From a patient, both a cell line incapable of secreting granulocyte colony-stimulating factor (G-CSF) (TC873) and a cell line capable of secreting G-CSF (TCM902) were established. The effector cells induced, with TC873 cells showed a high lytic capacity against two types of tumor cells. The effector cells induced by TCM902 cells did not show such capacity. Furthermore, the TCM902 cells excreted a factor suppressing the proliferation of lymphokine activated killer (LAK) cells and the autologous tumor cell lysis of tumor associated lymphocytes. Summary Two prey populations that share a common predator can interact indirectly by causing changes in the predator's foraging behaviour. Previous work suggests that adaptive choice of prey by the predator usually has two related consequences: (i) the predation rate on a particular prey species increases with the relative and/or absolute abundance of that prey; and (ii) increases in either prey population produce a short-term increase in the fitness of the other prey (short-term indirect mutualism between prey). This paper investigates how these two consequences are changed if the prey exhibit adaptive anti-predator behaviour. In this case, the predation rate on a particular prey often decreases as the prey's density increases. The predator then usually exhibits negative switching between prey. However, the presence of adaptive antipredator behaviour does not change the short-term mutualism between prey. In this case, as a prey becomes less common, it achieves a larger growth rate by reducing its anti-predator effort. These results imply that observations of the relationship between prey density and predation rate cannot be used to infer the nature of the behavioural indirect effect between prey that share a predator.  相似文献

Human CRK protein is a homolog of the chicken v-crk oncogene product and consists mostly of src homology region 2 (SH2) and SH3, which are shared by many proteins, in particular those involved in signal transduction. SH2 has been shown to bind specifically to phosphotyrosine-containing peptides. We report here that both SH2 and SH3 are required for signaling from CRK protein. Microinjection of the CRK protein induced neurite formation of rat pheochromocytoma cell line PC12. This activity was abolished by mutation of the CRK protein in either SH2 or SH3. The neuronal differentiation induced by the CRK protein was blocked by an excess amount of peptides containing CRK SH3. Moreover, we identified three proteins, of 118, 125, and 136 kDa, which bound specifically to CRK SH3. The CRK-induced neuronal differentiation was also suppressed by monoclonal antibodies against either CRK SH2 or p21ras. These results suggest that both SH2 and SH3 of the CRK protein mediate specific protein-protein binding and that the resulting multimolecular complex generates a signal for neurite differentiation through activation of p21ras.  相似文献

We have characterized an antiserum against basic fibroblast growth factor (bFGF) by immunoblot, investigated the location of bFGF-like immunoreactivity (bFGF-IR) in the trigeminal sensory system and perioral skin endowed with vibrissae, and demonstrated the site of bFGF mRNA expression in the vibrissae by in situ hybridization histochemistry. Light-microscopic immunohistochemistry has demonstrated that bFGF-IR is present not only in trigeminal ganglion neurons and their central and peripheral processes, but also in cells of the matrix, external root sheath and papillae of vibrissae and the stratum basale of the stratified squamous epithelium of the skin. Electron microscopy has revealed intense bFGF-IR mainly in cytoplasmic regions, other than the lumen of rough endoplasmic reticulum and the Golgi apparatus, in trigeminal ganglion neurons, in fibroblast-like cells in the papillae, and in capsules of vibrissae. In contrast, actively proliferating and/or differentiating cells in the matrix of vibrissae have intensely stained euchromatin and weakly labeled cytoplasm that, unlike that of the aforementioned cells, contain immunoreaction products in discrete spots less than 100 nm in diameter, implying the generation of different molecular forms of bFGF in cells of the matrix and papillae. Moreover, the accumulation of bFGF in the euchromatin appears to take place in cells at non-mitotic stages (possibly interphases), characterized by a conspicuous nucleolus and well-developed nuclear envelope. A digoxigenin-labeled cRNA probe for the demonstration of bFGF mRNA gives conspicuous hybridization signals mainly in the matrix of vibrissae. These findings suggest that bFGF is involved in the growth and differentiation of matrix cells during certain periods of the cell cycle and that it acts as a non-mitogenic mediator in the adult trigeminal sensory system.  相似文献

Summary Some properties of community structure are explored using co-evolutionary theory. We consider mathematical models of food webs in which all species in a community adopt foraging behaviours and antipredator behaviours that maximize individual fitness. If the antipredator behaviour of a prey is effective against all its enemies, the number of prey—predator links in a food web must be less than the sum of the numbers of prey and predator species. However, if an increase in a prey's attention to one type of predator decreases its attention to another type of predator, there may be no limit on the number of predator species using a common set of prey species. Predator-specific defence allows a much more complex community structure than non-specific defence. Predator-specific defence more frequently allows a large niche overlap between predators than does non-specific defence. The high connectivity of some fish communities in Lake Tanganyika may be an example of this phenomenon.  相似文献

Summary We analyse mathematical models of the evolution of a trait that determines ability in contest competition. We assume that the value of the competitive trait affects two different components of fitness, one measuring the benefit of winning contests and the other measuring the cost of developing the competitive trait. Unlike previous analyses, we include the population dynamical consequences of larger competitive trait values. Exaggeration of the competitive trait reduces the mean probability of survival during the non-competitive stage of the life cycle. The resulting lower population density reduces competition and, therefore, reduces the advantages of greater competitive ability. Models without population dynamics often predict dimorphism in the competitive trait when resource possession is decided by interactions with many other individuals. If the competition involves a contest with a single other individual, models without population dynamics often predict cycles of increase and collapse in the trait or a continual increase, possibly resulting in extinction. When population dynamics are included, both of these results become less likely and a single stable trait value becomes more likely. Population dynamics also make it possible to have dimorphism when individuals have a single pairwise contest and alternative stable trait values when an individual has many contests. Increases in the value of the resource being contested may increase or decrease the evolutionarily stable size of the trait. Competition between very differently sized species will often decrease size in the larger species (character convergence).  相似文献

It has been reported that growth factors activate Ras through a complex of an adaptor type SH2-containing molecule, Grb2, and a Ras guanine nucleotide-releasing protein (GNRP), mSos. We report on the involvement of another adaptor molecule, CRK, in the activation of Ras. Overexpression of wild-type CRK proteins CRK-I and CRK-II enhanced the nerve growth factor (NGF)-induced activation of Ras in PC12 cells, although the basal level of GTP-bound active Ras was not altered. In contrast, mutants with a single amino acid substitution in either the SH2 or SH3 domain of the CRK-I protein inhibited the NGF-induced activation of Ras. Two GNRPs for the Ras family, mSos and C3G, were coimmunoprecipitated with the endogenous Crk proteins in PC12 cells. The association between C3G and the CRK mutants was dependent upon the presence of intact SH3. The SH2 domain of CRK bound to the SHC protein phosphorylated on tyrosine residues by NGF stimulation. The results demonstrate that, in addition to Grb2, CRK participates in signaling from the NGF receptor and that two GNRPs appear to transmit signals from these adaptor molecules to Ras.  相似文献

We examined the propagation of an infectious disease and the eventual extinction of the host population in a lattice-structured population. Both the host colonization and pathogen transmission processes are assumed to be restricted to act between the nearest neighbor sites. The model is analyzed by an improved version of pair approximation (IPA). Pair approximation is a technique to trace the dynamics of the number of nearest neighbor pairs having particular states, and IPA takes account of the clustering property of lattice models more precisely. The results are checked by computer simulations. The analysis shows: (i) in a one-dimensional lattice population, a pathogen cannot invade a host population no matter how large is the transmission rate; (ii) in a two-dimensional lattice population, pathogens will drive the host to extinction if the transmission rate is larger than a threshold. These results indicate that spatially structured population models may give qualitatively different results from conventional population models, such as Lotka-Volterra ones, without spatial structure.  相似文献
