To explore potential links between plant communities, soil denitrifiers and denitrifier function, the impact of presence, diversity (i.e. species richness) and plant combination on nirK -type denitrifier community composition and on denitrifier activity was studied in artificial grassland plant assemblages over two consecutive years. Mesocosms containing zero, four and eight species and different combinations of two species were set up. Differences in denitrifier community composition were analysed by canonical correspondence analyses following terminal restriction fragment length polymorphism analysis of PCR-amplified nirK gene fragments coding for the copper-containing nitrite reductase. As a measure of denitrifier function, denitrifier enzyme activity (DEA) was determined in the soil samples. The presence as well as the combination of plants and sampling time, but not plant diversity, affected the composition of the nirK -type denitrifier community and DEA. Denitrifier activity significantly increased in the presence of plants, especially when they were growing during summer and autumn. Overall, we found a strong and direct linkage of denitrifier community composition and functioning, but also that plants had additional effects on denitrifier function that could not be solely explained by their effects on nirK -type denitrifier community composition.  相似文献

We have structurally characterized the c-ring from the thermoalkaliphilic Bacillus sp. strain TA2.A1 F1Fo-ATP synthase. Atomic force microscopy imaging and cryo-electron microscopy analyses confirm previous mass spectrometric data indicating that this c-ring contains 13 c-subunits. The cryo-electron microscopy map obtained from two-dimensional crystals shows less closely packed helices in the inner ring compared to those of Na+-binding c11 rings. The inner ring of α-helices in c11 rings harbors a conserved GxGxGxGxG motif, with glycines located at the interface between c-subunits, which is responsible for the close packing of these helices. This glycine motif is altered in the c13 ring of Bacillus sp. strain TA2.A1 to AxGxSxGxS, leading to a change in c-c subunit contacts and thereby enlarging the c-ring diameter to host a greater number of c-subunits. An altered glycine motif is a typical feature of c-subunit sequences in alkaliphilic Bacillus species. We propose that enlarged c-rings in proton-dependent F-ATP synthases may represent an adaptation to facilitate ATP synthesis at low overall proton-motive force, as occurs in bacteria that grow at alkaline pH.  相似文献

Crucial steps in geochemical cycles are in many cases performed by more than one group of microorganisms, but the significance of this functional redundancy with respect to ecosystem functioning is poorly understood. Ammonia-oxidizing archaea (AOA) and their bacterial counterparts (AOB) are a perfect system to address this question: although performing the same transformation step, they belong to well-separated phylogenetic groups. Using pig manure amended with different concentrations of sulfadiazine (SDZ), an antibiotic that is frequently used in veterinary medicine, it was possible to affect AOB and AOA to different degrees. Addition of manure stimulated growth of AOB in both soils and, interestingly, also growth of AOA was considerably stimulated in one of the soils. The antibiotic treatments decreased the manure effect notably on AOB, whereas AOA were affected to a lower extent. Model calculations concerning the respective proportions of AOA and AOB in ammonia oxidation indicate a substantial contribution of AOA in one of the soils that further increased under the influence of SDZ, hence indicating functional redundancy between AOA and AOB.  相似文献

The performance of root hemiparasites depends strongly on host species identity, but it remains unknown whether there exist general patterns in the quality of species as hosts for hemiparasites and in their sensitivity to parasitism. In a comparative approach, the model root hemiparasites Rhinanthus minor and R. alectorolophus were grown with 25 host species (grasses, forbs, and legumes) at two nutrient levels. Hosts grown without parasites served as a control. Host species identity strongly influenced parasite biomass and other traits, and both parasites grew better with legumes and grasses than with forbs. The biomass of R. alectorolophus was much higher than that of R. minor with all host plants and R. alectorolophus responded much more strongly to higher nutrient availability than R. minor. The performance of the two species of Rhinanthus with individual hosts was strongly correlated, and it was also correlated with that of R. alectorolophus and the related Odontites vulgaris in previous experiments with many of the same hosts, but only weakly with that of the less closely related Melampyrum arvense. The negative effect of R. minor on host biomass was less strong than that of R. alectorolophus, but stronger relative to its own biomass, suggesting that it is more parasitic. The impact of the two parasites on individual hosts did not depend on nutrient level and was correlated. Several legumes and grasses were tolerant of parasitism. While R. minor slightly reduced mean overall productivity, R. alectorolophus increased it with several species, indicating that the loss of host biomass was more than compensated by that of the parasite. The results show that closely related parasites have similar host requirements and correlated negative effects on individual hosts, but that there are also specific interactions between pairs of parasitic plants and their hosts.  相似文献

The magnitude of inbreeding depression (ID) varies unpredictably among environments. ID often increases in stressful environments suggesting that these expose more deleterious alleles to selection or increase their effects. More simply, ID could increase under conditions that amplify phenotypic variation (CV²), e.g., by accentuating size hierarchies among plants. These mechanisms are difficult to distinguish when stress increases both ID and phenotypic variation. We grew in- and outbred progeny of Mimulus guttatus under six abiotic stress treatments (control, waterlogging, drought, nutrient deficiency, copper addition, and clipping) with and without competition by the grass Poa palustris. ID differed greatly among stress treatments with δ varying from 7% (control) to 61% (waterlogging) but did not consistently increase with stress intensity. Poa competition increased ID under nutrient deficiency but not other stresses. Analyzing effects of initial size on performance of outbred plants suggests that under some conditions (low N, clipping) competition increased ID by amplifying initial size differences. In other cases (e.g., high ID under waterlogging), particular environments amplified the deleterious genetic effects of inbreeding suggesting differential gene expression. Interestingly, conditions that increased the phenotypic variability of inbred progeny regularly increased ID whereas variability among outbred progeny showed no relationship to ID. Our study reconciles the stress- and phenotypic variability hypotheses by demonstrating how specific conditions (rather than stress per se) act to increase ID. A micromodification for protein determination in tissue material using Amido black 10 B is described. Compared with the method of LOWRY et al. it requires a comparable time expenditure, but has three principal advantages: 1) it is 5-10fold more sensitive; 2) the calibration curve is linear over a virtually unlimited range; 3) it is feasible in the presence of a number of substances frequently used in protein analyses and making difficult or impossible measurement according to LOWRY et al.  相似文献

Summary This study examines the effect of different densities and the removal of all neighbours at different stages of development on all components of reproduction in the inbreeding annual Thlaspi arvense L. A 64-fold increase in density significantly reduced all repooductive components. The number of flower buds per plant was decreased most strongly; the order of decreasing plasticity among the other components was number of capsules per flower, individual seed weight, ovule number per capsule, flowers per flower bud and seeds per ovule. Removing neighbours at all stages of development increased seed yield of plants in comparison to the control without density reduction, but patterns of plasticity depended strongly on time of treatment. The main effect of the removal of neighbours at the vegetative stage was to increase the number of flowers per plant, but the number of ovules per capsule and seed weight increased also, and abortion of capsules decreased. Removing neighbours at the onset of flowering initially failed to affect flower number per plant, instead it resulted in a strong reduction of capsule abortion and an increase in seed weight. However, several weeks after flowering had initially ceased, fresh lateral inflorescences were produced, resulting in a second flush of reproduction. Removing neighbours at the stage of fruit ripening resulted at first only in an increase in seed witht, but later a second reproductive phase occurred. Fresh lateral branches were produced, but the apical meristem was also reactivated. The overall pattern of plasticity among all reproductive components in response to a removal of neighbours was the same as in response to density. The position of a capsule along the inflorescence influenced its number of ovules, the rate of seed abortion and the mean weight of seeds, with the type of effect depending on the developmental stage at which neighbours were removed. Significant negative correlations were found between the mean weight of seeds and the number of seeds in a capsule under all treatments.  相似文献

Strategies to globally protect biological diversity are often hampered by an insufficient ecological knowledge about target species. This also applies to Gagea spathacea (Liliaceae), a ‘vulnerable’ woodland spring geophyte with a distribution largely restricted to the lowlands of Central Europe. We studied whether the species’ linkage to highly fertile forest soils is related to its high nitrogen (N) demands during its short developmental cycle. We hypothesized that the species exhibits a highly efficient N (re)cycling strategy, characterized by efficient resorption of N from the leaves and reallocation to bulbs at the end of the growing season. To test this assumption, we conducted a 15N tracer experiment and quantified 15N flows between soil, leaves, bulbs, and roots. Our findings support our hypothesis that G. spathacea is exceptionally efficient in recycling N, shown by the resorption of 68% of leaf N and its reallocation to bulbs at the end of the growing season. After 6 weeks of growth the plant showed a distinct shift in its N metabolism: The C:N ratio of leaves strongly increased and those of bulbs decreased, leaf 15N enrichment and recovery started to decrease, while total plant 15N recovery remained constant, indicating no further N uptake from the soil. Leaf N reallocation to bulbs was accompanied by a twofold increase of the bulbs’ biomass. Because of the stenoecious behaviour of G. spathacea, a careful protection and sustainable management of G. spathacea forest habitats is necessary, particularly in its Central European core area.  相似文献
