Urea transporters (UTs) facilitate urea permeation across cell membranes in prokaryotes and eukaryotes. Bacteria use urea as a means to survive in acidic environments and/or as a nitrogen source. The UT from Actinobacillus pleuropneumoniae, ApUT, the pathogen that causes porcine pleurisy and pneumonia, was expressed in Escherichia coli and purified. Analysis of the recombinant protein using cross-linking and blue-native gel electrophoresis established that ApUT is a dimer in detergent solution. Purified protein was reconstituted into proteoliposomes and urea efflux was measured by stopped-flow fluorometry to determine the urea transport kinetics of ApUT. The measured urea flux was saturable, could be inhibited by phloretin, and was not affected by pH. Two-dimensional crystals of the biologically active ApUT show that it is also dimeric in a lipid membrane and provide the first structural information on a member of the UT family. 相似文献

To better understand visual processing abnormalities in autism we studied the attention orienting related frontal event potentials
(ERP) and the sustained attention related centro-parietal ERPs in a three stimulus oddball experiment. The three stimulus
oddball paradigm was aimed to test the hypothesis that individuals with autism abnormally orient their attention to novel
distracters as compared to controls. A dense-array 128 channel EGI electroencephalographic (EEG) system was used on 11 high-functioning
children and young adults with autism spectrum disorder (ASD) and 11 age-matched, typically developing control subjects. Patients
with ASD showed slower reaction times but did not differ in response accuracy. At the anterior (frontal) topography the ASD
group showed significantly higher amplitudes and longer latencies of early ERP components (e.g., P100, N100) to novel distracter
stimuli in both hemispheres. The ASD group also showed prolonged latencies of late ERP components (e.g., P2a, N200, P3a) to
novel distracter stimuli in both hemispheres. However, differences were more profound in the right hemisphere for both early
and late ERP components. Our results indicate augmented and prolonged early frontal potentials and a delayed P3a component
to novel stimuli, which suggest low selectivity in pre-processing and later-stage under-activation of integrative regions
in the prefrontal cortices. Also, at the posterior (centro-parietal) topography the ASD group showed significantly prolonged
N100 latencies and reduced amplitudes of the N2b component to target stimuli. In addition, the latency of the P3b component
was prolonged to novel distracters in the ASD group. In general, the autistic group showed prolonged latencies to novel stimuli
especially in the right hemisphere. These results suggest that individuals with autism over-process information needed for
the successful differentiation of target and novel stimuli. We propose the potential application of ERP evaluations in a novelty
task as outcome measurements in the biobehavioral treatment (e.g., EEG biofeedback, TMS) of autism. 相似文献

The didelphid marsupial, Didelphis aurita, is suggested as an intraguild predator and as key‐species in small mammal assemblages of the Atlantic Forest of Brazil. The field experiments required to test this hypothesis are complex to implement, but the recent revival of regression methods offers a viable alternative. Here we use the dynamic and static regression methods to determine the importance of D. aurita as a competitor and intraguild predator. Capture–recapture data from two localities in the Rio de Janeiro State were used, Garrafão (municipality of Guapimirim), a coastal forest of the Serra do Mar, and Barra de Maricá, a costal sand dune vegetation. Population and microhabitat variables were monitored from April 1997 to April 2003 in Garrafão, and from January 1986 to July 1990 in Barra de Maricá. Microhabitat variables were related to Canopy, Plant, Litter and Rock covers, Obstruction from 0 to 1.5 m, and Number of logs. Exploitation competition was tested by the dynamic method, which models the effects of D. aurita on the per capita growth rate of a species. Interference by predation or competition was tested by the static method, where the abundance of D. aurita at trap stations was regressed against the abundance of other small mammals, after removal of any variation associated with microhabitat factors. Exploitation competition was not detected, but the interference of D. aurita was pervasive, affecting all small mammals studied in the two localities. The clear avoidance of D. aurita by all small mammals tested in two localities of different physiognomies indicates that it functions as an intraguild predator, even if actual predation by D. aurita is an occasional event. 相似文献

We are currently in an interesting phase of plant biotechnology releases, both for the scientists responsible for these innovations who are beginning to see their ideas realized, and for the biotechnology companies that are starting to see a return on their investment. One of the most notable examples, is the introduction of transgenic crops that are engineered to express a Bacillus thuringiensis toxin that confers resistance to insect predation. Three porphyrin systems have been characterised for use in two-photon fluorescence imaging of biological samples. We have determined the two-photon absorption cross sections (sigma(2)) of the di-cation, free-base and metallated forms of hematoporphyrin derivative (HpD), hematoporphyrin IX (Hp9) and a boronated protoporphyrin (BOPP) using the open-aperture Z-scan and the two-photon induced fluorescence (TPIF) techniques at an excitation wavelength of 800 nm. The insertion of either protons or a metal ion into the macrocycle is shown not to significantly influence the sigma(2) of the porphyrins. Two-photon time-resolved fluorescence images of C6 glioma cells transfected with a free-base form of the BOPP have been obtained as a function of the porphyrin concentration. These studies reveal a maximum useful porphyrin concentration for fluorescence imaging purposes of approximately 30 microg mL(-1). 相似文献

The singlet oxygen formation quantum yield (Phi(Delta)) for solutions of the di-cation, free-base and metallated forms of hematoporphyrin derivative (HpD), hematoporphyrin IX (Hp9) and a boronated protoporphyrin (BOPP) are reported using the method of direct detection of the characteristic phosphorescence following polychromatic excitation. Values of Phi(Delta) for the free-base form of all the porphyrins and the di-cation forms of Hp9 and HpD are in the range of 0.44 to 0.85 in the solvents investigated. Incorporation of zinc ions into the macrocycle reduces Phi(Delta) for all three porphyrins. BOPP facilitates the coordination of certain transition metals (Mn, Co and Cu) compared to Hp9 and HpD and results in a dramatic decrease in Phi(Delta). The experimental data suggest the introduction of low energy charge transfer states associated with the disruption of the planarity of the macrocyclic ring provides alternative non-radiative deactivation pathways. In BOPP, this non-planarity is augmented by the large closo-carborane peripheral substituent groups. 相似文献

BH-3-only BIK functions at the endoplasmic reticulum to stimulate cytochrome c release from mitochondria 总被引:4,自引:0,他引:4
Stimulation of apoptosis by p53 is accompanied by induction of the BH-3-only proapoptotic member of the BCL-2 family, BIK, and ectopic expression of BIK in p53-null cells caused the release of cytochrome c from mitochondria and activation of caspases, dependent on a functional BH-3 domain. A significant fraction of BIK, which contains a predicted transmembrane segment at its COOH terminus, was found inserted in the endoplasmic reticulum (ER) membrane, with the bulk of the protein facing the cytosol. Restriction of BIK to this membrane by replacing its transmembrane segment with the ER-selective membrane anchor of cytochrome b(5) also retained the cytochrome c release and cell death-inducing activity of BIK. Whereas induction of cell death by BIK was strongly inhibited by the caspase inhibitor benzyloxycarbonyl-Val-Ala-Asp-fluoromethylketone, the inhibitor was without effect on the ability of BIK to stimulate egress of cytochrome c from mitochondria. This benzyloxycarbonyl-Val-Ala-Asp-fluoromethylketone-insensitive pathway for stimulating cytochrome c release from mitochondria by ER BIK was successfully reconstituted in vitro and identified the requirement for components present in the light membrane (ER) and cytosol as necessary for this activity. Collectively, the results identify BIK as an initiator of cytochrome c release from mitochondria operating from a location at the ER. 相似文献

MEN 11300 is a hybrid glycoprotein of 297 amino acids obtained by fusion of
the cDNA encoding GM-CSF with the cDNA encoding EPO followed by
transfection of the hybrid gene into CHO cells. The oligonucleotide
construct incorporated a spacing sequence between the two individual cDNAs
which encodes eight amino acids constituting a linker peptide intended to
separate the GM-CSF and EPO moieties. The recombinant MEN 11300 protein was
submitted to a detailed structural characterization including the
verification of the entire amino acid sequence, the assignment of the
disulfide bridges pattern, the identification of the glycosylation sites
and the definition of the glycosidic moiety, including site-specificity.
Partial processing of the C-terminal Arg residue and the occurrence of
N-glycosylation sites at Asn27, Asn155, Asn169, Asn214 were established.
Moreover, O-glycosylation at Ser257 and at the N-terminal region was also
detected. A large heterogeneity was observed in the N-glycans due to the
presence of differently sialylated and fucosylated branched complex type
oligosaccharides whereas O-linked glycans were constituted by GalGalNAc
chains with a different number of sialic acids. The disulfide bridges
pattern was established by direct FABMS analysis of the proteolytic digests
or by ESMS analysis of HPLC purified fractions. Pairing of the eight
cysteine residues resulted in Cys54-Cys96, Cys88-Cys121, Cys138-Cys292, and
Cys160-Cys164. This S-S bridges pattern is identical to that occurring in
the individual natural GM-CSF and EPO, thus showing that the two protein
moieties in MEN 11300 can independently acquire their native
three-dimensional structure.

Bacterial ice nucleation activity (INA+ phenotype) can be traced to the
product of a single gene, ina. A remarkably sparse distribution of this
phenotype within three bacterial genera indicates that the ina gene may
have followed an unusual evolutionary path. Southern blot analyses, coupled
with assays for ice-nucleating ability, revealed that within four bacterial
species an ina gene is present in some strains but absent from others.
Results of hybridization experiments using DNA fragments that flank the ina
gene suggested that the genotypic dimorphism of ina may be anomalous. A
phylogenetic analysis of 16S ribosomal RNA gene sequences from a total of
14 ina+ and ina- bacterial strains indicated that the ina+ bacteria are not
monophyletic but instead phylogenetically interspersed among ina- bacteria.
The relationships of ina+ bacteria inferred from ina sequence did not
coincide with those inferred from the 16S data. These results suggest the
possibility of horizontal transfer in the evolution of bacterial ina genes.
