Root colonization of cucumber with P aphanidermatum OP4 was examined in the absence and in the presence of either fluorescent pseudomonad strain CH31 or strain CHI. Quantification was performed by antigen-coated plate enzyme-linked immunosorbent assay (ACP-ELISA) using an antiserum obtained from a rabbit immunized with zoospore cysts: this antiserum did not cross-react with non-infested roots of cucumber, or with the fluorescent pseudomonads CH31 and CHI. Application of the previously developed biotest enabled us to assess the ability of the two strains of fluorescent pseudomonad to suppress root rot caused by P. aphanidermaium OP4.
At all bacterial densities tested, the fluorescent pseudomonad CH31 led to a significant reduction of both the root colonization of cucumber with P aphanidermatum OP4 and the severity of Pythium root rot. It has been hypothesized that humans with familial renal hypouricemia may have a generalized defect of urate transport across cell membranes due to the genetic deletion of a specific carrier, a defect similar to that reported in the Dalmatian dog. In this study the transport of urate labelled with carbon 14 by the erythrocytes of four patients with familial renal hypouricemia was identical to that of five healthy controls. The addition of hypoxanthine to the incubation medium inhibited the transport to a similar extent in the two groups of patients, demonstrating the presence of a carrier specific for urate. This carrier was also found to be present in the erythrocytes of Dalmatian and mongrel dogs. Thus, the renal anomaly causing the hypouricemia in both species is not related to a generalized deletion of a urate-transporting protein on cell membranes. 相似文献

The human cholesteryl ester transfer protein (CETP) facilitates the exchange of neutral lipids among lipoproteins. In order to evaluate the effects of increased plasma CETP on lipoprotein levels, a human CETP minigene was placed under the control of the mouse metallothionein-I promoter and used to develop transgenic mice. Integration of the human CETP transgene into the mouse genome resulted in the production of active plasma CETP. Zinc induction of CETP transgene expression caused depression of serum cholesterol due to a significant reduction of high density lipoprotein cholesterol. There was no change in total cholesterol content in very low and low density lipoproteins. However, there was a decrease in the free cholesterol/cholesteryl ester ratio in plasma and in all lipoprotein fractions of transgenic mouse plasma, suggesting stimulation of plasma cholesterol esterification. The results suggest that high levels of plasma CETP activity may be a cause of reduced high density lipoproteins in humans. 相似文献

Rats were pair-fed isocaloric diets containing either 25% (control diet) or 6% protein (low-protein diet) during the 5 weeks prior to conception and through the gestation and lactation periods; then, carnitine palmitoyltransferase I (CPT-I) activity was determined in liver and skeletal muscle mitochondria isolated from the corresponding pups. Maternal protein undernutrition increased the activity of hepatic CPT-I all along the suckling period, whereas the activity of the skeletal muscle enzyme was unaffected. Moreover, the sensitivity of hepatic CPT-I to inhibition by both malonyl-CoA and 4-hydroxyphenylglyoxylate was decreased in the low-protein group. These alterations in the properties of hepatic CPT-I may be involved in the appearance of hyperketonemia in the rat pup upon maternal administration of low-protein diets. 相似文献

In vitro-primed human lymphocytes proliferate in a secondary mixed lymphocyte reaction (MLR) under the control of MLR-S specificities. HL-A antigens are unable to induce a secondary Proliferation. In familial haploidentical combinations, the secondary proliferation is specific for the priming MLR-S specificity, i.e., as early as 24 to 48 hours after the re-stimulation, a clearcut response is observed toward the sensitizing MLR-S specificity. The secondary response is reflected in acceleration of the reaction rather than in the peak of (3H) TdR uptake. However, when either haploidentical familial primed responding cells or unrelated cells primed toward MLR-S homozygous cells were used, no early typing response was observed against unrelated cells. The level of (3H) TdR incorporation toward cells which possessed and those which did not possess the priming specificity was identical until day 3–4. Noneless, the peak response toward cells possessing the priming MLR-S specificity occurs regularly 24 to 48 hours prior to the peak response toward the cells negative for the priming specificity (day 3–4 as opposed to day 5). Technical improvements are therefore needed before such a technique will provide a clearcut MLR-S typing methodology. 相似文献
