Kynurenine is biosynthesised from tryptophan catalysed by indoleamine 2,3-dioxygenase (IDO). The abrogation of kynurenine production is considered a promising therapeutic target for immunological cancer treatment. In the course of our IDO inhibitor programme, formal cyclisation of the isothiourea moiety of the IDO inhibitor 1 afforded the 5-Cl-benzimidazole derivative 2b-6, which inhibited both recombinant human IDO (rhIDO) activity and cellular kynurenine production. Further derivatisation of 2b-6 provided the potent inhibitor of cellular kynurenine production 2i (IC50?=?0.34?µM), which unexpectedly exerted little effect on the enzymatic activity of rhIDO. Elucidation of the mechanism of action revealed that compound 2i suppresses IDO expression at the protein level by inhibiting STAT1 expression in IFN-γ-treated A431 cells. The kynurenine-production inhibitor 2i is expected to be a promising starting point for a novel approach to immunological cancer treatment.  相似文献

The structure of the tryptophan synthase beta2 subunit (Pfbeta2) from the hyperthermophile, Pyrococcus furiosus, was determined by X-ray crystallographic analysis at 2.2 A resolution, and its stability was examined by DSC. This is the first report of the X-ray structure of the tryptophan synthase beta2 subunit alone, although the structure of the tryptophan synthase alpha2beta2 complex from Salmonella typhimurium has already been reported. The structure of Pfbeta2 was essentially similar to that of the beta2 subunit (Stbeta2) in the alpha2beta2 complex from S. typhimurium. The sequence alignment with secondary structures of Pfbeta and Stbeta in monomeric form showed that six residues in the N-terminal region and three residues in the C-terminal region were deleted in Pfbeta, and one residue at Pro366 of Stbeta and at Ile63 of Pfbeta was inserted. The denaturation temperature of Pfbeta2 was higher by 35 degrees C than the reported values from mesophiles at approximately pH 8. On the basis of structural information on both proteins, the analyses of the contributions of each stabilization factor indicate that: (a) the higher stability of Pfbeta2 is not caused by either a hydrophobic interaction or an increase in ion pairs; (b) the number of hydrogen bonds involved in the main chains of Pfbeta is greater by about 10% than that of Stbeta, indicating that the secondary structures of Pfbeta are more stabilized than those of Stbeta and (c) the sequence of Pfbeta seems to be better fitted to an ideally stable structure than that of Stbeta, as assessed from X-ray structure data.  相似文献

The family Megalyridae is recorded from Japan for the first time on the basis of two new species, Carminator helios and C. japonicus. This is the northernmost record of living Megalyridae, although fossil species are known from regions of the Baltic Sea. Carminator helios can be distinguished from congeners of the Asian and Australian regions by the combination of a long head, deeply excavated frons with convex inner margins, entirely striated postgena, the number of fore tibial spines (more than 16), and a long R1 vein. Carminator japonicus is distinguishable from other species by the combination of an orange‐colored metasoma, a weakly excavated frons, and an elongate vertex. This species was collected from the canopy of Acer pictum. Additional characters that distinguish species of the genus Carminator are discussed.  相似文献

The defensive behavior of the aphidDinipponaphis autumna, which forms small, completely closed galls on leaves ofDistylium racemosum, was observed. In mature galls, in addition to tens of mature or nearly mature aphids of the 3rd generation, there remained 3 – 17 1st-instar nymphs of the same generation, which had well-sclerotized legs and the antennae with developed setae. Despite their minority, these 1st-instar nymphs clung to experimentally introduced insect larvae and stung them with their stylets. Fourth-instar wingpadded nymphs, the majority of the 3rd generation at the experiment, also attacked the introduced larvae, but they were readily spilt from the larvae. Four out of 205 1st-instar nymphs remaining in mature galls had the next instar cuticle developing inside, indicating that they are not destined to be sterile.  相似文献

Exogenous auxin applied to the apical bud of intact cucumberseedlings promoted elongation of the hypocotyl, mostly of theapical zone (zone I) and to a lesser extent, the middle andbasal zones (zones II and III). Geotropic curvature was clearlyobserved 30 min after geostimulation. The curvature began atzone I and moved toward the basal zones with prolonged geostimulation.Decapitation (removal of apex and cotyledons) did not produceany appreciable effect on the geotropic response, but the curvaturewas reduced with the removal of zone I. Auxin application toseedlings without zone I restored their ability to respond togravity. The lower epidermis of horizontally oriented hypocotylsplayed a more important role than the upper in the geotropicresponse. The extensibility of the lower epidermal cell wallwas increased significantly by the geostimulus before the onsetof the curvature response. (Received August 24, 1973; )  相似文献

Spinach chloroplasts pre-incubated at various temperatures, changed their photosynthetic activities (measured at 15°C) as the incubation temperature was raised; these activities showed characteristic activitytemperature profiles as follows:
  1. The profiles were shifted to lower temperatures if various aliphatic compounds were present in the incubation mixture.
  2. In general, the extent of the shift was proportional to the product of the concentration and the partition coefficient of the compound, i.e. to the concentration of the compound partitioned in the lipophilic chloroplast phase.
The combined effects of heat and the aliphatic compounds tested indicated that the lipophilic groups in these compounds play a determinative role in the heat denaturation of thylakoids. The consequence of such structure alterations is inactivation of photosynthetic functions. The lipophilic properties of the spinach thylakoid membrane as compared with certain artificial and natural membranes are described.  相似文献

The relationship between clinical parameters and pathological changes was investigated in an animal model of mononeuropathy, by behavioral, electrophysiological and histopathological methods. Mononeuropathy was induced in rats by loosely tying ligatures around the sciatic nerve. Eighty-four rats were used, and these were divided into fourteen groups to determine chronological changes in the withdrawal reflex latency, nerve conduction velocity and ultrastructure of the nerve from 1 to 84 days after nerve ligation surgery. Pathological changes around the ligated nerves were divisible in three phases: the first week was an inflammatory phase, when axonal degeneration, phagocyte infiltration and interstitial edematous changes were observed. The second and third weeks were a nerve-sprouting phase, when numerous axonal sprouts and remyelination were seen. The fourth to twelfth weeks were a recovery phase in which maturing myelination and interstitial fibrosis were characteristic. In the inflammatory phase, withdrawal reflex latencies were shortened, and sensory nerve conduction velocities (SCV) and motor nerve conduction velocities (MCV) gradually decreased. In the nerve-sprouting phase, the latency values remained low, and SCV and MCV were minimal. The parameters examined gradually returned to control levels during the recovery phase. In conclusion, these findings increase the knowledge of disease progression in mononeuropathy with hyperalgesia in human and animal models.  相似文献

The causes and timing of seed death in early regeneration process of Siebold's beech (Fagus crenata Blume) was studied at 15 sites along a snowfall gradient in Japan, in order to clarify why the seedling density of the species has geographic difference remarkably. Seed production did not significantly differ along the snowfall gradient. Pre-dispersal seed mortality by insect damage was higher at sites with light snowfall than at sites with heavy snowfall, but this only seemed to be a minor factor influencing the population. A large proportion of the viable nuts that fall in autumn ware killed in winter before germination. Winter mortality was much higher at sites with thin snow cover than that at sites with thick snow cover, and this factor was strongly correlated with the geographic variation of seedling regeneration probability. There was little seed mortality by winter desiccation. The main factor contributing to the geographic difference seemed to be a seed predation by rodents in winter. Deep snow cover may reduce the success of rodents finding seeds in winter. Thus the observed relationship between snowpack depth and early mortality may be due to an indirect effect through the process of seed predation.p>  相似文献

In an aortic smooth muscle cell line, A10 cells, we investigated the effect of sphingosine 1-phosphate on the induction of heat shock protein 27 (HSP27), a low-molecular-weight heat shock protein. Sphingosine 1-phosphate significantly induced the accumulation of HSP27 in a pertussis toxin-sensitive manner. The effect was dose-dependent in the range between 0.1 and 30 microM. Sphingosine 1-phosphate stimulated an increase in the levels of mRNA for HSP27. Sphingosine 1-phosphate stimulated both p42/p44 mitogen-activated protein (MAP) kinase and p38 MAP kinase activation. PD98059, an inhibitor of the upstream kinase that activates p42/p44 MAP kinase, did not affect sphingosine 1-phosphate-stimulated HSP27 induction. In contrast, SB203580, an inhibitor of p38 MAP kinase, reduced sphingosine 1-phosphate-induced HSP27 induction. SB203580 reduced the levels of mRNA for HSP27 induced by sphingosine 1-phosphate. These results indicate that sphingosine 1-phosphate stimulates the induction of HSP27 via p38 MAP kinase activation in aortic smooth muscle cells.  相似文献

Deletion of the citC gene, coding for isocitrate dehydrogenase, arrests sporulation of Bacillus subtilis at stage I after bipolar localization of the cell division protein FtsZ but before formation of the asymmetric septum. A spontaneous extragenic suppressor mutation that overcame the stage I block was found to map within the spoVG gene. The suppressing mutation and other spoVG loss-of-function mutations enabled citC mutant cells to form asymmetric septa and to activate the forespore-specific sigma factor sigmaF. However, little induction of mother cell-specific, sigmaE-dependent sporulation genes was observed in a citC spoVG double mutant, indicating that there is an additional defect(s) in compartmentalized gene expression in the citC mutant. These other defects could be partially overcome by reducing the synthesis of citrate, by buffering the medium, or by adding excess MnCl2. Overexpression of the spoVG gene in wild-type cells significantly delayed sigmaF activation. Increased expression and stability of SpoVG in citC mutant cells may contribute to the citC mutant phenotype. Inactivation of the spoVG gene caused a population of otherwise wild-type cells to produce a small number of minicells during growth and caused sporulating cells to complete asymmetric septation more rapidly than normal. Unlike the case for inactivation of the cell division inhibitor gene minD, many of these minicells contained DNA and appeared only when the primary sporulation signal transduction pathway, the Spo0A phosphorelay, was active. These results suggest that SpoVG interferes with or is a negative regulator of the pathway leading to asymmetric septation.  相似文献
