities with distinct historical development was found in the analysis. These genetic results provide relevant information for studies in historical sciences, archaeology, forensic genetics and genealogy.  相似文献
