S100A1 is a member of the Ca2+-binding S100 protein family. It is expressed in brain and heart tissue, where it plays a crucial role as a modulator of Ca2+ homeostasis, energy metabolism, neurotransmitter release, and contractile performance. Biological effects of S100A1 have been attributed to its direct interaction with a variety of target proteins. The (patho)physiological relevance of S100A1 makes it an important molecular target for future therapeutic intervention. S-Nitrosylation is a post-translational modification of proteins, which plays a role in cellular signal transduction under physiological and pathological conditions. In this study, we confirmed that S100A1 protein is endogenously modified by Cys85
S-nitrosylation in PC12 cells, which are a well established model system for studying S100A1 function. We used isothermal calorimetry to show that S-nitrosylation facilitates the formation of Ca2+-loaded S100A1 at physiological ionic strength conditions. To establish the unique influence of the S-nitroso group, our study describes high resolution three-dimensional structures of human apo-S100A1 protein with the Cys85 thiol group in reduced and S-nitrosylated states. Solution structures of the proteins are based on NMR data obtained at physiological ionic strength. Comparative analysis shows that S-nitrosylation fine tunes the overall architecture of S100A1 protein. Although the typical S100 protein intersubunit four-helix bundle is conserved upon S-nitrosylation, the conformation of S100A1 protein is reorganized at the sites most important for target recognition (i.e. the C-terminal helix and the linker connecting two EF-hand domains). In summary, this study discloses cysteine S-nitrosylation as a new factor responsible for increasing functional diversity of S100A1 and helps explain the role of S100A1 as a Ca2+ signal transmitter sensitive to NO/redox equilibrium within cells. 相似文献

The commonly used preclinical animal model of postmenopausal osteoporosis is the mature ovariectomized rat, whereby cessation of ovarian oestrogen production consequently results in bone volume reduction. The study aim was to precisely define the time course of structural changes resulting from ovariectomy and thereby reduce the time animals have to be treated to judge the effects of osteoporosis treatment. For this purpose, we assessed architectural changes by microcomputed tomography (μCT) during 10 weeks following ovariectomy or sham surgery at two-week intervals. Moreover, the trabecular microarchitecture of the lumbar vertebrae was assessed after necropsy. Besides this, serum biomarkers of bone turnover were determined. These data were in a new approach additionally correlated to femur mRNA expression profiles. We selected the osteoblast marker genes osteocalcin and type I collagen as well as the two osteoclast marker genes cathepsin k and tartrate-resistant acid phosphatase 5. The gene expression analysis suggested an activation of osteoblasts as well as octeoclasts. The significantly induced serum levels of osteocalcin and collagen degradation fragments also revealed this higher rate of bone turnover. Our results indicate that as soon as four weeks after ovariectomy the bone volume fraction exhibited a decline of 30% and 50% of the connectivity density. In addition, significant decreases of trabecular number and thickness as well as of the bone volume fraction were only observed in vertebrae of ovariectomized animals. Purpose
Promoting the economic and environmental performance of Swiss farms is a major objective of Swiss agricultural policy. In the present paper, we investigate the relationship between the economic and global environmental performance of the Swiss dairy farms in the alpine area.Methods
The analysis relies on a sample of 56 dairy farms from the Farm Accountancy Data Network, for which highly precise and comprehensive Life Cycle Assessments have been carried out. The work income per family work unit is used as indicator of the economic performance of a farm. The so-called global environmental performance of a farm is, for its part, measured by means of an eco-efficiency indicator for each environmental impact considered (demand for non-renewable energy resources, eutrophication potential, aquatic ecotoxicity potential, human toxicity potential and land use) and of an aggregate eco-efficiency indicator assessed using a Data Envelopment Analysis-based approach. The relationship between economic and global environmental performance is assessed by means of non-parametric Spearman??s rank correlation analysis.Results
The results of the analysis reveal the existence of a positive relationship between economic and global environmental performance. This positive relationship exists for all environmental issues considered and thus also for the aggregate eco-efficiency indicator. Its strength, however, substantially varies from one issue to another.Conclusions
This study provides the evidence that there is no trade-off between economic and global environmental farm performance. When they improve their economic performance, farms also tend to improve their global environmental performance and vice versa. This finding is of central relevance for policy-makers as it should contribute to improving the acceptance among farmers of the environmental objectives of Swiss agricultural policy in terms of an increase in environmental resource use productivity. During human pregnancy, placental trophoblasts differentiate and syncytialize into syncytiotrophoblasts that sustain progesterone production [1]. This process is accompanied by mitochondrial fragmentation and cristae remodeling [2], two facets of mitochondrial apoptosis, whose molecular mechanisms and functional consequences on steroidogenesis are unclear. Here we show that the mitochondria-shaping protein Optic atrophy 1 (Opa1) controls efficiency of steroidogenesis. During syncytialization of trophoblast BeWo cells, levels of the profission mitochondria-shaping protein Drp1 increase, and those of Opa1 and mitofusin (Mfn) decrease, leading to mitochondrial fragmentation and cristae remodeling. Manipulation of the levels of Opa1 reveal an inverse relationship with the efficiency of steroidogenesis in trophoblasts and in mouse embryonic fibroblasts where the mitochondrial steroidogenetic pathway has been engineered. In an in vitro assay, accumulation of cholesterol is facilitated in the inner membrane of isolated mitochondria lacking Opa1. Selenium analogues (4b-4h, and 4j) of two known sulfur compounds were synthesized and tested their anticancer activities. The selenium compound 4b had comparable activity with its sulfur analogue 4a, while DNA-binding study showed these two compounds had similar interaction with ct-DNA, the K(b) was 8.23 and 2.36, respectively. The primary results showed that most compounds had moderate anticancer activities with IC(50) values between 10(-6) and 10(-5) M. Another selenium analogue 4j showed the highest activity with the IC(50) values around 5.3 μM against K562 and MCF-7 cell lines. More importantly, the organochalcogen compounds exhibited stronger anticancer activities against K562 cell line than the other cell lines tested. 相似文献

During developmental and tumor angiogenesis, semaphorins regulate blood vessel navigation by signaling through plexin receptors that inhibit the R-Ras subfamily of small GTPases. R-Ras is mainly expressed in vascular cells, where it induces adhesion to the extracellular matrix (ECM) through unknown mechanisms. We identify the Ras and Rab5 interacting protein RIN2 as a key effector that in endothelial cells interacts with and mediates the pro-adhesive and -angiogenic activity of R-Ras. Both R-Ras-GTP and RIN2 localize at nascent ECM adhesion sites associated with lamellipodia. Upon binding, GTP-loaded R-Ras converts RIN2 from a Rab5 guanine nucleotide exchange factor (GEF) to an adaptor that first interacts at high affinity with Rab5-GTP to promote the selective endocytosis of ligand-bound/active β1 integrins and then causes the translocation of R-Ras to early endosomes. Here, the R-Ras/RIN2/Rab5 signaling module activates Rac1-dependent cell adhesion via TIAM1, a Rac GEF that localizes on early endosomes and is stimulated by the interaction with both Ras proteins and the vesicular lipid phosphatidylinositol 3-monophosphate. In conclusion, the ability of R-Ras-GTP to convert RIN2 from a GEF to an adaptor that preferentially binds Rab5-GTP allows the triggering of the endocytosis of ECM-bound/active β1 integrins and the ensuing funneling of R-Ras-GTP toward early endosomes to elicit the pro-adhesive and TIAM1-mediated activation of Rac1. 相似文献

Aims
Nutrients play a key role in arbuscular mycorrhizal (AM) symbiosis. We quantified the response of AM symbiosis of seedlings and adult plants of Plantago lanceolata to fertilization under field conditions in managed grasslands differing in nutrient availability and soil moisture.Methods
The AM symbiosis was measured as the total extent of AM fungal colonization and frequency of arbuscules or vesicles, and as the relative proportions of morphotypes. We further examined the effects of the surrounding vegetation upon AM symbiosis.Results
Fertilization decreased total AM colonization and relative arbuscular frequency of the whole mycorrhizal community and of Acaulospora and “fine endophyte” morphotypes in seedling roots, but it had no effect upon the mycorrhiza in adult plants. The decline in arbuscular frequency in seedling roots due to fertilization was greater at the sites with higher nutrient availability and lower N:P ratio. Seedlings surrounded by more forbs had a greater total AM colonization and higher vesicular frequency.Conclusions
Increased nutrient availability in the initial stages of seedling development has a prominent effect upon AM symbiosis development, but these effects seem to diminish over the long term, as evidenced by the results obtained for adult plants and from the limited effects of parameters characterizing long-term nutrient availability. 相似文献1000.
