Summary Production of antibodies against peptides or poorly antigenic proteins by conventional methods often requires either large quantities of the native immunogen or some chemical modification to increase their antigenicity. In this study an in vivo and in vitro immunization protocol has been used to generate monoclonal antibodies against the decapeptide luteinizing hormone-releasing hormone (LHRH). Two injections of 100 μg of avian LHRH-I into BALB/c mice were given 7 d apart. Dissociated splenocytes were collected under sterile conditions. They were incubated with 100 μg of the immunogen in 75-cm2 tissue culture flasks in thymocyte-conditioned media. After 5 to 8 d exposure to the antigen, splenocytes were fused with SP2/O myeloma cells by polyethylene glycol. The cells were plated into 24 wells and then incubated in hypoxanthine aminopterin and thymidine selective media. After 14 d an initial screening was done by enzyme immunoassay. The positive wells (6/24) were expanded into 96-well plates and rescreened. Selected lines were cloned out 3 times by limiting dilution and the most positive expanded for ascites production. The antibody was affinity purified in a protein A column. The antibody cross-reacted with LHRH-I and II but preferentially to LHRH-I, as shown by competitive assay. A hypothalamic extract from a mature chick showed a higher response than preparations from whole brain explants of 1- to 3-d posthatched chicks, mature quail, and mature mouse. Chromaffin cells cultured in serum-free medium secreted a smaller percentage of their catecholamine stores in response to stimulation by high K+ (55 mM) than did cells cultured in serum-containing medium. Addition of insulin-like growth factor-I (IGF-I) to serum-free medium restored high K(+)-stimulated catecholamine secretion to the levels seen in serum-treated cultures. In contrast, addition of IGF-I to serum-containing medium had little effect on catecholamine secretion. These results suggest that serum contains IGF-I or another factor that maintains the secretory responsiveness of chromaffin cells. IGF-I not only enhanced high K(+)-stimulated catecholamine secretion, but also augmented secretion elicited by the nicotinic agonist dimethyl-phenylpiperazinium, the dihydropyridine agonist Bay K 8644, and Ba2+. IGF-I did not affect the dependence of catecholamine secretion on extracellular Ca2+ concentration nor did it affect the time course of secretion. Experiments using 45Ca2+ demonstrated that IGF-I treatment enhanced Ca2+ uptake into the cells. When cells were permeabilized by treatment with digitonin, Ca2(+)-dependent catecholamine secretion was slightly, but consistently, greater from IGF-I-treated cells than from untreated cells. Our results suggest that IGF-I may enhance catecholamine secretion partly by increasing Ca2+ entry into the cells and partly by affecting a step distal to Ca2+ entry.  相似文献

Five-day-old, dark-grown seedlings of theEpinastic (Epi) tomato mutant (Lycopersicon esculentum Mill.) and its parent, cultivar VFN8, were used as a system for assessing the role of ethylene in theEpi phenotype. The distinguishing features ofEpi seedlings are an increase in hypocotyl diameter and reduced hypocotyl length. Treatment of VFN8 seedlings with 0.5 l/liter ethylene closely mimicked theEpi phenotype. The rate of ethylene production by 5-day-old, dark-grownEpi seedlings was double that of VFN8 seedlings. Nevertheless, treatment ofEpi seedlings with inhibitors of ethylene biosynthesis (aminoethoxyvinylglycine or Co2+) or ethylene action (silver thiosulfate or norbornadiene) failed to normalize theEpi phenotype.Epi seedlings grown in sealed jars containing ethylene and CO2 adsorbants also expressed the characteristicEpi phenotype. The results indicate that the physiological lesion resulting from theEpi gene mutation is not simply an overproduction of ethylene.  相似文献

The importance of litter to nutrient and organic matter storage and the possible influence of species selection on soil fertility in ten stands each consisting of a separate tree species were examined in this study. The plantations had been grown under similar conditions in an arboretum in the Luquillo Experimental Forest, Puerto Rico. The species involved were: Anthocephalus chinensis, Eucalyptus × patentinervis, E. saligna, Hernandia sonora, Hibiscus elatus, Khaya nyasica, Pinus caribaea var. hondurensis, P. elliottii var. densa, Swietenia macrophylla, and Terminalia ivorensis. After 26 yr, litter mass ranged from 5 mg ha-1 in the H. sonora stand to 27.2 Mg ha-1 in the P. caribaea stand. Nutrients in the litter (N, P, K, Ca, and Mg) also varied widely, but stands were ranked in different order when ranked by nutrients in the litter than then ranked according to accumulation of mass. Only E. saligna and A. chinensis stands were ranked similarly in accumulation of both nutrients and mass, and the stand of H. elatus was ranked higher with respect to nutrient accumulation than to accumulation of mass. The nutrient concentration in standing leaf litter generally increased in the order of recently fallen <old intact< fragmented. Nutrient concentration of standing leaf litter appears to increase with age and depth in the litter layer. The amount of nutrients stored in the litter compartment of these plantations was in the same order of magnitude as the quantity of available nutrients in the top 10-cm of mineral soil. Total litter mass was negatively correlated with the mass-weighted concentration of N, K, and Mg. The same relationship was found for Ca in the leaf litter and N in the fine wood litter compartments. In some stands (notably P. caribaea, P. elliottii, and E. saligna), leaf litter derived from species other than the species planted in that particular stand had higher nutrient concentration than leaf litter from the planted species. Soils of the 10 stands were classified in the same soil series and had similar texture (clay soils). However, significantly different chemical characteristics were found. Results obtained by analysis of covariance and by limiting comparisons to adjacent stands with similar soil texture, indicate that different species have had different influences on the concentration of available nutrients in soil.  相似文献

The differentiation of T Lymphocytes within the thymus is an important biological phenomenon during wich these cell acquire their functions to further control the immune system. Numerous experiments under various conditions have been devised to understand the different mechanisms involved in this complex process. Nevertheless, interpretation of these experiments lead to still contradictory debatable hypotheses. Modelisation of this process through classical simulation methods cannot be envisaged because they are not adapted to modifications of the model structure, which is the point of interest. For these reasons, we proposed a new approach of automatic search for model. The program consists of four independent connected modules : The generator produces model, based on the rationale of formal grammars. Protocol and experimental data are stored in a set of experiments. The simulator using a protocol and a model provides simulated results. Finally, the supervisor by comparing simulated results and experimental data, adapts the model parameters to increase their fit and either chooses a new experiment to explore, or modifies the model structure. Change of the model structure is performed among still unexplored models according to their promise level, which is iteratively evaluated relatively to previously explored models through a proposed model distance. The generator is written in Prolog and the other modules in C++. The architecture of the program allows us to modify or complete a module without changing anything in the other modules. As a consequence, the proposed modeling approach conceived to study T lymphocyte differentiation within the thymus remains independent of this biological phenomenon and can be applied to other biological problems.  相似文献

The putative periclinal chimeraRhododendron xlimbatum President Roosevelt was used to study the origin of shoots in vitro. Genotypic segregation readily occurred in vitro. Numerous phenotypes were observed, although most shoots were either entirely green or maintained the original variegation pattern. Derivatives of the third apical layer were rarely involved in shoot formation. A reversed chimeral form was isolated. Adventitious shoots were usually miniaturized and rapidly proliferating, but axillary shoots had thicker stems, larger leaves and proliferated more slowly. Corolla tissue produced stunted, leafy shoots; no variegated shoots were produced from floret explants. In shoot tip cultures the addition of 40M 2iP without IBA resulted in the greatest number of shoots. Explant choice was the most critical factor for maintenance of foliar variegation.  相似文献

Summary The enzyme loading needed to achieve substrate saturation appeared to be the most economical enzyme concentration to use for hydrolysis, based on percentage hydrolysis. Saturation was reached at 25 filter paper units per gram substrate on Solka Floc BW300, as determined by studying (a) initial adsorption of the cellulase preparation onto the substrate, (b) an actual hydrolysis or (c) a combined hydrolysis and fermentation (CHF) process. Initial adsorption of the cellulases onto the substrate can be used to determine the minimal cellulase requirements for efficient hydrolysis since enzymes initially adsorbed to the substrate have a strong role in governing the overall reaction. Trichoderma harzianum E58 produces high levels of -glucosidase and is able to cause high conversion of Solka Floc BW300 to glucose without the need for exogenous -glucosidase. End-product inhibition of the cellulase and -glucosidase can be more effectively reduced by employing a CHF process than by supplemental -glucosidase.Offprint requests to: C. M. Hogan  相似文献
