Summary Genes for the major storage protein of potato, patatin, have been mapped genetically and physically in both the potato and tomato genomes. In potato, all patatin genes detected by the cDNA clone pGM01 map to a single locus at the end of the long arm of chromosome 8. By means of pulsed field gel electrophoresis (PFGE) it was possible further to delimit this locus, containing 10–15 copies of the gene, to a maximum size of 1.4 million base pairs. Hybridizations with class-specific clones suggest that the locus is at least partially divided into domains containing the two major types of patatin genes, class I and II. In tomato, patatin-homologous sequences were found to reside at the orthologous locus at the end of chromosome 8. The approximately three copies in tomato were localized by PFGE to a single fragment of 300 kilobases. Whereas the class II-specific 5 promoter sequences reside in tomato at the same locus as the coding sequences, the single class I-specific copy of the 5 promoter sequences was localized on chromosome 3 with no coding sequence attached to it. A clone from this chromosome 3 locus of tomato was isolated and by restriction fragment length polymorphism mapping it could be further shown that a similar class I-specific sequence also exists on chromosome 3 of potato. As in tomato, this copy on chromosome 3 is not linked to a coding sequence for patatin. The results are discussed with respect to genome evolution and PFGE analysis of complex gene families. 相似文献

A discrete-time, age-independent SIR-type epidemic model is formulated and analyzed. The effects of vaccination are also included in the model. Three mathematically important properties are verified for the model: solutions are nonnegative, the population size is time-invariant, and the epidemic concludes with all individuals either remaining susceptible or becoming immune (a property typical of SIR models). The model is applied to a measles epidemic on a university campus. The simulated results are in good agreement with the actual data if it is assumed that the population mixes nonhomogeneously. The results of the simulations indicate that a rate of immunity greater than 98% may be required to prevent an epidemic in a university population. The model has applications to other contagious diseases of SIR type. Furthermore, the simulated results of the model can easily be compared to data, and the effects of a vaccination program can be examined. 相似文献

Buffer-extractable proteins from leaves of Spinacia oleracea L. were separated by non-denaturing polyacrylamide gel electrophoresis. Gels were stained for adenosine diphosphoglucose (ADPglucose)-dependent glucan-synthase (GS) activity (EC 2.4.1.21). Three major forms of activity were observed. No staining was detectable when ADPglucose was replaced by an equimolar concentration of either uridine, guanosine or cytosine diphosphoglucose. Two of the three GS forms exhibited both primed and citrate-stimulated unprimed activity whereas one enzyme form was strictly dependent upon the presence of an exogenous glucan. For intracellular localization, mesophyll protoplasts and intact chloroplasts were isolated and their enzyme pattern was compared with that of the leaf extract. Intactness and purity of the chloroplast preparations were ascertained by polarographic measurement of the ferricyanide- or CO2-dependent oxygen evolution, by determination of marker-enzyme activities, and by electrophoretic evaluation of the content of chloroplast- and cytosol-specific glucanphosphorylase forms (EC 2.4.1.1). The three GS forms were present in mesophyll protoplasts. Intact chloroplasts possessed both primer-independent enzyme forms but lacked the primer-dependent one. The latter form was enriched in supernatant fractions of leaf homogenates when the intact chloroplasts had been pelleted by centrifugation. Thus, in spinach-leaf mesophyll cells soluble ADPglucose-dependent GS is located both inside and outside the chloroplast.Abbreviations GS

The social environment affects both behavioral and physiological responses to separation from the mother. Less information
is available on the impact of the social environment on the response to separation in peer-reared infant monkeys. This study
reports the responses of peer-reared pigtail macaque infants to repeated separations, and the impact of social versus isolation
housing during the separation. The responses of two pairs of monkeys were studied during four three-day separations. One of
each pair was housed in isolation during the separation, and the other was with another pair of peers, with whom they had
been living for one month prior to the separation. The isolation-housed peer responded to the separation with behavioral agitation,
but no depression. The socially-housed peer's behavior did not differ from baseline during the separation. During successive
reunions, all the separated monkeys, regardless of housing condition, exhibited declining levels of behaviors related to maintaining
proximity to their attachment figure. Although the number of subjects is small, the results suggest that the presence of social
support, in the form of a familiar peer, can ameliorate the response to separation, and that with repeated separations the

Fully adult partial skeletons attributed to Australopithecus afarensis (AL 288-1, “Lucy”) and to Homo habilis (OH 62, “Lucy's child”), respectively, both include remains from upper and lower limbs. Relationships between various limb bone dimensions of these skeletons are compared to those of modern African apes and humans. Surprisingly, it emerges that OH 62 displays closer similarities to African apes than does AL 288-1. Yet A. afarensis, whose skeleton is dated more than 1 million years earlier, is commonly supposed to be the ancestor of Homo habilis. If OH 62, classified as Homo habilis by its discoverers, does indeed represent a stage intermediate between A. afarensis and later Homo, a revised interpretation of the course of human evolution would be necessary. 相似文献

Summary
Drosophila imaginal disc cell lines show a characteristic pattern of aggregation in culture, which appears to be due to cell-cell rather than cell-substrate interactions. We have examined the distribution of PS integrins in wing and leg cell lines, and find that these integrin homologues are expressed preferentially in aggregates. Populations ofTococa occidentalis (Melastomataceae) and the inhabiting ants (Myrmelachista sp.) were observed for more than eight months in the Peruvian Amazon (Sira mountains). They represent a complex coevolutionary system: the plants offer shelter (leaf domatia, hollow stems) and food (leaf glands), whereas the ants kill all surrounding plants, including large trees up to 10 m, by chemical weapons. Experiments with exposed plants revealed a highly specialized way to attack meristematic tissue and leaf nervature, which leads to a quick decay of the plant individuals. The clearing of the vegetation by the ants allows theTococa population to expand mostly by vegetative shoots to large monocultures (up to 30 m in diameter) free from any other plant species. Artificially introduced plant individuals, from differentT. occidentalis populations, are regarded as a foreign species by the ants.The succession of such aTococa-Myrmelachista system begins with one or a few founder plants on a light place in the midst of the vegetation.Myrmelachista soon inhabits their host plants which otherwise would not survive and begin to clear the place from all foreign plant species.Tococa expands quickly, forming circle shaped populations. Distantly situated canopy trees shade theTococa population after a number of years and cause their decay. The whole place appears contaminated for years and no other plant can establish itself.
