The dehydration of d-mannose and the demethanolization of methyl-α-d-mannopyranoside (MαMP) or methyl-α-d-mannofuranoside (MαMF) were examined using microwave-assisted heating for a 3-min irradiation at temperature from 120 to 280 °C in ordinary or dry sulfolane without any catalyst. The microwave-assisted heating of MαMP and MαMF smoothly proceeded to selectively afford the anhydromannoses, 1,6-anhydro-β-d-mannopyranose (AMP) and 1,6-anhydro-β-d-mannofuranose (AMF), respectively, in high yields. For MαMP in ordinary sulfolane at 240 °C, AMP was selectively obtained in the AMF:AMP ratio of 4:96, whereas AMF was the major product at the AMF:AMP ratio of 97:3 from MαMF in dry sulfolane at 220 °C. 相似文献

Quantitative changes in fractional catabolic and synthetic rates of the myosin-actin pool in rat muscle under starvation and refeeding, during growth or after treatment with hydrocortisone were studied by estimating urinary excretion of Nτ-methylhistidine (3-methyl- histidine; Me-His).Following deprivation of food, urinary Me-His output increased from 0.35 mg/day to 0.45 mg/day during first 2 day in spite of decreasing body Me-His pool. This high rate of Me-His excretion was maintained for the following 4 days of starvation and then decreased. When rats were refed a 20% casein diet after 10 days of starvation, Me-His excretion continued to decrease even after 3 days of refeeding. On the fifth day of refeeding, it began to rise progressively. During starvation, fractional catabolic rate of myosin-actin was about 3.7 %/day in comparison with 2.6 %/day of fed rats. After refeeding, the fractional catabolic rate decreased rapidly to a minimum value of 1.7 %/day on the third day. After that, it reached to a value of 2.6 %/day of fed rats. On the other hand, fractional synthetic rate of myosin-actin dropped immediately after fasting and the low rate of about 0.4 %/day was maintained during starvation period. Fractional synthetic rate recovered quickly after refeeding.Urinary output of nitrogen and creatinine rose quickly on the first day after administration of hydrocortisone and on the second day it fell to their normal value. While Me-His excretion increased after injection of hydrocortisone up to 0.52 mg/day on the second day and this high excretion rate remained until the following day. From these results, it was shown that administration of hydrocortisone to rats enhances catabolism and reduces synthesis of myosin-actin. The results also show that the effect of this hormone on myofibrillar protein catabolism appears to last longer than its effect on nitrogen metabolism in the whole body judged from urinary nitrogen output.Fractional rates of catabolism and synthesis of rat myosin-actin were 3.3 %/day (half- life of 21 days) and 7.2%/day, respectively, at the growth stage of 129 g body weight. These rates were 2.3 %/day (half-life of 30 days) and 2.8 %/day, respectively, at the mature stage of 363 g body weight.Under the dietary conditions in this experiment, fractional synthetic rate changed far more dramatically than catabolic rate. This suggests that mass of muscle protein is primarily regulated by the rate of synthesis, although the rate of catabolism should not be neglected. 相似文献

to clarify the genetic diversity and structure of Japanese populations for evaluating on the bottleneck effect and an endangered
species, we analyzed the ND5 region of mitochondrial DNA (mtDNA) and 45 single nucleotide polymorphisms (SNPs) in 640 lacustrine sockeye salmon in Japan
and 80 anadromous sockeye salmon in Iliamna Lake of Alaska. The genetic diversity of the Japanese population in both mtDNA
and SNPs was significantly less than that of the Iliamna Lake population. Moreover, all Japanese populations had SNP loci
deviating from the HWE. In spite of low genetic diversity, the SNP analyses resulted that the Japanese population was significantly
divided into three groups. These suggest that Japanese sockeye salmon populations should be protected as an endangered species

Non-disk-shaped molecules self-assembled to form columnar-type helical aggregates with gel ability through π-stacking interactions among the central tris(phenylethynylphenyl)adamantane moieties, hydrogen-bonding interactions among the alanine parts with amide groups, and van der Waals interactions among the alkyl groups in nonpolar n-alkanes. Food contains various taste substances. Among them, umami substances play an important role with regard to the perception of the taste of food, but, few studies have examined the taste characteristics of representative umami substances other than monosodium L-glutamate (MSG). By conducting mouse behavioral studies (the 48-h 2-bottle preference test and the conditioned taste aversion test) and assessing gustatory nerve responses, we investigated the taste characteristics of unique umami substances, including sodium succinate, L-theanine, betaine, and the enantiomer of MSG, D-MSG. Furthermore, we examined the synergy of umami with inosine 5'-monophoshate (IMP). In the case of the mice, sodium succinate had an umami taste and showed strong synergy with IMP. L-theanine showed synergy with IMP but did not have an umami taste without IMP. In contrast, betaine did not have an umami taste or synergy with IMP. D-MSG might have weak synergy with IMP. 相似文献

We have designed cancer antiproliferative compounds, starting from aniline or phenol derivative, which comprise one or two nitrooxymethylphenyl groups as do the hybrid drugs NCX4040 and NCX530. Compound 2a with p-nitrooxymethylbenzoyl-oxy and -amino groups as well as 8a with a p-nitrooxymethylbenzoylamino group showed more promising effects than NCX4040 against human colon and breast cancer cells. Since 2a and 8a, but not NCX4040, arrested human colon carcinoma HCT116 cells in the M phase, the former two compounds may inhibit cell growth differently from NCX4040. Merged images of immunofluorescence-stained α-tubulin and Hoechst-stained nuclei in human fibrosarcoma HT1080 cells showed that 2a and 8a disrupted microtubule formation just as did vincristine, the tubulin polymerization inhibitor. In experiments in vivo, the intraperitoneal administration of 8a at 80 mg/kg/day reduced the growth of HCT116 xenografts in nude mice to T/C 55%. 相似文献

The outer dynein arms (ODAs) of the flagellar axoneme generate forces needed for flagellar beating. Elucidation of the mechanisms underlying the chemomechanical energy conversion by the dynein arms and their orchestrated movement in cilia/flagella is of great importance, but the nucleotide-dependent three-dimensional (3D) movement of dynein has not yet been observed. In this study, we establish a new method for reconstructing the 3D structure of the in vitro reconstituted ODA-microtubule complex and visualize nucleotide-dependent conformational changes using cryoelectron microscopy and image analysis. As the complex went from the rigor state to the relaxed state, the head domain of the beta heavy chain shifted by 3.7 nm toward the B tubule and inclined 44 degrees inwards. These observations suggest that there is a mechanism that converts head movement into the axonemal sliding motion. 相似文献
