Leishmania major: nature of immunity induced by immunization with a mutagenized avirulent clone of the parasite in mice

A chemically mutagenized avirulent form of Leishmania major was used to immunize BALB/c and C57B1/6 mice against challenge with virulent L. major. Immunity was elicited when the avirulent parasite was injected intravenously or intraperitoneally, but not subcutaneously. In fact, the latter route of immunization sometimes resulted in exacerbation of a subsequent infection with virulent L. major. Mice immunized with avirulent L. major developed upon challenge with virulent L. major cutaneous lesions which were significantly smaller and contained substantially fewer parasites than lesions on control nonimmune animals. Finally, the protection conferred by immunization with avirulent L. major could be adoptively transferred with T cells of the CD4+ lineage but not the CD8+ lineage.     

An improved method for the extraction of corticosterone from cell homogenates and subcellular fractions of the rat adrenal cortex

An improved technique is described for the extraction and analysis of corticosterone (11β,21-dihydroxy-4-pregnene-3,20-dione) from homogenates and subcellular fractions of the rat adrenal cortex. Factors influencing complete extraction of corticosterone were the nature of the organic solvent system and the concentration of the tissue being extracted. The continued activity of steroidogenic enzymes during subcellular fractionation was prevented by 0.1 mM 1-benzylimidazole. For optimum extraction, homogenates were diluted 1:12 (v/v) in 0.25 M sucrose, containing 0.1 M potassium hydroxide. Dilute homogenate was mixed with absolute ethanol (1:10, v/v) and extracted three times with diethyl ether (1:5, v/v). Following extraction, corticosterone in each sample was isolated by thin-layer chromatography (TLC), quantitated by radioimmunoassay (RIA), and corrected by measuring the recovery of added ³H corticosterone. With these procedures, 90–100% of corticosterone found in extracts of adrenal homogenates was recovered in extracts of subcellular fractions of the homogenates.     

Growth and mineral nutrition of two Chlorococcalean algae

The effect of composition of the medium and pH on the growth of Pediastrum duplex Meyen and Dictyosphaerium pulchellum Wood was studied. Both species showed preference to alkaline conditions. The ammonium nitrate grown colonies of D. pulchellum lacked mucilage and showed a more compact form, resembling D. pulchellum var. minitum Deflandre. From this it appears that D. pulchellum var. minitum is a nutritional variant of the species and not a stable variety.     

Evidence for subclasses of SH groups in (Na++K+)-ATPase.

Changes in the chemical reactivity of the sulfhydryl groups of (Na⁺ + K⁺)-dependent ATPase can be indicative of conformational changes induced by activating ions. Cyanylation of these groups by 5 mM 2-nitro-5-thiobenzoic acid caused a partial inhibition of enzymatic activity. Both this loss and the incorporation of radioactive cyanide from the ¹⁴C-labeled reagent were reduced by inclusion of 50 mM ATP and 150 mM Na⁺ in the incubation. When 10 mM Mg⁺⁺ was added in addition, the inactivation was not different from that produced by cyanylation reagent alone, but the radioactive labeling of protein increased significantly. The data indicate that the sulfhydryl groups of this enzyme exist in two populations, one of which must be free if the enzyme is to function. The other, not essential for enzymatic activity, becomes accessible only when the Na⁺ and Mg⁺⁺-dependent phosphorylation of the enzyme alters its conformation. Inactivation of the enzyme by freezing and thawing increases the incorporation of radioactivity but destroys the responsiveness of labeling to cations and ATP.     

Insecticidal activity of Origanum majorana L. essential oil as anti‐cholinergic agent

The present study was focused on exploring the presence of active compounds in Origanum majorana essential oil (OmEO), and its various knock‐down effects against the rice moth, Corcyra cephalonica. GC–MS analysis detected the existence of major compounds such as monoterpenes, cis‐β‐terpineol and terpinen‐4‐ol with the total proportion of 52.16%. Fumigant toxicity against adult and larvae was calculated with an LC₅₀ value of 11.31 and 49.83 μL/L air, respectively. The contact toxicity against adult, pupa, larvae and eggs was observed with LC₅₀ value 2.54, 0.95, 2.78, and 0.49 μL/L, respectively. Furthermore, the influential repellent behavior against adults has been observed. Acetylesterase (AChE) inhibition activity of OmEO was observed against adult and larvae of C. cephalonica with an IC₅₀ value of 35.89 and 118.54 μL/mL, respectively. Moreover, computational docking study revealed the binding affinity of Cis‐β‐terpineol and terpinen‐4‐ol towards the active binding sites of AChE. On the other hand, Fluorescence‐assisted cytometry and comet assay confirmed the cytotoxic and genotoxic effect of OmEO at various concentrations on C. cephalonica. Altogether, the results showed the knock‐down effect of OmEO against C. cephalonica, and it could be a potential biocontrol measure against the stored product pest.
