Carrageenan yield, physicochemical properties, and antiviral activity of the carrageenan from Solieria chordalis (Rhodophyceae, Solieriaceae) harvested at the Brittany coast (France) were investigated. S. chordalis carrageenan was extracted by conventional and the microwave-assisted extraction (MAE) methods. The effect of different parameters during MAE extraction such as alkali concentration (0, 0.5 and 1% KOH), extraction time (10, 20, and 25 min) and temperature (90 and 105 °C) were evaluated. Native carrageenan extracted by MAE had the highest yield (29.3%) after 10 min at 90 °C. After alkali treatment, carrageenan yield ranged from 10.7 to 18.4%. No significant differences in the carrageenan yield were observed between MAE and conventional method under alkaline conditions. Chemical analysis and FTIR spectra revealed the presence of a predominant iota-carrageenan. Evaluation of the antiviral activity of S. chordalis carrageenan against HSV-1 (Herpes simplex virus type 1) showed a EC50 of the iota-carrageenans fractions in the range of 3.2 to 54.4 μg mL?1 (MOI 0.01 ID50 mL?1) without cytotoxicity in that range of concentrations.  相似文献

The Pichia pastoris expression system was used to produce functionalized single-chain antibody fragments (scFv) directed against the ED-B domain of the B-fibronectin (B-Fn) isoform which was found to be present only in newly formed blood vessels during tumor angiogenesis. Therefore, scFv antibody fragments recognizing the ED-B domain are potential markers for angiogenesis. We constructed four functionalized scFv antibody fragments for direct labeling with radioactive molecules or toxins or for attachment to liposomes serving as carriers for cytotoxic or antiangiogenic compounds. The C-termini of the scFv antibody fragments contain 1-3 cysteine residues that are separated by a hydrophilic linker (GGSSGGSSGS) from the binding domain and are accessible for site-specific functionalization with thiol-reactive reagents. Plasmid expression, culture conditions, and purification were optimized in 1-L cultures. The scFv antibody fragments were purified by anion exchange chromatography. The yields were 5-20 mg/L culture medium. The large-scale production of one scFv antibody fragment in a 3.7-L fermenter gave a yield of 60 mg. The reactivity of the cyteines was demonstrated by labeling with the thiol-reactive fluorescent dye ABD-F. The four scFv antibody fragments bound specifically to ED-B-modified Sepharose and binding was further confirmed by immunofluorescence on cell cultures using ED-B-positive human Caco-2 tumor cells. Furthermore, we could demonstrate specific binding of scFv-modified liposomes to ED-B-positive tumor cells. Our results indicate that the P. pastoris expression system is useful for the large-scale production of cysteine-functionalized alpha-ED-B scFv antibody fragments.  相似文献

The Top 10 fungal pathogens in molecular plant pathology   总被引:18,自引:0,他引:18  
The aim of this review was to survey all fungal pathologists with an association with the journal Molecular Plant Pathology and ask them to nominate which fungal pathogens they would place in a 'Top 10' based on scientific/economic importance. The survey generated 495 votes from the international community, and resulted in the generation of a Top 10 fungal plant pathogen list for Molecular Plant Pathology. The Top 10 list includes, in rank order, (1) Magnaporthe oryzae; (2) Botrytis cinerea; (3) Puccinia spp.; (4) Fusarium graminearum; (5) Fusarium oxysporum; (6) Blumeria graminis; (7) Mycosphaerella graminicola; (8) Colletotrichum spp.; (9) Ustilago maydis; (10) Melampsora lini, with honourable mentions for fungi just missing out on the Top 10, including Phakopsora pachyrhizi and Rhizoctonia solani. This article presents a short resumé of each fungus in the Top 10 list and its importance, with the intent of initiating discussion and debate amongst the plant mycology community, as well as laying down a bench-mark. It will be interesting to see in future years how perceptions change and what fungi will comprise any future Top 10.  相似文献

Summary Previous studies have suggested that gap junctions may have a role in various uterine functions, including parturition. Because nickel has been demonstrated to increase uterine contractility in vitro, the effect of nickel (II) chloride on gap junctional communication was assessed in a tumorigenic uterine cell line, SK-UT-1 (ATCC HTB 114). Cells were exposed in vitro to 25 and 50 μM NiCl2 for 24 h or 100 μM NiCl2 for 3, 12, and 24 h, then functional gap junctional communication was measured as the transfer of Lucifer yellow dye from microinjected donor cells to their primary neighbor cells. Dye transfer was significantly increased only in cell cultures exposed to 100 μM NiCl2 for 24h, compared to untreated controls, lower doses, and shorter exposure periods. This response was inhibited by the simultaneous co-treatment of SK-UT-1 cells with magnesium by adding 100 μM MgSO4 to the dosing medium. Possible mechanisms and implications for these findings are discussed.  相似文献

Disturbances affect almost all terrestrial ecosystems, but it has been difficult to identify general principles regarding these influences. To improve our understanding of the long-term consequences of disturbance on terrestrial ecosystems, we present a conceptual framework that analyzes disturbances by their biogeochemical impacts. We posit that the ratio of soil and plant nutrient stocks in mature ecosystems represents a characteristic site property. Focusing on nitrogen (N), we hypothesize that this partitioning ratio (soil N: plant N) will undergo a predictable trajectory after disturbance. We investigate the nature of this partitioning ratio with three approaches: (1) nutrient stock data from forested ecosystems in North America, (2) a process-based ecosystem model, and (3) conceptual shifts in site nutrient availability with altered disturbance frequency. Partitioning ratios could be applied to a variety of ecosystems and successional states, allowing for improved temporal scaling of disturbance events. The generally short-term empirical evidence for recovery trajectories of nutrient stocks and partitioning ratios suggests two areas for future research. First, we need to recognize and quantify how disturbance effects can be accreting or depleting, depending on whether their net effect is to increase or decrease ecosystem nutrient stocks. Second, we need to test how altered disturbance frequencies from the present state may be constructive or destructive in their effects on biogeochemical cycling and nutrient availability. Long-term studies, with repeated sampling of soils and vegetation, will be essential in further developing this framework of biogeochemical response to disturbance.  相似文献

Switchgrass (Panicum virgatum L.) is being developed as a biofuel feedstock for the United States. Efficient and accurate methods to estimate switchgrass biomass feedstock supply within a production area will be required by biorefineries. Our main objective was to determine the effectiveness of indirect methods for estimating biomass yields and composition of switchgrass fields. Indirect measurements were conducted in eastern Nebraska from 2003 to 2007 in which switchgrass biomass yields were manipulated using three nitrogen rates (0 kg N ha-1, 60 kg N ha-1, and 120 kg N ha-1) and two harvest periods (August and post-killing frost). A modified Robel pole was used to determine visual obstruction, elongated leaf height, and canopy height measurements. Prediction models from the study showed that elongated leaf height, visual obstruction, and canopy height measurements accounted for >?91%, >?90%, and >?82% of the variation in switchgrass biomass, respectively. Regression slopes were similar by cultivar (“Cave-in-Rock” and “Trailblazer”), harvest period, and across years indicating that a single model is applicable for determining biomass feedstock supply within a region, assuming similar harvesting methods. Sample numbers required to receive the same level of precision were as follows: elongated leaf height<canopy height<visual obstruction. Twenty to 30 elongated leaf height measurements in a field could predict switchgrass biomass yield within 10% of the mean with 95% confidence. Visual obstruction is recommended on switchgrass fields with low to variable stand densities while elongated leaf height measurements would be recommended on switchgrass fields with high, uniform stand densities. Incorporating an ocular device with a Robel pole provided reasonable frequency estimates of switchgrass, broadleaf weeds, and grassy weeds at the field scale.  相似文献

Profiles for the exoplasmic face (EF) of the freeze-fractured plasma membrane from the root storage tissue of red beets are reconstructed by microdensitometry of micrographs of surface-shadowed-platinum carbon replicas. Autocovariance functions (ACFs) are computed from those profiles. The initial portions of the ACFs have a Gaussian form whose parameters (root mean square surface roughness and autocovariance length) are estimated. The parameter estimates are used to show that the pits on the EF faces are in good complementarity with the intramembrane particles seen on the complementary protoplasmic fracture faces.  相似文献

This article gives an overview of the electrochemical biosensors that incorporate genetically modified enzymes. Firstly, the improvements on the sensitivity and selectivity of biosensors that integrate mutated enzymes are summarised. Next, new trends focused on the oriented immobilisation of mutated enzymes through specific functional groups located at their surface are reviewed.
