Four main molecular forms of acetylcholinesterase (AChE) can be solubilized from newborn rat superior cervical ganglia (SCG), homogenized in the presence of a high-ionic-strength, detergent-containing medium. These forms, respectively referred to as 16, 10, 6.5, and 4 S, are characterized by their sedimentation coefficients. Their relative proportions in SCG are notably different in vivo during postnatal maturation, and in culture. The 16-S AChE appears to be mainly neuronal in origin, is maintained in culture independently of original presynaptic in vivo elements, and its cellular pool is not depleted in the presence of tetrodotoxin (TTX).  相似文献

Previous studies revealed that whistled songs of starlings can be separated into different categories, among which five themes are found in the repertoires of all (or most) individuals. These “specific” themes show local variations but the different themes have different sizes of dialect areas. This study investigates the temporal evolution of this complex dialectal system. Observations were made in an area where dialects were mapped three years earlier. They reveal an obvious temporal evolution of dialects, although the degree of variation differs according to the theme. No structural change was observed for two themes (CC and ST), but some temporal dynamics in their dialect boundaries did appear. There were changes in the structures and boundaries of two other themes (IT and HT). However, they showed:
  • — a relationship between new and old structures, either with persistence of the old variants or with an apparent drift. Almost no really new structures were observed.
  • — all the 1980 variants recorded in 1983 were found at sites not far from the original one. The main changes consisted of extensions or regressions of variants along boundaries.
Lastly, the temporal evolution of RT dialects is unusual in that the changes are more sudden. The variants have been replaced by different ones in some cases, but in others they have remained stable. The complexity of the dialectal system is persistent on a temporal level, varying from stability for ST to sudden changes for RT, with IT and HT intermediate.  相似文献

Developmental aspects of the neuromuscular system in mouse embryos chronically paralyzed in utero with tetrodotoxin (TTX) between embryonic days 14 and 18 were studied using biochemical and histological methods. The number of lumbar spinal motoneurons (MNs) was higher in inactive embryos than in controls suggesting a decreased motoneuron cell death. In association with the increase in MN number, choline acetyltransferase activity was significantly increased in both spinal cord and peripheral synaptic sites. Paralyzed muscles exhibited a decreased number of mature myofibers and the nuclei were centrally located. Creatine kinase activity was greatly decreased and total acetylcholine receptor and receptor cluster numbers per myofiber were significantly increased in paralyzed muscles. A similar pattern of changes occurs in the neuromuscular system of the mutant mouse muscular dysgenesis (mdg). However, in contrast to the mdg mutant, tetrodotoxin-treated muscles were similar to controls in their innervation pattern, in the ultrastructural aspects of the excitation–contraction coupling system (i.e., dyads and triads) and in the extent of dihydropyridine binding. Thus, neuromuscular inactivity is not sufficient to impair the pattern of muscle innervation or the appearance of either the triadic junctions or dihydropyridine receptors. These results indicate that alterations of dihydropyridine binding sites and triads in muscular dysgenesis cannot be accounted for by inactivity but rather must reflect a more primary defect involving the structural gene(s) regulating the development of one or more aspects of muscle differentiation.  相似文献

A model is proposed for the population dynamics of an annual plant (Sesbania vesicaria) with a seed bank (i.e. in which a proportion of seeds remain dormant for at least one year). A simple linear matrix model is deduced from the life cycle graph. The dominant eigenvalue of the projection matrix is estimated from demographic parameters derived from field studies. The estimated values for population growth rate () indicates that the study population should be experiencing a rapid exponential increase, but this was not the case in our population.The addition of density dependent effects on seedling survivorship and adult fecundity, effects for which field studies provide evidence, considerably improves our model. Depending on the demographic parameters, the model leads to stable equilibrium, oscillations, or chaos. Study of the behaviour of this model in the parameter space shows that the existence of a seed bank allows higher among-year variation of adult fecundity, without leaving the region of demographic stability. Field data obtained over 3 years confirm this prediction.  相似文献

Our understanding of the gas exchange mechanisms in plant organs critically depends on insights in the three-dimensional (3-D) structural arrangement of cells and voids. Using synchrotron radiation x-ray tomography, we obtained for the first time high-contrast 3-D absorption images of in vivo fruit tissues of high moisture content at 1.4-microm resolution and 3-D phase contrast images of cell assemblies at a resolution as low as 0.7 microm, enabling visualization of individual cell morphology, cell walls, and entire void networks that were previously unknown. Intercellular spaces were always clear of water. The apple (Malus domestica) cortex contains considerably larger parenchyma cells and voids than pear (Pyrus communis) parenchyma. Voids in apple often are larger than the surrounding cells and some cells are not connected to void spaces. The main voids in apple stretch hundreds of micrometers but are disconnected. Voids in pear cortex tissue are always smaller than parenchyma cells, but each cell is surrounded by a tight and continuous network of voids, except near brachyssclereid groups. Vascular and dermal tissues were also measured. The visualized network architecture was consistent over different picking dates and shelf life. The differences in void fraction (5.1% for pear cortex and 23.0% for apple cortex) and in gas network architecture helps explain the ability of tissues to facilitate or impede gas exchange. Structural changes and anisotropy of tissues may eventually lead to physiological disorders. A combined tomography and internal gas analysis during growth are needed to make progress on the understanding of void formation in fruit.  相似文献

Ecological niche theory predicts that coexistence is facilitated by resource partitioning mechanisms that are influenced by abiotic and biotic interactions. Alternative hypotheses suggest that under certain conditions, species may become phenotypically similar and functionally equivalent, which invokes the possibility of other mechanisms, such as habitat filtering processes. To test these hypotheses, we examined the coexistence of the giant redfin Pseudobarbus skeltoni, a newly described freshwater fish, together with its congener Pseudobabus burchelli and an anabantid Sandelia capensis by assessing their scenopoetic and bionomic patterns. We found high habitat and isotope niche overlaps between the two redfins, rendering niche partitioning a less plausible sole mechanism that drives their coexistence. By comparison, environment–trait relationships revealed differences in species–environment relationships, making habitat filtering and functional equivalence less likely alternatives. Based on P. skeltoni's high habitat niche overlap with other species, and its large isotope niche width, we inferred the likelihood of differential resource utilization at trophic level as an alternative mechanism that distinguished it from its congener. In comparison, its congener P. burchelli appeared to have a relatively small trophic niche, suggesting that its trophic niche was more conserved despite being the most abundant species. By contrast, S. capensis was distinguished by occupying a higher trophic position and by having a trophic niche that had a low probability of overlapping onto those of redfins. Therefore, trophic niche partitioning appeared to influence the coexistence between S. capensis and redfins. This study suggests that coexistence of these fishes appears to be promoted by their differences in niche adaptation mechanisms that are probably shaped by historic evolutionary and ecological processes.  相似文献

Despite numerous advances in the identification of the molecular machinery for clathrin-mediated budding at the plasma membrane, the mechanistic details of this process remain incomplete. Moreover, relatively little is known regarding the regulation of clathrin-mediated budding at other membrane systems. To address these issues, we have utilized the powerful new approach of subcellular proteomics to identify novel proteins present on highly enriched clathrin-coated vesicles (CCVs). Among the ten novel proteins identified is the rat homologue of a predicted gene product from human, mouse, and Drosophila genomics projects, which we named enthoprotin. Enthoprotin is highly enriched on CCVs isolated from rat brain and liver extracts. In cells, enthoprotin demonstrates a punctate staining pattern that is concentrated in a perinuclear compartment where it colocalizes with clathrin and the clathrin adaptor protein (AP)1. Enthoprotin interacts with the clathrin adaptors AP1 and with Golgi-localized, gamma-ear-containing, Arf-binding protein 2. Through its COOH-terminal domain, enthoprotin binds to the terminal domain of the clathrin heavy chain and stimulates clathrin assembly. These data suggest a role for enthoprotin in clathrin-mediated budding on internal membranes. Our study reveals the utility of proteomics in the identification of novel vesicle trafficking proteins.  相似文献

Everything else being equal, insect herbivores can be expected to oviposit on host plants that provide the qualitatively and quantitatively best food for larvae. However, the selection of a plant for oviposition may be influenced by such ecological factors as natural enemies, host distribution, host patch size or host patch density. We performed a field study to test whether spatial proximity between two host plant species influences the oviposition patterns and larval distribution of the alpine leaf beetle Oreina elongata. In the population studied, O. elongata oviposits and feeds on two host plants, that belong to the same family (Asteraceae): Adenostyles alliariae and Cirsiumspinosissimum. The first species contains pyrrolizidine alkaloids that are sequestered by the beetle as a chemical defence, whereas the second plant does not contain any alkaloids but has hairy and spiny leaves that might give some mechanical protection to beetle larvae.
During two consecutive summers, we quantified oviposition and larval distribution on randomly chosen C. spinosissimum that grew spatially isolated from A. alliariae, on C. spinosissimum that grew in leaf contact with A. alliariae and on A. alliariae that grew in leaf contact with C. spinosissimum (isolated A. alliariae was not considered, because it is rare in the study population). In both years, more eggs were laid on C. spinosissimum than on A. alliariae and more on those C. spinosissimum that were growing close to A. alliariae than on those growing isolated. Large numbers of larvae moved from C. spinosissimum to A. alliariae during the season. Patch size did not influence egg and larval numbers. Eggs survived better on C. spinosissimum than on A. alliariae in the field. The data suggest that C. spinosissimum may provide eggs with better protection against stormy weather. In a separate study of the same population, we found that larval performance was better on A. alliariae than on C. spinosissimum. Our present data suggest that O. elongata preferentially oviposits on plants of the species that maximizes egg survival and that grow in close proximity to plants of the species that provides better food and chemical defence.  相似文献
