Hydrogen peroxide was formed in isolated cell walls from Marchantiapolymorpha L. in the presence of MnCl2 by either NADH or NADPHoxidation. This reaction was stimulated by phenols such as 2,4-dichlorophenolor p-coumarate, suggesting a reaction similar to that proposedfor the last step of lignification in higher plant cells, althoughbryophytes have been reported to be devoid of lignin. Stone parsley, soybean, sunflower, sweet potato, potato, andadlay cultivated in a Cd2+-containing medium had Cd-bindingcomplexes with molecular weights of about 4,000 in the roottissues. The complexes were similar to the complex previouslyfound in water hyacinth roots in their absorption and CD spectraand their amino acid compositions. Human liver BCKADH complex was purified. On SDS-polyacrylamide gel electrophoresis, the purified enzyme complex gave three major bands having molecular weights of 51,000, 46,000, and 36,000, and one minor band with a molecular weight of 55,000. The minor band corresponded in molecular weight to lipoamide oxidoreductase which was purified separately. The purified BCKADH represented only approximately 20% of the maximum activity when assayed without addition of exogenous lipoamide oxidoreductase, indicating that lipoamide oxidoreductase component was readily dissociable from the complex. The BCKADH effectively oxidized all of KIV, KIC, and KMV, yielding apparent Km values in the range of 14-17 microM for those alpha-keto acids. Vmax values obtained were 0.86, 0.61, and 0.51 mumole NADH produced/min/mg of protein for KIV, KIC, and KMV, respectively, in the presence of excess amount of lipoamide oxidoreductase. This ratio of Vmax values was practically identical to those of specific activities obtained with respective branched-chain alpha-keto acids at each purification step. The enzyme complex also oxidized pyruvate and alpha-ketoglutarate to a lesser extent. Kinetic experiments gave Km values of 0.98 and 2.9 mM for pyruvate and alpha-ketoglutarate, respectively, with Vmax of 0.43 and 0.08 mumole NADH produced/min/mg of protein. NAD and CoASH were absolutely required for the reaction. Km values for NAD and CoASH were estimated to be 47 and 25 microM, respectively. 相似文献

Resting spore formation during short time-scale upwelling and its significance were investigated in the field and by a simple
theoretical model. Field observations of spore formation ofLeptocylindrus danicus were made off Izu Peninsula, Japan. A rapid increase in ratio of resting spore to vegetative cell numbers indicated thatL. danicus formed resting spores quickly as a response to nutrient depletion in the upwelled water, although only a very low number
of resting spores was found in the upwelling. A simple model was constructed to investigate the possible advantages of spore
formation during short time-scale upwelling. This showed that there is a critical time-scale for resting spore formation to
be advantageous. The nutrient depletion period of the upwelling off Izu was shorter than the critical time-scale determined
by the model. Rapid-sinking of resting spores may increase further the critical time-scale, unless spores return with upwelling
water. For short time-scale upwelling, the vegetative cell may be better suited than the resting spore for enduring a short
period of nutrient depletion.

The distribution of dens and den use by the red fox was studied in Kumamoto Prefecture between 1968 and 1982. A total of 80
dens were classified into natal, residential, and temporarily-visited dens. Natal dens comprised 12.4% of all dens. Relative
proportions were quite stable for a long period. This indicates a constant number of reproducing females in the area for a
considerable length of time. Seasonally the utilization of den showed three peaks in February, May and October. The peaks
coincided well with the peaks of reproductive activity of female foxes. The results of multiple regression analysis of distribution
of dens with respect to selected environmental parameters around densites indicated that the fox selectively utilize open
land, rather than densely vegetated areas. The persistence of dens, the selective utilization by foxes of open land and the
sizes of dens were all related to the breeding activities of female foxes inhabiting the area. Reproductive females usually
used plural dens (“den group”) in one reproductive season. The numbers of those females occuring in the area were maintained
at a steady level for fairy long periods. These indicate that the existence of a stable social relationship between females

Summary We investigated possible association of and linkage between HLA and familial polyposis coli (FPC). In 182 individuals from 66 pedigrees of FPC and 108 individuals from a normal population, HLA-A,-B, and-C antigens were determined. When the frequencies of HLA antigens in 66 unrelated patients and in normal controls were compared, no association of FPC with HLA was observed. For the linkage analysis, HLA haplotypes of 17 affected sib pairs were investigated by the affected sib pair method. The number of pairs which shared two, one, and no haplotypes identical by descent was not significantly different from the number expected with random occurrence (P>0.95). Finally, seven families were analyzed using Morton's sequential test. A maximum lod score of-0.056 at a recombination fraction of 0.4, and a lod of-3.089 at a recombination fraction of 0.05 were obtained. cAMP in Anabaena cylindrica: Rapid Changes in Cellular Levels in Response to Changes in Extracellular Environments 总被引:2,自引:0,他引:2
The cellular level of cyclic 3',5'-AMP (cAMP) in the cyanobacteriumAnabaena cylindrica changed transiently in response to changesin extracellular environments. When the cells were transferedfrom dark to light, or anaerobic to aerobic conditions in thedark, the cAMP level rapidly decreased within one min and thengradually recovered. Addition of carbonyl cyanide m-chlorophenylhydrazone(CCCP) which inhibits ATP synthesis caused an increase in cAMPlevel in the light but not in the dark. The level of cAMP increasedseveral fold by lowering the pH from 8 to 6. On the contrary,a rise of pH from 6 to 8 caused a decrease in the cAMP level.It is suggested that the change in membrane electrochemicalpotential is involved in the regulation of cellular cAMP concentration. Based on ultrastructural features of cellular components of a hemangiopericytoma, hyperplastic cells are classifiable into fibroblast-like (group I), endotheloid (group II) and pericyte-like (group III) cells. The transformation of the group I cells to the group II, or to the group III cells, is pronounced in our electron micrographs and this may imply that the group I cell is the principal cell of origin in this neoplasm. The smooth muscle-like (group IV) cells comprising the media of the arteries and veins in this neoplasm may represent modified, possibly de-differentiated smooth muscle cells reacted to the neoplastic proliferation of the surrounding adventitial (group I) cells. 相似文献
