ABSTRACT.   The Louisiana Waterthrush (waterthrush; Seiurus motacilla ) is a forest-dwelling, Nearctic-Neotropical migratory passerine that nests along streams. We attached radiotransmitters (0.6–0.8 g) to 12 nestling waterthrushes using snug, elastic loops. At three nests, adult waterthrushes were videotaped removing radio-tagged young from the nest. In addition, we recovered nine radio-backpacks (with two still attached to the carcasses of nestlings) near nests within a few days after attaching transmitters. Only one of 12 radio-tagged young was relocated more than 24 h after attaching the transmitter. Thus, the method of transmitter attachment we used was not effective. Using snug, nonelastic loops (e.g., nylon) for the harness may reduce the loss of transmitters, but may injure the skin as fledglings grow. Other possible alternatives include (1) gluing the transmitter to skin on the back of nestlings, (2) capturing fledglings in mist nets and attaching transmitters a week or more after fledging by which time contour feathers have grown and the likelihood of a parent removing the transmitter may be reduced, or (3) attempting to monitor fledglings without attaching transmitters. The success of the latter two alternatives would likely be enhanced by attaching transmitters to adults and then tracking them to locate their still-dependent fledglings.  相似文献

The effect of environmental conditions, especially ammonium concentration, on community composition and nitrification activity of nitrifying bacterial biofilms in a pilot wastewater treatment plant was examined. A decreasing ammonium gradient was created when four aerated tanks with suspended carrier material were serially fed with wastewater. Community composition was analysed using fluorescence in situ hybridization (FISH) probes as well as partial 16S rRNA and amoA gene analysis using polymerase chain reaction-denaturating gradient gel electrophoresis (PCR-DGGE) and sequencing. Fluorescence in situ hybridization probes identified at least five ammonia-oxidizing bacterial (AOB) and two nitrite-oxidizing bacterial (NOB) populations. A change in nitrifying community was detected in the tanks, indicating that ammonium was an important structuring factor. Further, we found support for different autoecology within the Nitrosomonas oligotropha lineage, as at least one population within this lineage increased in relative abundance with ammonium concentration while another population decreased. Absolute numbers of AOB and NOB growing in biofilms on the carriers were determined and the cell specific nitrification rates calculated seemed strongly correlated to ammonium concentration. Oxygen could also be limiting in the biofilms of the first tank with high ammonium concentrations. The response of the nitrifying community to increased ammonium concentrations differed between the tanks, indicating that activity correlates with community structure.  相似文献

We report on a study where 4-day old chicken embryos from different flocks were pre-treated with 50 Hz magnetic fields (MF) prior to a 60-min UV-C exposure (1.7 mW/cm(2)) to investigate the possible protective effect of MF exposure on UV-induced embryo death. Different flux densities (0.010, 0.025, 0.050, 0.10, and 0.20 mT), field directions (vertical and horizontal), as well as MF exposure times (10, 20, and 60 min) were employed. We did not find any significant effects by MF exposure, irrespective of exposure time, flux density, or field direction on the survival of embryos. Neither could we find any flock dependency on sensitivity to MF exposure.  相似文献

Genetic evidence links the Arabidopsis MONOPTEROS (MP) and PIN-FORMED1 (PIN1) genes to the patterning of leaf veins. To elucidate their potential functions and interactions in this process, we have assessed the dynamics of MP and PIN1 expression during vascular patterning in Arabidopsis leaf primordia. Both genes undergo a dynamic process of gradual refinement of expression into files one to two cells wide before overt vascular differentiation. The subcellular distribution of PIN1 is also gradually refined from a non-polar distribution in isodiametric cells to strongly polarized in elongated procambial cells and provides an indication of overall directions of auxin flow. We found evidence that MP expression can be activated by auxin exposure and that PIN1 as well as DR5::GUS expression is defective in mp mutant leaves. Taken together the results suggest a feedback regulatory loop that involves auxin, MP and PIN1 and provide novel experimental support for the canalization-of-auxin-flow hypothesis.  相似文献

Extremely low frequency (ELF) magnetic fields in cell culture incubators have been measured. Values of the order of tens of μT were found which is in sharp contrast to the values found in our normal environment (0.05-0.1 μT). There are numerous examples of biological effects found after exposure to MF at these levels, such as changes in gene expression, blocked cell differentiation, inhibition of the effect of tamoxifen, effects on chick embryo development, etc. We therefore recommend that people working with cell culture incubators check for the background magnetic field and take this into account in performing their experiments, since this could be an unrecognised factor of importance contributing to the variability in the results from work with cell cultures.  相似文献

Early events in the cellular response to DNA damage, such as double strand breaks, rely on lesion recognition and activation of proteins involved in maintenance of genomic stability. One important component of this process is the phosphorylation of the histone variant H2AX. To investigate factors explaining the variation in carcinogenic potency between different categories of polycyclic aromatic hydrocarbons (PAHs), we have studied the phosphorylation of H2AX (H2AXγ). A549 cells were exposed to benzo[a]pyrene diol epoxide [(+)-anti-BPDE] (a bay-region PAH) and dibenzo[a,l]pyrene diol epoxide [(−)-anti-DBPDE] (a fjord-region PAH) and H2AXγ was studied using immunocytochemistry and Western blot. Hydrogen peroxide (H2O2) was used to induce oxidative DNA damage and strand breaks. As showed with single cell gel electrophoresis, neither of the diol epoxides resulted in DNA strand breaks relative to H2O2. Visualisation of H2AXγ formation demonstrated that the proportion of cells exhibiting H2AXγ staining at 1 h differed between BPDE, 40% followed by a decline, and DBPDE, <10% followed by an increase. With H2O2 treatment, almost all cells demonstrated H2AXγ at 1 h. Western blot analysis of the H2AXγ formation also showed concentration and time-dependent response patterns. The kinetics of H2AXγ formation correlated with the previously observed kinetics of elimination of BPDE and DBPDE adducts. Thus, the extent of H2AXγ formation and persistence was related to both the number of adducts and their structural features.  相似文献

Recent work on the fragile fiber mutants of Arabidopsis has identified microtubule-associated proteins that affect the orientation of cellulose microfibrils in cell walls, a major determinant of plant elongation growth. Gonadal hormones and the relative fatty acid composition of serum lecithin and cholesterol ester were studied on four occasions during one cycle in twenty-two regularly menstruating women. The most evident change during the menstrual cycle was a decrease in the sum of polyunsaturated fatty acids of the linoleic series in the late luteal phase. Concomitantly an increase in oleic acid as well as palmitic acid was recorded. These changes were considered to be dietary influenced since a shift of the oleic/linoleic acid ratio is often seen when fat is replaced by sugar and some women are known to increase their intake of refined carbohydrates premenstrually. The only correlation found for fatty acids and hormone levels was a correlation of the ratio oleic/linoleic acids and 17-beta-estradiol. This pattern is not seen after administration of exogenous estrogens and obviously there is a discrepancy between endogenous and exogenous estrogens in this context. Whether physiological fluctuations of sex hormones during the menstrual cycle directly influence the fatty acid composition of serum lecithin and cholesterol ester is uncertain. No changes in dihomo-gamma-linolenic acid or arachidonic acid, the major precursors of prostaglandin synthesis were recorded.  相似文献

Eight ovariectomized women with an intact uterus received 12.5 micrograms ethinylestradiol for 14 days, in a cross over fashion. Ethinylestradiol was administered as a paper disc for sublingual application or as a tablet for oral administration. The parameters studied were FSH secretion, plasma protein synthesis, endometrial morphology, cervical mucus properties and the karyopyknotic index. Both types of treatment resulted in estrogenic effects with a more pronounced effect on sperm penetration and the Spinnbarkeit test after paper disc treatment than after conventional tablet treatment. The post-treatment FSH concentration 20 h after ethinylestradiol administration was significantly lower than the pretreatment value only in the case of the paper disc treatment. There was no evidence of any difference in effects on protein synthesis. The results indicate that the availability of ethinylestradiol for peripheral tissues may be increased by sublingual administration compared to oral tablet administration, whereas the effects on liver protein synthesis may remain unchanged.  相似文献

FAST AXONAL TRANSPORT IN VITRO IN THE SCIATIC SYSTEM OF THE FROG   总被引:7,自引:3,他引:4  
Abstract— An in vitro system from the frog has been used to study fast axonal protein transport. The preparation, which was incubated in a specially made chamber, consisted of the gastrocnemius muscle, the sciatic nerve, the dorsal ganglia and part of the spinal cord. The parts were separated from each other by silicone grease barriers, which made it possible to follow the migration of labelled proteins from the spinal cord and ganglia, along the sciatic nerve, towards the muscle. About 80 per cent of transported proteins in the sciatic nerve originated from the dorsal spinal ganglia and moved antidromically at a rate of 60–90 mm per day at 18°C. The rapidly transported proteins were 90 per cent particulate and mainly associated with structures sedimenting in the microsomal fraction.
The effects of cyclohexirnide showed that the synthesis of rapidly moving proteins and their transport were separate processes. A low concentration of colchicine inhibited the transport when it was present in the medium surrounding the ganglia, but had no effect even at a higher concentration, when it was added to the nerve compartment. The presence of vinblastine at a low concentration in either of the two compartments completely arrested the protein transport. Likewise N-ethylmaleimide or p-chloromercuribenzene sulphonic acid in the nerve medium effectively blocked the fast transport. Results from experiments performed to test the possibility of disto-proximal flow and of transfer of proteins from the muscle to the nerve are discussed.  相似文献
