Background
Reduced estimated glomerular filtration rate (eGFR) and proteinuria are risk factors for end-stage renal disease (ESRD), of which benign nephrosclerosis is a common cause. However, few biopsy-based studies have assessed these associations.Methods
We performed retrospective cohort study of 182 Japanese patients who underwent renal biopsy from June 1985 through March 2014 and who were diagnosed with benign nephrosclerosis. Competing risk regression analyses were used to investigate the effect of eGFR and proteinuria levels at the time of renal biopsy on the risk for renal events (ESRD or a 50% decline in eGFR from baseline).Results
During a median 5.8-year follow-up, 63 (34.6%) patients experienced renal events. The incidence of renal events increased with lower baseline eGFR and greater baseline proteinuria levels. After adjustment for baseline covariates, lower eGFR levels (subhazard ratios [SHRs], 1.30; 95% confidence interval [CI], 1.01–1.67, per 10 mL/min/1.73 m2) and higher proteinuria levels (SHR, 1.52; 95% CI, 1.23–1.87, per 1.0 g/day) at the time of renal biopsy were associated independently with higher risk for renal events. Lower levels of serum albumin (SHR, 2.07; 95% CI, 1.20–3.55 per 1.0 g/dL) were also associated with renal events. Patients with both eGFR <30 mL/min/1.73 m2 and proteinuria ≥0.5 g/day had a 26.7-fold higher risk (95% CI, 3.97–179.4) of renal events than patients with both eGFR ≥60 mL/min/1.73 m2 and proteinuria <0.5 g/day.Conclusions
Reduced eGFR and increased proteinuria as well as lower serum albumin at the time of renal biopsy are independent risk factors for renal events among patients with biopsy-proven benign nephrosclerosis. 相似文献952.

Background
Lung transplantation has been established as the definitive treatment option for patients with advanced lymphangioleiomyomatosis (LAM). However, the prognosis after registration and the circumstances of lung transplantation with sirolimus therapy have never been reported.Methods
In this national survey, we analyzed data from 98 LAM patients registered for lung transplantation in the Japan Organ Transplantation Network.Results
Transplantation was performed in 57 patients as of March 2014. Survival rate was 86.7% at 1 year, 82.5% at 3 years, 73.7% at 5 years, and 73.7% at 10 years. Of the 98 patients, 21 had an inactive status and received sirolimus more frequently than those with an active history (67% vs. 5%, p<0.001). Nine of twelve patients who remained inactive as of March 2014 initiated sirolimus before or while on a waiting list, and remained on sirolimus thereafter. Although the statistical analysis showed no statistically significant difference, the survival rate after registration tended to be better for lung transplant recipients than for those who awaited transplantation (p = 0.053).Conclusions
Lung transplantation is a satisfactory therapeutic option for advanced LAM, but the circumstances for pre-transplantation LAM patients are likely to alter with the use of sirolimus. 相似文献953.

Glucose transport systems in cultured neuronal cells and astrocytes of rats were characterized by measuring the uptake of 2-deoxy-D-[3H]glucose ([3H]2-DG) into the cells. Various sugars inhibited 2-DG uptake by neuronal cells and astrocytes similarly, a finding indicating that the substrate specificities of the transporters in the two types of cells were almost the same. However, the Km values for 2-DG of neuronal cells and astrocytes were 1.7 and 0.36 mM, respectively. The uptake of 2-DG was strongly inhibited by cytochalasin B. Nucleosides, such as adenosine, inosine, and uridine, inhibited 2-DG uptake competitively in both neuronal cells and astrocytes. The uptake by both types of cells were also inhibited by forskolin, but not by cyclic AMP, an observation suggesting that forskolin bound directly to the transporters to cause inhibition. Its inhibition was competitive in astrocytes and noncompetitive in neuronal cells. Astrocytes contained a glucose transporter with a subunit molecular weight of 45K, as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis after photoaffinity labeling using [3H]cytochalasin B as a probe. 相似文献

Rapid growth in culture of Equisetum arvense gametophytes wasobtained using Murashige-Skoog's medium plus 3% (w/v) sucroseand continuous illumination. High-performance liquid chromatographic (HPLC) methods have been developed for the detection of uniconazole-P [(E)-1-(4-chlorophenyl)-4,4,-dimethyl-2-(1,2,4-triazol-1-yl)-1-penten-3-ol; XE-1019; the active ingredient in Prunit and Sumagic] in soil and plant tissue samples. Methanolic extracts of soil and plant samples were dried to the aqueous phase, the pH adjusted to 11, and partitioned against methylene chloride. The methylene chloride phases were washed with pH 11 water and then passed through C-18 solid phase extraction (SPE) columns. The soil extracts were then dried and the residues taken up in 1 ml acetonitrile of which 20 l were injected directly onto a C-18 reverse phase analytical column for HPLC analysis. Plant tissue extracts were purified by partitioning and passing through a sequence of Florisil/C-18/Florisil SPE columns before HPLC analysis. Recovery of uniconazole-P was 70% from soils and 40% from plant tissues. Quantitative detection of 10 parts per billion (ppb) uniconazole-P in plant tissues and soil samples was feasible following these procedures. The soil cleanup procedures were also used to detect uniconazole-P in leachates collected from container-grown plants. 相似文献

Summary Adenine phosphoribosyltransferase (APRT) deficiency causing 2,8-dihydroxyadenine urolithiasis and renal failure is present at a high frequency among the Japanese but not other ethnic groups. A special type of mutant allele, designated APRT*J, with a nucleotide substitution at codon 136 from ATG (Met) to ACG (Thr) is carried by approximately 79% of all Japanese 2,8-dihydroxyadenine urolithiasis patients. We analyzed mutant alleles of 39 APRT deficient patients using a specific oligonucleotide hybridization method after in vitro amplification of a part of the genomic APRT sequence. We found that 24 had only APRT*J alleles. Determination of the haplotypes of 194 APRT alleles from control Japanese subjects and of the 48 different APRT*J alleles indicated that normal alleles occur in four major haplotypes, whereas all APRT*J alleles occur in only two. These results suggest that all APRT*J alleles have a single origin and that this mutant sequence has been maintained for a long period, as calculated from the frequency of the recombinant alleles. 相似文献

Methylazoxymethanol (MAM)-induced cerebral hypoplasia resulted in a significant increase in densities of both serotonin uptake sites in frontal cortex and dopamine uptake sites in striatum, suggesting serotonergic and dopaminergic axon terminals were compressed in the smaller brain volumes. A cell suspension culture, prepared fromPerilla frutescens var.crispa callus induced by Murashige and Skoog (1962) medium containing 2,4-dichlorophenoxyacetic acid (2,4-D, 1.0 ml/l) and kinetin (0.1 mg/l), contained caffeic acid derivatives as the phenolic components. Fresh and dry weights of the cells increased exponentially
for about 11 days after transfer to a fresh medium. The contents of caffeic acid and protein also reached a maximum on the
11th day, but α-amino nitrogen phenylalanine and tyrosine continued to increase in amount until the 20th to 23rd day. Caffeic
acid formation in the cells was increased by lowering the concentration of 2,4-D. The administration ofl-2-aminooxy-3-phenylpropionic acid (l-AOPP), 2-aminooxyacetic acid (AOA) andN-(phosphonomethyl)glycine (glyphosate) to the cells inhibited caffeic acid formation to a large extent. An 80% inhibition
of caffeic acid formation was caused by 10−4Ml-AOPP whereas phenylalanine and tyrosine contents of the cells became 7.5 and 2.3 times higher at thisl-AOPP concentration than those in the control. An 85% inhibition of caffeic acid formation was achieved at 10−3M glyphosate concentration, while 10−3M AOA inhibited caffeic acid formation by 95% and also growth rate by 80%. Summary We have examined the conditions for cultivation of enzymatically dispersed cells from 34 human urothelial transitional cell
carcinomas (TCC) of various types. By employing two culture methods, stationary and tapping suspension, and by using the synthetic
medium DM 160 supplement with human umbilical cord serum and fetal bovine serum, six cell strains were established. In two
strains the tapping suspension culture method was suitable for growth of highly malignant cancer cells that detach easily
from the glass surface in stationary cultures. Each of the six cell strains has been maintained in culture for over 30 months
with repeated subcultures of 32 to 128 times. The histopathological features of the original TCC were three differentiated
papillary types and three anaplastic nonpapillary types. In two cell strains from TCC with low malignancy, however, the cancer
masses that formed in nude mice differed from the original TCC in which they became more malignant, and one cell strain resembled
the original TCC closely. In three stationary culture cell strains the epithelial nature was demonstrated by the presence
of desmosomes and tonofilaments. In one cell strain only tonofilaments were present. In two tapping suspension culture cell
strains the presence of desmosomes was not shown clearly, but fine tonofilaments were observed in one cell strain.
