Orphanin FQ/nociceptin (OFQ/N), the endogenous ligand for the "orphan" opioid receptor ORL-1 (NOP(1)) was first identified in 1995. In the years since its discovery, a large body of evidence has accumulated showing that OFQ/N and its receptor are widely distributed in the nervous system, and showing that OFQ/N has potent and indiscriminate inhibitory actions on neurons in many regions. However, numerous studies investigating the functional role of OFQ/N in physiology or behavior have failed to provide a coherent view. Pain and analgesia have been the best studied, and administration of OFQ/N is reported to have no effect, to produce hyperalgesia, analgesia or anti-hyperalgesia. Effects of OFQ/N receptor antagonists have proved similarly contentious. In an attempt to resolve this controversy, we investigated the actions of OFQ/N on the activity of physiologically characterized neurons in the rostral ventromedial medulla, a region with a well-documented role in pain modulation(Heinricher et al., 1997). The results of those experiments demonstrate that this peptide is neither "anti-opioid" or "anti-hyperalgesic". It is simply inhibitory. For this reason, the effects seen in functional studies will only be fully understood when examined in the context of identified neural circuits. 相似文献

It has been suggested that the use of avidin-biotin immunohistochemical techniques for antigen detection in neural tissue produces nonspecific background staining. For this reason neural tissue was used to test the quality, sensitivity and specificity of four commercially available antibody detection kits which use avidin or streptavidin binding to biotin. Free-floating, thick-section immunohistochemistry on perfusion fixed rat central nervous system revealed variability among staining kits for all parameters analyzed under the same experimental conditions. The reagents from the Vector 'Elite' kit were the most sensitive and specific, and received the highest overall rating for quality. Most commercial products tested could be used at greater dilutions than those recommended by the manufacturers without compromising specific staining. No staining was evident when the primary and secondary antibodies were omitted. This suggests that nonspecific binding is unlikely to be due to endogenous ligands, charge of hydrophilic reactions between these tertiary complexes and the tissue sections. 相似文献

Simultaneous induction of two enzymes sensitive to catabolite repression does not lead to an additive decrease of the specific activity of the two. Exogenously added cAMP increases the specific activity of catabolically repressed enzymes, irrespective of whether the enzyme is induced separately or simultaneously with another enzyme. In the presence of 12 different substrates metabolized by inducible enzymes glucose does not bring about catabolite repression. Many studies have examined positive feedbacks between invasive plant traits and nutrient cycling, but few have investigated whether feedbacks arise from introduction of pre-adapted species or from eco-evolutionary feedback that develops after introduction. Eco-evolutionary feedback could occur between an invader's leaf tissue C:N ratio and its response to litter accumulation. Previous modeling predicts that occurrence of this feedback would be reflected by: (1) field data showing higher litter:biomass ratios in the invasive range; (2) high C:N genotypes benefiting more from experimental litter additions than low C:N genotypes; (3) this beneficial effect on high C:N genotypes inducing a critical transition toward invader dominance when a critical amount of litter is added to a native species-dominated community experiencing low nutrient conditions. Here, we empirically tested these predictions for the invasive grass Phalaris arundinacea, which has undergone post-introduction evolutionary change toward attaining higher C:N ratios under high nutrient conditions. We performed a biogeographical comparison of litter:biomass ratios in the native (Europe) and invasive (USA) range, and an experiment with mesocosms from the invasive range under low nutrient conditions. Low and high C:N Phalaris genotypes were introduced into native-dominated and bare mesocosms, to which varying litter amounts were added. The biogeographical comparison revealed that litter:biomass ratios were higher in the invasive range. The mesocosm experiment showed that when grown in isolation, only high C:N genotypes responded positively to litter. This effect, however, was not strong enough to stimulate Phalaris when exposed to competition with native species. Our results suggest that eco-evolutionary feedback between Phalaris’ C:N ratio and litter accumulation could occur, but only under high nutrient conditions. Our experiments suggest that eco-evolutionary feedback may select for specialist rather than superior genotypes. Hence, genotypic variation induced by post-introduction admixture may be subject to context-dependent selection due to eco-evolutionary feedback, increasing trait variation within invasive populations. 相似文献

The poly-γ-d-glutamic acid capsule of Bacillus anthracis is a barrier to infection by B. anthracis-specific bacteriophages. A possible role for cAMP in the mechanism of ecdysteroid induction of morphogenesis in imaginal discs of Drosophila melanogaster was explored in a variety of experiments. Neither cAMP, supplied externally, nor theophylline at concentrations which increase internal cAMP levels 2–3 fold will substitute for the hormone in the induction of morphogenesis. Hormone action in imaginal discs is neither enhanced nor repressed by externally supplied cAMP or theophylline. Internal cAMP levels in discs, determined by kinase binding assays, are not altered by juvenile hormone or 20-hydroxy-ecdysone under incubation conditions which permit ecdysteroid stimulation of RNA synthesis and induction of morphogenesis. Adenylate cyclase activity in disc extracts is not stimulated or depressed by 20-hydroxy-ecdysone or juvenile hormone, but is stimulated by NaF. These findings suggest that ecdysteroid action in imaginal discs does not involve changes in the internal concentration or metabolism of cAMP. 相似文献
