Introduction

Granulocyte-macrophage colony-stimulating factor (GM-CSF) has been shown to be important in the development of inflammatory models of rheumatoid arthritis and there is encouraging data that its blockade may have clinical relevance in patients with rheumatoid arthritis. The aims of the current study were to determine whether GM-CSF may also be important for disease and pain development in a model of osteoarthritis.

Methods

The role of GM-CSF was investigated using the collagenase-induced instability model of osteoarthritis. We studied both GM-CSF-/- mice and wild-type (C57BL/6) mice treated prophylactically or therapeutically with a monoclonal antibody to GM-CSF. Disease development (both early and late) was evaluated by histology and knee pain development was measured by assessment of weight distribution.

Results

In the absence of GM-CSF, there was less synovitis and matrix metalloproteinase-mediated neoepitope expression at week 2 post disease induction, and less cartilage damage at week 6. GM-CSF was absolutely required for pain development. Therapeutic neutralization of GM-CSF not only abolished the pain within 3 days but also led to significantly reduced cartilage damage.

Conclusions

GM-CSF is key to the development of experimental osteoarthritis and its associated pain. Importantly, GM-CSF neutralization by a therapeutic monoclonal antibody-based protocol rapidly and completely abolished existing arthritic pain and suppressed the degree of arthritis development. Our results suggest that it would be worth exploring the importance of GM-CSF for pain and disease in other osteoarthritis models and perhaps clinically for this form of arthritis.  相似文献

Phenotypically cryptic lineages comprise an important yet understudied part of biodiversity; in particular, we have much to learn about how these lineages are formed and maintained. To better understand the evolutionary significance of such lineages, we studied a hybrid zone between two morphologically cryptic phylogeographic lineages in the rainforest lizard, Lampropholis coggeri. Analyzing a multilocus genetic dataset through cline inference, individual-based methods and population measures of disequilibrium and using simulations to explore our genetic results in context of theoretical expectations, we inferred the processes maintaining this hybrid zone. We find that these lineages meet in a hybrid zone that is narrow (≈400 m) relative to inferred dispersal rate. Further, the hybrid zone exhibits substantial genetic disequilibrium and sharply coincident and largely concordant clines. Based on our knowledge about the region's biogeography, the species' natural history, and our simulation results, we suggest that strong selection against hybrids structures this system. As all clines show a relatively narrow range of introgression, we posit that this hybrid zone might not yet be in equilibrium. Nonetheless, our results clearly show that phylogeographic lineages can evolve substantial reproductive isolation without concomitant morphological diversification, suggesting that such lineages can constitute a significant component of evolutionary diversity.  相似文献

Novel C-5 aminomethyl pyrrolotriazines were prepared and optimized for dual EGFR and HER2 protein tyrosine kinase inhibition. The homopiperazine, 1p, emerged as a key lead and it showed promising oral efficacy in EGFR and dual EGFR/HER2 driven human tumor xenograft models. A protocol for somatic embryogenesis in Azadirachta indica A Juss. has been standardized using in vivo leaflets. Experiments were carried out to examine the effect of various auxins, cytokinins, sucrose, inorganic and organic salts on subsequent somatic embryo induction and maturation. Embryogenic calli were induced on Murashige and Skoog (MS) medium supplemented with 1.5 mg dm−3 kinetin and 1.5 mg dm−3 indole-3-acetic acid and subsequently all the stages of somatic embryo development (globular, cordate, torpedo and cotyledonary) were observed. Maturation of these embryos was accomplished with the same growth regulators after three subcultures. The histological study of somatic embryos showed resembles to zygotic embryos. The matured somatic embryos were transferred onto half strength MS-medium devoid of growth regulators for their germination (82 %). Plantlets were acclimatized in the field with a survival rate of 80–83.5 %.  相似文献

The effect of high salt concentration on photosystem II (PS II) electron transport rates and chlorophyll a fluorescence induction kinetics was investigated in coupled and uncoupled spinach thylakoid membranes. With increase in salt concentration, the rates of electron transport mediated by PS II and the F v/F m ratio were affected more in uncoupled thylakoids as compared to coupled thylakoid membranes. The uncoupled thylakoid membranes seemed to behave like coupled thylakoid membranes at high NaCl concentration (∼1 M). On increasing the salt concentration, the uncoupler was found to be less effective and Na+ probably worked as a coupling enhancer or uncoupling suppressor. We suggest that positive charge of Na+ mimics the function of positive charge of H+ in the thylakoid lumen in causing coupled state. The function of NaCl (monovalent cation) could be carried out by even lower concentration of Ca2+ (divalent cation) or Al3+ (trivalent cation). We conclude that this function of NaCl as coupling enhancer is not specific, and in general a positive charge is required for causing coupling in uncoupled thylakoid membranes. The importance of internal hydrogen bonding in a series of benzothiadiazine and 1,4-benzothiazine NS5b inhibitors has been explored. Computational analysis has been used to compare the protonated vs. anionic forms of each series and we demonstrate that activity against HCV NS5b polymerase is best explained using the anionic forms. Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), commonly called gram pod borer, is a serious pest of pulse crops in India and damages chickpea (Cicer arietinum L). On average a 30% crop loss is reported. One of the potential natural enemies reported for its biological control is Campoletis chlorideae Uchida (Hymenoptera: Ichneumonidae), an idiobiont parasitic wasp that attacks second instar larvae of the pod borer. Male-biased sex ratios hinder efforts to mass release parasitic Hymenoptera for biological control by making the production of females costly. Parental age at time of mating is known to affect the progeny sex ratio in some Braconidae. In this view, we examined the reproduction and survival of the parasitoid C. chlorideae in the laboratory [22 ± 4°C, 70 ± 10% R.H. and 10:14 h light:dark photoperiod]. All the nine (i.e. 3 × 3) combinations of possible mating were made for males and females, being each of different age groups (0–12, 48–60 and 96–108 h old). Second instar larvae of the pod borer were reared on chickpea and exposed to mated females of different mating age group combinations. Results revealed that (a) newly emerged females (0–12 h old) mated to aged males yielded more progeny contrasted to the older females inseminated by newly emerged males (0–12 h old). This implies that increased maternal age at mating caused greater reduction in the progeny yield than that of paternal age; (b) the progeny sex ratio was lowest (0.344 ± 0.057) (mean ± SD) when parents mated just after emergence (0–12 h old). It became more and more male biased as parents were deprived of mate and was highest (0.666 ± 0.701) for 96–108 h old parents. The analysis of variance of the data shows that mate deprived males caused greater contribution in the production of sons, resulting a male-biased progeny sex ratio. Our conclusion indicate that newly emerged parasitoids (0–12 h old) are most fecund and should be used in rearing practices in order to enhance the progeny yield, particularly with respect to daughters.
