Paleodietary analysis on the prehistoric population of El Hierro (Canary Islands)

In order to deepen our knowledge of the dietary habits of the prehispanic inhabitants of El Hierro, we have determined bone strontium (Sr), manganese (Mn), magnesium (Mg), copper (Cu), zinc (Zn), and calcium (Ca) in 52 human tibiae (23 belonging to males and 20 to females individuals) buried in a single burial cave; in 21 modern individuals who served as controls; and in 11 bones of herbivores found at archeological sites of the Canary Islands. Results suggest that females consumed a more vegetarian diet, although site-corrected Sr/Ca ratio of both males and females speaks for a mixed-diet consumption.     

Nuclear inositide signaling: An appraisal of phospholipase C β1 behavior in myelodysplastic and leukemia cells

Location impinges on function of some of the main players of nuclear inositol lipid cycle. Here we have discussed the behavior of PI-PLCβ1 in myelodysplastic and cultured leukemia cells.The presence of a cryptic deletion of PI-PLCβ1 gene in high-risk MDS patients is accompanied by altered expression of its mRNA in that the overall decrease of mRNA is characterized by a dramatic decrease of the splicing variant 1a, which is cytosolic and partially nuclear, whilst the splicing variant 1b is still highly represented. This suggests that altered expression of nuclear PI-PLCβ1 could be involved in a disregulation of the cell cycle and have also important effects on cell apoptotic pathways.Moreover, in cultured leukemia cells (Felc) it has been reported by means of a proteomic approach that the splicing factor SRp20 interacts with nuclear PI-PLCβ1 and its expression is modulated by this signaling molecule.All in all, it appears more and more evident that nuclear signaling elicited by PI-PLCβ1 is a key event in the control of cell cycle progression.     

Synthesis,RNAi activity and nuclease-resistant properties of apolar carbohydrates siRNA conjugates

Oligoribonucleotide conjugates carrying apolar carbohydrates at the 5′-end and the corresponding siRNA duplexes have been prepared using phosphoramidite chemistry. All the carbohydrate-siRNA derivatives were compatible with RNA interference machinery if transfected with oligofectamine. In the absence of a transfection agent, some of them exerted certain reduction of gene expression. Double-tailed permethylated glucose conjugated to siRNA through a long spacer inhibited gene expression up to 26% compared to the scrambled duplex. Such modifications contribute positively to the stability of oligoribonucleotides against 5′-exonuclease degradation.     

Interspecific interaction: The analysis of complex structures in carnivorous zooplankton populations

Linear unidirectional dependence of one population upon another is rather atypical among zooplankton species, as carnivory is not only characteristic of high trophic levels but also part of omnivory in many species of copepods. The feeding on its predatorjuveniles may be important for the survival of a copepod population, more in regard to the impact on the predator than to the nutritional benefit obtained. Such complex interrelationships are important for predictive ecosystem models. Ctenophore-copepod interrelationships have been analysed in experiments, and the results produced have been used for constructing simulation models. Details of experimentation and of modelling are described and discussed.     

Associations between beta-tubulin and mitochondria in adult isolated heart myocytes as shown by immunofluorescence and immunoelectron microscopy

We have investigated the associations between beta-tubulin and mitochondria in freshly isolated cardiac myocytes from the rat. Beta-tubulin was identified by using monoclonal antibodies for immunofluorescence and high resolution immunogold electron microscopy. In addition, conventional transmission and scanning electron microscopic studies were performed. After chemical stabilization in a formaldehyde solution, the myocytes were shock-frozen at -150 degrees C, cryosectioned at -70 degrees C and subsequently processed for immunohistochemical and immunocytochemical microscopy. A characteristic of the rod shaped myocytes is the presence of a dense network of microtubules in the cytoplasm displaying a pattern of strong anti-beta-tubulin reaction. The complexity of this network however varies considerably among the myocytes reflecting microtubule dynamic instability. Further, our findings demonstrate that the beta-tubulin label in rod cells is confined to the perinuclear and interfibrillar spaces and, therefore, is largely colocalized with the cytoplasmic organelles. In myocytes undergoing severe contracture the distribution of beta-tubulin is entirely restricted to the outer mitochondrial-containing domain. This implies that, in a cell model with marked segregation of the contractile filaments and organelles, mitochondria are codistributed with microtubules in the total absence of desmin intermediate filaments. Moreover, our immunogold preparations demonstrate anti-beta-tubulin labelling in the outer mitochondrial membrane as well as of fibres in close apposition to this membrane. These results indicate the presence of a specific beta-tubulin binding to the outer mitochondrial membrane that probably also involves microtubule based translocators and/or MAPs.
