This study was carried out for the purpose of detecting the relationship between soil-inhabiting tardigrade communities and environmental factors of various forests. Nine forests in the southern part of Kanagawa Prefecture, Central Japan, were selected for this study. Four vegetation types were designated; broadleaved (evergreen/deciduous), coniferous and orchard. In these sites, dry weight of leaf litter, soil pH, soil hardness, soil moisture content and porosity ratio were measured. Wet soil faunal frequencies were also described. The Baermann funnel method was adopted for collecting tardigrades, and DIC microscopy was used for specific identification. To clarify the correlation between environmental variables and tardigrade faunae, multivariate analysis was applied. The tardigrade fauna occurred as two distinct groups. The first group primarily contained Macrobiotus species. The second group contained the genus Diphascon (e.g. D. nobilei, D. patanei, D. prorsirostre). An exception to this was D. pingue, included in the former. Most of the Diphascon species were concentrated in Nebu (coniferous forest site), while, Macrobiotus species were dominant in other sites. Distinct environmental factors could not be identified, but the nematodal frequency was recognized as the main factor in forming these groups. The uniqueness of Nebu, which does not correspond to large-scaled vegetational classification, was substantiated by statistical data. Nebu’s coniferous forest apparently has created a unique environment for sustaining rare species (Diphascon species). This study concluded that forests should be evaluated not only by the macroscopic factors, such as landscape, but also by the microscopic communities, such as those including the tardigrades.  相似文献

Activation-induced cytidine deaminase (AID) is essential to all three genetic alterations required for generation of antigen-specific immunoglobulin: class switch recombination, somatic hypermutation, and gene conversion. Here we demonstrate that AID molecules form a homodimer autonomously in the absence of RNA, DNA, other cofactors, or post-translational modifications. Studies on serial deletion mutants revealed the minimum region between Thr27 and His56 responsible for dimerization. Analyses of point mutations within this region revealed that the residues between Gly47 and Gly54 are most important for the dimer formation. Functional analyses of these mutations indicate that all mutations impairing the dimer formation are inefficient for class switching, suggesting that dimer formation is required for class switching activity. Dimer formation and its requirement for the function of AID are features that AID shares with APOBEC-1, an RNA editing enzyme of apolipoprotein B100 mRNA.  相似文献

The intensity-response (I-R) relations for four wind-sensitive giant interneurons (GIs 8-1, 9-1, 9-2 and 9-3) in the fourth-, sixth- and last-instar nymphs of the cricket, Gryllus bimaculatus, were investigated using a unidirectional air current stimulus in order to explore the functional changes of GIs during postembryonic development. Contrary to our expectations, the response properties of GIs in nymphs were largely different from those in adults. The response magnitude of GI 8-1 in an intact cricket decreased during development, i.e. the GI in younger insects showed a larger response magnitude. Although the response magnitudes of GIs 9-1 and 9-2 were almost identical during the nymphal period, a significant decrease was observed after the imaginal ecdysis. During the nymphal period, the response magnitude of GI 9-3 increased according to the developmental stage. However, it decreased significantly after the imaginal ecdysis. We also investigated the response magnitudes of the GIs in nymphs after unilateral cercal ablation. From the results of ablation experiments, the changes in excitatory and/or inhibitory connections between filiform hairs and each GI during postembryonic development were revealed.  相似文献

We have previously reported the graft transmission of target specificity for RNA silencing using transgenic Nicotiana benthamiana plants expressing the coat protein gene (CP, including the 3′ non-translated region) of Sweet potato feathery mottle virus. Transgenic plants carrying the 5′ 200 and 400 bp regions of CP were newly produced. From these plants, two silenced and two non-silenced lines were selected to investigate the manifestation of transitive RNA silencing by graft experiments. Non-silenced scions carrying the entire transgene were grafted onto either 5′ or 3′ silencing inducer rootstocks. When non-silenced scions were grafted onto 5′ silencing inducer rootstocks, RNA silencing was induced in the non-silenced scions and spread toward the 3′ region of the transgene mRNA. Similarly, when non-silenced scions were grafted onto 3′ silencing inducer rootstocks, RNA silencing was induced in the non-silenced scions, but was restricted to the 3′ region of the transgene and did not spread to the 5′ region. In addition, results from crossing experiments, involving non-silenced and 3′ silencing inducer plants, confirmed the above finding. This indicates that RNA silencing spreads in the 5′–3′ direction, not in the 3′–5′ direction, along the transgene mRNA.  相似文献

The pollen morphology and tapetal membranes of 17 species of Trillium from North America have been examined by scanning and transmission electron microscopy. The pollen grains of the genus are consistently spherical and inaperturate. The exine is unstratified and ultra-thin. Five types of exine structure are recognized, namely, echinate-, corrugate-, granulate-, verrucate- and spinulate-type, based on the variations in the components of the exine. The granulate-type is the most common and is divided into six subtypes. The features of tapetal membranes also vary in relation with the variation of the exine structures. This study demonstrates the potential of palynological evidence in assessment of taxonomic relationships within the genus.  相似文献

The role of macrophages in the process of antigen-induced production of mediators in cellular immune response was studied, using the antigen-induced production of migration inhibitory factor (MIF) as a measure of the activation of immune lymphocytes.The production of MIF by guinea pig immune lymph node cells in response to the stimulation with PPD was abolished when the lymph node cells were depleted of adherent cell population by passing the cells through a Tetron fiber column and incubating the effluent cells in plastic dishes. These purified immune lymphocytes did not respond to particle-bound PPD, either. However, the response was obviously restored by the addition of a small number of the purified peritoneal adherent cells (macrophages) which had been pulse-treated with PPD. The PPD-pulsed macrophages produced no MIF by themselves. Thus, the results clearly indicated the requirement for macrophages in the process of antigen-induced MIF production by immune lymphocytes. Destruction of PPD-pulsed macrophages by freezing and thawing or by homogenization abrogated their ability to stimulate immune lymphocytes. Attempts to restore the response of the purified immune lymphocytes to PPD by adding 2-mercaptoethanol or the culture supernatant of macrophages to the medium have so far been unsuccessful.  相似文献

Abstract Fourteen species of ciliates, seven of which are new, were found living in a sample of anoxic water collected from a small lake in Spain. The species belong to all six orders in which anaerobic ciliates have been described and they include the first anaerobic representatives of the order Prostomatida. This surprising diversity is probably sustained because it embraces all ciliate feeding types, and because protozoa are the only important consumers of the diversity of microbes in anoxic habitats. Six of the anaerobic ciliate species have aerobic congeners; this strengthens the contention that anaerobic ciliates evolved independently from aerobes belonging to several taxonomic groups.  相似文献

We raised monoclonal antibodies against homogenates of ovaries of Ciona intestinalis . We obtained an antibody named GC-1 which specifically recognized early germ cells in C. intestinalis and C. savignyi . Using GC-1 as a marker in immunoelectron microscopy, we determined the morphological sequence of early oogenesis in the ovaries of Ciona . In the stratified epithelium composing the wall of the ovarian tubes, the oocytes were identifiable at the early stages of meiotic prophase according to nuclear features such as condensed chromatin with synaptonemal complexes. GC-1 recognized these early oocytes. We found round cells with large and homogeneous nuclei clustered at the marginal end of the stratified epithelium. We identified these cells as oogonia on the basis of: (1) features of the nucleus, (2) reactivity to GC-1, and (3) early emergence in the developing ovaries. The oogonia were classified into three types: type A was large (7–9 μm in diameter) and clear, type B was intermediate in size (5–6 μm) and electron-density, and type C was small (4–5 μm) and dark. In the developing ovaries of juvenile C. intestinalis, type A oogonia appeared first (before 11 days after settlement) and types B and C followed (15 days after settlement). Thus we see that the type A is the oogenetic stem cell, type B is the proliferating oogonium, and type C is the final oogonium just before meiosis. The oocytes appeared 18 days after metamorphosis.  相似文献

1. Ileal tensions to iodoacetic acid (IAA) develop when tissue ATP concentration falls below approximately 60-55% of the control. 2. As the IAA concentration is increased (0.1-10 mM), the ATP concentrations decrease rapidly, and both the time of the onset and the duration of the contraction shorten. 3. In the presence of lactate, IAA failed to decrease the tissue ATP concentration and did not develop tension. 4. The contraction to IAA developed in the presence of Ca2+ antagonist, D-600 or in Ca2(+)-free solution, however, onset time was prolonged. 5. These results suggest that the contraction to IAA is referred to as 'rigor' because it increases with decreasing tissue ATP concentration in ileum.  相似文献
