Defective thymine dimer excision in radiation-sensitive mutants rad10 and rad16 of Saccharomyces cerevisiae

Summary Two rad mutants of yeast, rad10 and rad16, are shown to be defective in the removal of UV-induced pyrimidine dimers since DNAs obtained from irradiated cells following a post-irradiation incubation in the dark still retain UV-endonuclease-sensitive sites. Both rad10 and rad16 mutants are in the same pathway of excision-repair as the rad1, rad2, rad3 and rad4 mutants.     

Uptake of glycylglycine by the scutellum of germinating barley grain

The scutella separated from germinating barley grains (Hordeum vulgare L. cv. Himalaya) took up the dipeptide [¹⁴C]glycylglycine (Gly-Gly) rapidly from incubation media. The pH optimum of the process was about 4.5, and the rate of uptake conformed to Michaelis-Menten kinetics with an apparent K_m of 2.3 mm and V_max of 41 μmole gram⁻¹ hour⁻¹. The uptake was strongly inhibited by dinitrophenol and cyanide and by lack of O₂.     

Ecological studies on the deep-water pelagic community of Korsfjorden (western Norway)

As part of a project aimed at assessing the available energy in a pelagic system and the efficiency of its transfer, monthly biomass estimates of the 23 main macroplanktonic and mesoplanktonic species have been made in Korsfjorden (Norway). Sampling was carried out over approximately 3 years (1971–1974) and results are presented as mg dry weight in a 690 m³ water column. The total annual stocks of the various species were not constant in the 3 years of sampling. Principal components analyses of the monthly stocks in each of the 3 years show striking differences in the correlation matrices from year to year. On the basis of known and supposed feeding habits, the species have been classified as herbivores, omnivores, or carnivores. The monthly biomass estimates of these groups show a consistent pattern from year to year. This, in contrast to the variable correlations between individual species, indicates that there can be considerable flexibility in the specific composition of a planktonic community without correspondingly marked effects on its trophic structure.     

Resistance in tomato to the greenhouse whitefly: analysis of population dynamics

Life history parameters including longevity, developmental time, and reproduction were determined for whiteflies, Trialeurodes vaporariorum (Westwood), kept in clip-on cages on susceptible parent, Lycopersicon esculentum, resistant parent, L. hirsutum glabratum, and intermediate breeding lines of tomato. Using the Lewontin triangular reproductive function, the intrinsic rate of increase (r_m) was calculated for each genotype. This is an elegant tool for detecting resistance, as it incorporates all salient factors of life history. Comparing r_m to the other parameters measured, selection criteria were chosen. Developmental time is not a suitable selection criterion as it varies very little even between the most susceptible and the most resistant genotypes. Additionally, developmental time of T. vaporariorum offspring was found to be highly significantly correlated to parental age on all tomato genotypes except L. hirsutum glabratum. Total reproduction, truncated population counts, and longevity appear good criteria for selection. This test, focusing on antibiotic factors, shows large differences between the resistant and susceptible parent genotypes, but much smaller differences between the breeding lines and the susceptible parent. Earlier screenings relying on a variety of tests appear to have maintained antixenotic rather than antibiotic properties in the breeding lines.     

Chloroplast phosphoribulokinase associates with yeast phosphoriboisomerase in the presence of substrate

Pea chloroplastic phosphoribulokinase and yeast phosphoriboisomerase partition independently of one another in a two-phase polyethyleneglycol, dextran system, but apparent interaction is seen when ribose-5-phosphate is added to the two-phase system. It appears that the pea leaf of kinase recognizes yeast isomerase when it is carrying metabolite.
