The C-terminal trigger sequence is essential in the coiled-coil formation of GCN4-p1; its conformational properties are thus
of importance for understanding this process at the atomic level. A solution NMR model structure of a peptide, GCN4p16–31,
encompassing the GCN4-p1 trigger sequence was proposed a few years ago. Derived using a standard single-structure refinement
protocol based on 172 nuclear Overhauser effect (NOE) distance restraints, 14 hydrogen-bond and 11 ϕ torsional-angle restraints,
the resulting set of 20 NMR model structures exhibits regular α-helical structure. However, the set slightly violates some
measured NOE bounds and does not reproduce all 15 measured 3J(HN-HCα)-coupling constants, indicating that different conformers of GCN4p16–31 might be present in solution. With the aim to resolve
structures compatible with all NOE upper distance bounds and 3J-coupling constants, we executed several structure refinement protocols employing unrestrained and restrained molecular dynamics
(MD) simulations with two force fields. We find that only configurational ensembles obtained by applying simultaneously time-averaged
NOE distance and 3J-coupling constant restraining with either force field reproduce all the experimental data. Additionally, analyses of the
simulated ensembles show that the conformational variability of GCN4p16–31 in solution admitted by the available set of 187
measured NMR data is larger than represented by the set of the NMR model structures. The conformations of GCN4p16–31 in solution
differ in the orientation not only of the side-chains but also of the backbone. The inconsistencies between the NMR model
structures and the measured NMR data are due to the neglect of averaging effects and the inclusion of hydrogen-bond and torsional-angle
restraints that have little basis in the primary, i.e. measured NMR data. 相似文献

The recently described antimicrobial peptide melectin (MEP, GFLSILKKVLPKVMAHMK-NH2) exhibits high antimicrobial activity against Gram-positive and Gram-negative bacteria. Here we describe the synthesis and biological activities of 23 new analogues of MEP. We studied the influence of dimerization and tetramerization (MAP-constructs of MEP) on the antimicrobial and hemolytic activities, as well as the role of Met in positions 14 and 17 of the peptide chain. Oxidation of the Met to Met(O) and Met(O2) decreases antimicrobial activity of all tested bacteria if the peptide is in the monomeric form, however, only to Staphylococcus aureus if in the form of dimer or tetramer. Dimerization and tetramerization increase the undesirable hemolytic activity of the peptides. Interestingly, substitution of Leu for Val in position 6 leads to the decrease of hemolytic activity. Introduction of the isosteric amino acid Nle into positions 14 or 17 or both leads to slight increase of hemolytic activity under preservation of high antimicrobial activities. Type IV pili (T4P) are bacterial surface-exposed appendages that have been extensively studied in Gram-negative pathogenic bacteria. Despite recent sequencing efforts, little is known regarding these structures in non-pathogenic anaerobic Gram-positive species, particularly commensals of the mammalian gut. Early studies revealed that T4P in two ruminal Gram-positive species are associated with growth on cellulose, suggesting possible associations of T4P with substrate utilization patterns. In the present study, genome sequences of 118 taxonomically diverse, mainly Gram-positive, bacterial strains isolated from anaerobic (gastrointestinal) environments, have been analysed. The genes likely to be associated with T4P biogenesis were analysed and grouped according to T4P genetic organization. In parallel, consortia of Carbohydrate Active enZYmes (CAZymes) were also analysed and used to predict carbohydrate utilization abilities of selected strains. The predictive power of this approach was additionally confirmed by experimental assessment of substrate-related growth patterns of selected strains. Our analysis revealed that T4P systems with diverse genetic organization are widespread among Gram-positive anaerobic non-pathogenic bacteria isolated from different environments, belonging to two phylogenetically distantly related phyla: Firmicutes and Actinobacteria. 相似文献

Phenotypes of plants, and thus their ecology and evolution, can be affected by the environmental conditions experienced by their parents, a phenomenon called parental effects or transgenerational plasticity. However, whether such effects are just passive responses or represent a special type of adaptive plasticity remains controversial because of a lack of solid tests of their adaptive significance. Here, we investigated transgenerational effects of different nutrient environments on the productivity, carbon storage and flowering phenology of the perennial plant Plantago lanceolata, and whether these effects are influenced by seasonal variation in the maternal environment. We found that maternal environments significantly affected the offspring phenotype, and that plants consistently produced more biomass and had greater root carbohydrate storage if grown under the same environmental conditions as experienced by their mothers. The observed transgenerational effects were independent of the season in which seeds had matured. We therefore conclude that transgenerational effects on biomass and carbon storage in P. lanceolata are adaptive regardless of the season of seed maturation. 相似文献

Nonlinear oscillatory systems, playing a major role in biology, do not exhibit harmonic oscillations. Therefore, one might
assume that the average value of any of their oscillating variables is unequal to the steady-state value. For a number of
mathematical models of calcium oscillations (e.g. the Somogyi–Stucki model and several models developed by Goldbeter and co-workers),
the average value of the cytosolic calcium concentration (not, however, of the concentration in the intracellular store) does
equal its value at the corresponding unstable steady state at the same parameter values. The average value for parameter values
in the unstable region is even equal to the level at the stable steady state for other parameter values, which allow stability.
This holds for all parameters except those involved in the net flux across the cell membrane. We compare these properties
with a similar property of the Higgins–Selkov model of glycolytic oscillations and two-dimensional Lotka–Volterra equations.
Here, we show that this equality property is critically dependent on the following conditions: There must exist a net flux
across the model boundaries that is linearly dependent on the concentration variable for which the equality property holds
plus an additive constant, while being independent of all others. A number of models satisfy these conditions or can be transformed
such that they do so. We discuss our results in view of the question which advantages oscillations may have in biology. For
example, the implications of the findings for the decoding of calcium oscillations are outlined. Moreover, we elucidate interrelations
with metabolic control analysis.

Cell nuclei were isolated from leaf tissues of wild banana (Musa balbisiana, M. acuminata ssp.banksii andM. acuminata ssp.errans) and of the two vegetative clones of diploid cultivar “Pisang Mas”. Relative fluorescence intensity was measured on propidium iodide-stained nuclei by flow cytometry. Nuclei isolated fromGlycine max with known nuclear genome size were used as internal standard to determine nuclear DNA content ofMusa in absolute units. The results of the study showed that the size of nuclear genome ofMusa is smaller than previously estimated. In general, it is smaller in comparison with many other angiosperms. Furthermore, it was found that nuclear DNA content ofM. balbisiana (genome BB) is significantly lower than that ofM. acuminata subspecies and cultivars (genome AA). This finding should permit estimation of genome composition in triploidMusa clones with expected hybrid composition. The research of the spider fauna of Slovenia dates back to the very beginning of binomial nomenclature, and has gone through more and less prolific phases with authors concentrating on taxonomy, faunistics, ecology and zoogeographic reviews. Although the body of published works is remarkable for a small nation, the faunistic data has remained too scattered for a thorough understanding of regional biotic diversity, for comparative and ecological research, and for informed conservation purposes. A national checklist is long overdue. Here, a critical review of all published records in any language is provided. The species list currently comprises 738 species, is published online at http://www.bioportal.si/katalog/araneae.php under the title Araneae Sloveniae, and will be updated in due course. This tool will fill the void in cataloguing regional spider faunas and will facilitate further araneological research in central and southern Europe. 相似文献

IgE against mixtures of common food or respiratory allergens were determined by ELISA in healthy (n = 38) and allergic (n = 62) mothers and their children. Significantly higher level of IgE against respiratory allergens was found in sera of allergic mothers and in cord blood of their children. No correlation between antibody level in maternal and newborn's sera was found; this argues against the transfer of IgE from mother to fetus and points rather to offspring's intrauterine sensitization. Specific IgE level in cord blood was higher in children who developed later allergy than in children who did not. Specific IgE level in colostrum was low both in healthy and allergic mothers; there was no correlation between high concentration of IgE against respiratory allergens in sera of allergic mothers and their colostrum, which does not support the idea of IgE transport from blood to mammary gland. Only slightly increased colostral IgE was detected in allergic mothers whose children manifested allergy later. Allergy of the mother and high level of anti-allergen IgE in her serum and in cord blood are the main predictive factors of future occurrence of allergy in the offspring. A combination of several predictive factors could have higher prognostic value. 相似文献
