Little is known about the primary amino acid structure of human cartilage link protein (CRTL1). We screened a human genomic library with a cDNA encoding the 3' untranslated region and the adjoining B1 domain of chicken link protein. One clone was isolated and characterized. A 3.5-kb EcoRI-KpnI fragment from this genomic clone that contains the human B1 exon was used to map the gene to chromosome 5q13----q14.1. The same fragment was used to screen a cDNA library prepared from mRNA of Caco-2, a human colon tumor cell line. Two overlapping clones were isolated and shown to encode all of CRTL1. The deduced amino acid sequence is 354 residues long. The amino acid sequence shows a striking degree of identity to the porcine (96%), rat (96%), and chicken (85%) link protein sequences. Furthermore, there is greater than 86% homology between the 3' untranslated region of the genes encoding human and porcine link proteins. These results indicate that there has been strong evolutionary pressure against changes in the coding and 3' untranslated regions of the gene encoding cartilage link protein.  相似文献

The structures of the tetrahydrogenated menaquinones fromActinomadura angiospora, Faenia rectivirgula, andSaccharothrix australiensis were determined by mass spectrometry and proton nuclear magnetic resonance spectrometry. The positions of saturation of the tetrahydrogenated menaquinones fromFaenia rectivirgula andSaccharothrix australiensis were units II plus III (counting from the ring system), whereas that ofActinomadura angiospora had units III and VIII hydrogenated. The tetrahydrogenated menaquinones fromFaenia rectivirgula andSaccharothrix australiensis are similar to those characterized from other Gram-positive taxa to date, whereas that fromActinomadura angiospora represents a hitherto unknown isomer.  相似文献

Chemotaxonomic studies were performed on some heterofermentative lactobacilli of uncertain taxonomic position. Two strains from beer and six strains from a variety of habitats were found to be distinct from each other and all other Lactobacillus species examined on the basis of DNA-DNA hybridizations and warrant new species for which the names L. malefermentans and L. parabuchneri , respectively, are proposed.  相似文献

This overview presents data showing that glucose use increases and that excitatory amino acids (i.e., glutamate, aspartate), taurine and ascorbate increase in the extracellular fluid during seizures. During the cellular hyperactive state taurine appears to serve as an osmoregulator and ascorbate may serve as either an antioxidant or as a pro-oxidant. Finally, a unifying hypothesis is given for seizure-induced brain damage. This unifying hypothesis states that during seizures there is a release of excitatory amino acids which act on glutamatergic receptors, increasing neuronal activity and thereby increasing glucose use. This hyperactivity of cells causes an influx, of calcium (i.e. calcium stress) and water movements (i.e., osmotic stress) into the cells that culminate in brain damage mediated by reactive oxygen species.Special issue dedicated to Dr. Frederick E. A previously undescribed, H2-oxidizing CO2-reducing acetogenic bacterium was isolated from gut contents of the wood-feeding termite, Pterotermes occidentis. Cells of representative strain APO-1 were strictly anaerobic, Gram-negative, endospore-forming motile rods which measured 0.30–0.40×6–60 m. Cells were catalase positive, oxidase negative, and had 51.5 mol percent G+C in their DNA. Optimum conditions for growth on H2+CO2 were at 30–33°C and pH (initial) 7.8, and under these conditions cells formed acetate according to the equation: 4 H2+2 CO2CH3COOH+2 H2O. Other energy sources supporting good growth of strain APO-1 included glucose, ribose, and various organic acids. Acetate and butyrate were major fermentation products from most organic compounds tested, however propionate, succinate, and 1,2-propanediol were also formed from some substrates. Based on comparative analysis of 16S rRNA nucleotide sequences, strain APO-1 was related to, but distinct from, members of the genus Sporomusa. Moreover, physiological and morphological differences between strain APO-1 and the six known species of Sporomusa were significant. Consequently, it is proposed herewith that a new genus, Acetonema, be established with strain APO-1 as the type strain of the new species, Acetonema longum. A. longum may contribute to the nutrition of P. occidentis by forming acetate, propionate and butyrate, compounds which are important carbon and energy sources for termites.  相似文献

Clostridium mayombei sp. nov., a previously undescribed H2-oxidizing CO2-reducing acetogenic bacterium, was isolated from gut contents of the African soilfeeding termite, Cubitermes speciosus. Cells were anaerobic, Gram positive, catalase and oxidase negative, endospore-forming motile rods which measured 1×2 – 6 m and which had a DNA base composition of 25.6 mol% G+C (strain SFC-5). Optimum conditions for growth on H2+CO2 were at 33°C and pH 7.3, and under these conditions cells produced acetate according to the equation: 4 H2+2 CO2CH3COOH+2 H2O. Other substrates supporting good growth included carbohydrates (e.g. glucose, xylose, starch), sugar alcohols, and organic and amino acids, and with these substrates acetate was almost always the principle fermentation product. Comparative analysis of 16S rRNA nucleotide sequences confirmed that C. mayombei was closely related to various members of the genus Clostridium. However, morphological and physiological differences between C. mayombei and other homoacetogenic clostridia were deemed significant enough to warrant creation of a new taxon. Results are discussed in light of the diversity of H2/CO2 acetogens recently isolated from various termites, and in terms of the relative importance of H2/CO2 acetogenesis to termite nutrition.  相似文献

Body morphology is said to be the all important factor in determining swimming prowess in copepods. Fusion and differentiation of the body (tagmosis) is coupled with advance into the pelagic realm of the Gymnoplea and is thought, by the provision of a rigid thoracic tagma, to promote swimming efficiency. Thus pelagic copepods are believed to be secondarily derived from bottom dwelling predecessors. Experimental evidence is presented to show that the majority of bottom dwelling harpacticoid families, including the most primitive and the most advanced, have representatives that undergo active sustained swimming movements. Such a widespread occurrence is indicative of a conservative evolutionary trait. This primitive behaviour is linked to precopulatory association which takes place necessarily in the water column; it is a feature retained by representatives of all copepod orders. The implication of cephalic appendage vibration (feeding currents) is the essential feature in the swimming success of the Gymnoplea; planktonic efficiency in these is suggested to have evolved coincident with, rather than because of increased tagmosis.  相似文献

The coupling factor ATPase (F1) from photosynthetic membranes of the cyanobacterium Spirulina platensis was purified to homogeneity by a combination of ion-exchange chromatography and sucrose density gradient centrifugation. The ATPase activity of purified Spirulina F1 is latent but can be elicited by trypsin treatment, resulting in specific activities (CaATPase) of 27-37 mumol Pi min-1 mg protein-1. On denaturing sodium dodecyl sulfate-polyacrylamide gradient gels, Spirulina F1 is resolved into five subunits with molecular weights of 53,400, 51,600, 36,000, 21,100, and 14,700, similar to the molecular weights of the subunits of spinach chloroplast coupling factor (CF1). As determined by native polyacrylamide gradient gel electrophoresis, the molecular weight of the Spirulina F1 holoenzyme was estimated to be 320,000, somewhat smaller than the estimated molecular weight of spinach CF1 (392,000). Spirulina F1 was shown to be an active coupling factor by its ability to reconstitute phenazine methosulfate-dependent cyclic photophosphorylation in membrane vesicles which had been depleted of coupling factor content by 2 M NaBr treatment. We estimate the Spirulina F1 content of membrane vesicles to be 1 F1 per 830 chlorophylls or 0.12 mol F1 mol P700(-1), based on the specific ATPase activities of the membrane vesicles and the purified Spirulina F1, the molecular weight of F1, and the P700 content of the vesicles.  相似文献
