—Guinea pigs were killed by asphyxiation with nitrogen and the soluble proteins were extracted from the brain at various times post mortem. The quantity of extractable brain protein decreased by 21 per cent when the animals remained at room temperature for 2 h post mortem. This decrease was not a consequence of extensive proteolysis or variations in blood volume but was probably a result of precipitation. After death, the pH of the brain fell rapidly to a minimum of ~6CE4 within about 35 min. Examination of the patterns of brain proteins after acrylamide gel electrophoresis showed a concomitant decrease in the content of several protein bands.  相似文献

Microwave exposure has been reported to affect various components of the immune system. In this study, we examined the effect of a single whole-body exposure of hamsters to microwave (mw) energy (2.45 GHz; 5-25 mW/cm2; 1 h) on the IgM antibody (Ab) response of spleen cells to sheep red blood cells (SRBC). MW-exposed, sham-exposed, and cage-control hamsters were immunized with SRBC and plaque-forming cells (PFC) in spleens assayed using the direct hemolytic plaque assay. In cage-control hamsters the Ab response was highest between days 4 and 5, returning to baseline by day 9. MW exposure (25 mW/cm2 for 1 h) significantly augmented PFC response only on days 4 and 5 postimmunization, causing approximately a 4.3- and 3.5-fold increase over controls, respectively. Exposure to 15 mW/cm2 caused a lesser, but significant increase in PFC. Exposure to intensities below 15 mW/cm2 for 1 h did not produce any increase in Ab response. Immunization with different concentrations of SRBC following 1 h of 25 mW/cm2 MW exposure revealed a stimulation in PFC at all concentrations ranging from 5 X 10(7) to 5 X 10(8) SRBC. Pretreatment of hamsters with MW radiation prior to immunization showed that the animals retained an increased sensitivity to SRBC for as long as 4 days after MW exposure. In contrast, exposure of hamsters to MW energy on different days after immunization showed an effect of the PFC response only if given between 0 and 1 day after immunization. These results suggest that MW exposure augments the primary IgM response to SRBC by affecting some early event in the immune response process. The various possible explanations for this phenomenon are discussed.  相似文献

Mutants of Zymomonas mobilis were selected for decreased alcohol dehydrogenase activity by using consecutively higher concentrations of allyl alcohol. A mutant selected by using 100 mM allyl alcohol produced acetaldehyde at a level of 4.08 g/liter when the organism was grown in aerated batch cultures on a medium containing 4.0% (wt/wt) glucose. On the basis of the amount of glucose utilized, this level of acetaldehyde production represents nearly 40% of the maximum theoretical yield. Acetaldehyde produced during growth was continuously air stripped from the reactor. Acetaldehyde present in the exhaust stream was then trapped as the acetaldehyde-bisulfite addition product in an aqueous solution of sodium bisulfite and released by treatment with base. Acetaldehyde was found to inhibit growth of Z. mobilis at concentrations as low as 0.05% (wt/wt) acetaldehyde. An acetaldehyde-tolerant mutant of Z. mobilis was isolated after both mutagenesis with nitrosoguanidine and selection in the presence of vapor-phase acetaldehyde. The production of acetaldehyde has potential advantages over that of ethanol: lower energy requirements for product separation, efficient separation of product from dilute feed streams, continuous separation of product from the reactor, and a higher marketplace value.  相似文献

The effect of increased sympathetic activity on skeletal muscle blood flow during acute anemic hypoxia was studied in 16 anesthetized dogs. Sympathetic activity was altered by clamping the carotid arteries bilaterally below the carotid sinus. One group (n = 8) was beta blocked by administration of propranolol (1 mg/kg); a second group (n = 8) was untreated. Venous outflow from the left hindlimb was isolated for measurement of blood flow and O2 uptake (VO2). After a 20-min control period, both carotid arteries were clamped (CC) for 20 min followed by a 20-min recovery period. The sequence was repeated after hematocrit was lowered to about 15% by dextran exchange for blood. Prior to anemia, CC did not alter cardiac output or limb blood flow in either group. After induction of anemia, hindlimb resistance was higher with CC in the beta block than in the no block group. Both limb blood flow and VO2 fell in the beta-block group with CC during anemia. Beta block also prevented the additive increases in whole body VO2 seen with CC and induction of anemia. The data showed that the increased vasoconstrictor tone that was obtained with beta block during anemia was successful in redistributing the lower viscosity blood away from resting skeletal muscle, even to the point that muscle VO2 was decreased.  相似文献

Human leukocyte complement receptor type three (CR3) was shown to be lectin-like and to resemble bovine serum conglutinin (K) in that it bound to both iC3b and unopsonized yeast (Saccharomyces cerevisiae), and was inhibited by EDTA or N-acetyl-D-glucosamine (NADG). CR3 and K also bound to zymosan (Z), a yeast cell wall extract that contains primarily polysaccharide and no detectable protein. However, structural differences and the absence of K on bovine phagocytes indicated that CR3 was not the human homologue of bovine K. Phagocytic and respiratory responses to unopsonized Z were CR3 dependent because they were inhibited by monoclonal antibodies specific for the alpha-chain of CR3 and did not occur with phagocytes from patients with a genetic deficiency of CR3. The binding of CR3 to Z did not require opsonization of the Z with neutrophil-secreted C3, as Z binding and responses were not inhibited by Fab anti-C3. In addition, CR3-dependent binding of yeast occurred with neutrophils from which protein secretion was blocked by fixation with paraformaldehyde. Rabbit erythrocytes (RaE) also bound weakly to neutrophil CR3 and triggered ingestion. Anti-CR3 not only blocked the binding and ingestion of RaE but also blocked selectively the ingestion of RaEC3b without affecting the strong binding mediated by CR1. Even though sheep E and sheep EC3b were not ingested by neutrophils, a weak binding of CR3 to sheep E was suggested by the finding of 20 to 40% inhibition of sheep EAIgG ingestion by anti-CR3. Such inhibition was only observed in buffers that allowed activity of the CR3 binding site and not in buffers containing either EDTA or NADG. An apparently contradictory finding was that the weak CR3-dependent binding of Z triggered neutrophil ingestion and a superoxide burst, whereas the avid CR3-dependent binding of sheep EC3bi did not induce significant ingestion or a respiratory burst. Blocking studies with monoclonal antibodies specific for different epitopes of the alpha-chain of CR3 suggested that this might result from the presence of two distinct binding sites in CR3: one site for fixed iC3b that did not trigger functions, and a second function-triggering site for Z that did not bind to fixed iC3b.  相似文献

Summary Bacteria, isolated from lakewater and capable of growing at the expense of commercial synthetic 2-stroke lubricant oils released 2-ethylhexanol from this substrate by means of esterases. The alcohol accumulated to approximatedly 6mM in these incubations but was itself mineralized by a strain of Pseudomonas putida isolated from the same lakewater by elective culture. The initial steps in the degradative pathway of 2-ethylhexanol were catalysed by alcohol and aldehyde dehydrogenases. The resultant 2-ethylhexanoic acid was further metabolised via -oxidation. The accumulation of intermediates in culture fluids facilitated by acrylate addition and higher isocitrate lyase activities in cell extracts, suggested that 2-ethylhexanoic acid was cleaved to two butyrate moieties which were then further metabolised to acetate. The pathway shows the organism has an interesting -oxidation system capable of utilizing branched substrates.The inducible alcohol dehydrogenase system consisted of two different enzymes, one heat-stable and NAD+-linked, the other heat-labile and PMS-linked, with widely different pH optima of pH 7.0 and 9.0, respectively. Activity staining of extracts resolved by non-denaturing polyacrylamide gel electrophoresis was used to confirm that the enzymes were substantially different moieties.  相似文献

We have examined the relative deficits in tension development and O2 uptake in contracting skeletal muscle during severe hypoxic hypoxia. Anesthetized mongrel dogs were ventilated to maintain an end-tidal PCO2 between 35 and 40 Torr. Venous outflow from the gastrocnemius muscle was measured using an electromagnetic flow probe. The tendon was cut and attached to a strain gauge. The muscle was stimulated to contract isometrically at 2 or 4 Hz for 20 min. Hypoxia (9% O2 in N2) was then imposed for 30 min, followed by 30 min of normoxia. Blood flow first increased in proportion to the contraction frequency and then increased further a similar amount in both groups during hypoxia. O2 extraction and blood flow reached maximal levels during hypoxia in the 2-Hz group. The further O2 deficit that was accumulated during 4 Hz and hypoxia was, therefore, a result of the greater discrepancy between O2 supply and demand. O2 uptake decreased more in hypoxia than did developed tension. These results are best explained by ATP supplementation from nonaerobic energy sources that was promoted by the free-flow condition of hypoxic hypoxia.  相似文献
