Quinones are well established as key players in the production of reactive oxygen species within cellular environments. Many factors govern their cytotoxicity but most studies have been restricted to a few, core, derivatives. A new strategy for the in situ production of quinone derivatives has been developed such that libraries of diverse functionality can be rapidly created without recourse to extensive synthetic procedures. The approach relies upon nucleophilic addition by reduced thiol derivatives to the quinone core within a pre-culture assay mixture and provides a generic strategy that exploits the large reservoir of commercial thiols currently available. A readily accessible chromatographic method has been developed that allows the derivatisation process to be easily monitored and the purity of the resulting one pot preparation to be assessed. The viability of the combinatorial approach has been fully validated through comparison with a range of quinone-S-conjugates prepared using conventional bench synthesis. Metabotropic glutamate receptor type 5 (mGluR5) modulates dopamine and glutamate neurotransmission at central synapses. In this study, we addressed the role of mGluR5 in l-DOPA-induced dyskinesia, a movement disorder that is due to abnormal activation of both dopamine and glutamate receptors in the basal ganglia. A selective and potent mGluR5 antagonist, 3-[(2-methyl-1,3-thiazol-4-yl)ethynyl] pyridine, was tested for its ability to modulate molecular, behavioural and neurochemical correlates of dyskinesia in 6-hydroxydopamine-lesioned rats treated with l-DOPA. The compound significantly attenuated the induction of abnormal involuntary movements (AIMs) by chronic l-DOPA treatment at doses that did not interfere with the rat physiological motor activities. These effects were paralleled by an attenuation of molecular changes that are strongly associated with the dyskinesiogenic action of l-DOPA (i.e. up-regulation of prodynorphin mRNA in striatal neurons). Using in vivo microdialysis, we found a temporal correlation between the expression of l-DOPA-induced AIMs and an increased GABA outflow within the substantia nigra pars reticulata. When co-administered with l-DOPA, 3-[(2-methyl-1,3-thiazol-4-yl)ethynyl] pyridine greatly attenuated both the increase in nigral GABA levels and the expression of AIMs. These data demonstrate that mGluR5 antagonism produces strong anti-dyskinetic effects in an animal model of Parkinson's disease through central inhibition of the molecular and neurochemical underpinnings of l-DOPA-induced dyskinesia.  相似文献

  • 1 Blue whale locations in the Southern Hemisphere and northern Indian Ocean were obtained from catches (303 239), sightings (4383 records of ≥8058 whales), strandings (103), Discovery marks (2191) and recoveries (95), and acoustic recordings.
  • 2 Sighting surveys included 7 480 450 km of effort plus 14 676 days with unmeasured effort. Groups usually consisted of solitary whales (65.2%) or pairs (24.6%); larger feeding aggregations of unassociated individuals were only rarely observed. Sighting rates (groups per 1000 km from many platform types) varied by four orders of magnitude and were lowest in the waters of Brazil, South Africa, the eastern tropical Pacific, Antarctica and South Georgia; higher in the Subantarctic and Peru; and highest around Indonesia, Sri Lanka, Chile, southern Australia and south of Madagascar.
  • 3 Blue whales avoid the oligotrophic central gyres of the Indian, Pacific and Atlantic Oceans, but are more common where phytoplankton densities are high, and where there are dynamic oceanographic processes like upwelling and frontal meandering.
  • 4 Compared with historical catches, the Antarctic (‘true’) subspecies is exceedingly rare and usually concentrated closer to the summer pack ice. In summer they are found throughout the Antarctic; in winter they migrate to southern Africa (although recent sightings there are rare) and to other northerly locations (based on acoustics), although some overwinter in the Antarctic.
  • 5 Pygmy blue whales are found around the Indian Ocean and from southern Australia to New Zealand. At least four groupings are evident: northern Indian Ocean, from Madagascar to the Subantarctic, Indonesia to western and southern Australia, and from New Zealand northwards to the equator. Sighting rates are typically much higher than for Antarctic blue whales.
  • 6 South‐east Pacific blue whales have a discrete distribution and high sighting rates compared with the Antarctic. Further work is needed to clarify their subspecific status given their distinctive genetics, acoustics and length frequencies.
  • 7 Antarctic blue whales numbered 1700 (95% Bayesian interval 860–2900) in 1996 (less than 1% of original levels), but are increasing at 7.3% per annum (95% Bayesian interval 1.4–11.6%). The status of other populations in the Southern Hemisphere and northern Indian Ocean is unknown because few abundance estimates are available, but higher recent sighting rates suggest that they are less depleted than Antarctic blue whales.

Question: Is ombrotrophic bog vegetation in an oceanic region of southwestern Sweden changing in the same direction over a five year period (1999 ‐ 2004) as northwest European bogs in the last 50 years, i.e. towards drier and more eutrophic vegetation? Location: The province of Halland, southwestern Sweden. Methods: Changes in species composition were monitored in 750 permanently marked plots in 25 ombrotrophic bogs from 1999 to 2004. Changes in species occurrences and richness were analysed and a multivariate statistical method (DCA) was used to analyse vegetation changes. Results: The species composition changed towards wetter rather than drier conditions, which is unlike the general pattern of vegetation change on bogs in northwestern Europe. Species typical of wetter site conditions including most Sphagnum species increased in abundance on the bogs until 2004. The total number of species per plot increased, mostly due to the increased species richness of Sphagnum species. Nitrogen‐demanding (eutrophic) species increased in occurrence. Conclusions: Ombrotrophic bog vegetation in an oceanic region in Sweden became wetter and was resilient to short‐term climatic shifts, after three years of below normal precipitation followed by several years with normal precipitation levels. Shifts towards more nitrogen demanding species were rapid in this region where the deposition levels have been high for several decades.  相似文献

The need for contraception in the successful management of captive wild animals is becoming increasingly apparent. Because concerns exist regarding the reversibility of the contraceptive implant melengestrol acetate (MGA), reproductive data for 94 female Amur (Panthera tigris altaica) and Sumatran tigers (Panthera tigris sumatrae) were analyzed using survival analyses to evaluate return to reproductive status after implant removal or assumed expiration. Females placed in potential breeding situations after MGA implants were surgically removed showed a 62% return to reproduction by 5.25 years, whereas females with implants that were assumed to have expired showed only a 30% return to reproduction by 6 years. Implanted females did not reproduce as successfully as non‐implanted control females, which showed an 85% probability of reproducing after placement in a new breeding situation by 2.66 years. Parturition increased the probability of reproducing in non‐implanted females, but not in implanted females. Litter size, stillbirths, and offspring survival were not significantly different between non‐implanted, implant‐removed and implant‐expired female tigers. Ten female tigers reproduced both before and after implant placement, and the differences in litter size, stillbirths, and offspring survival were not significant, nor were they significantly different from non‐implanted females. Prior parturition, age when implant was removed, and duration of implantation did not affect the probability of reproducing for females after implant removal. These results show substantial reversibility of MGA implants, leading to 62% successful reproduction after implant removal. The reasons for lower successful reproduction in animals previously treated with the contraceptive compared to non‐implanted females are not known, but a greater delay in reversibility was seen when implants were left in place and only presumed expired. Species abundance distributions (SADs) follow one of ecology's oldest and most universal laws – every community shows a hollow curve or hyperbolic shape on a histogram with many rare species and just a few common species. Here, we review theoretical, empirical and statistical developments in the study of SADs. Several key points emerge. (i) Literally dozens of models have been proposed to explain the hollow curve. Unfortunately, very few models are ever rejected, primarily because few theories make any predictions beyond the hollow-curve SAD itself. (ii) Interesting work has been performed both empirically and theoretically, which goes beyond the hollow-curve prediction to provide a rich variety of information about how SADs behave. These include the study of SADs along environmental gradients and theories that integrate SADs with other biodiversity patterns. Central to this body of work is an effort to move beyond treating the SAD in isolation and to integrate the SAD into its ecological context to enable making many predictions. (iii) Moving forward will entail understanding how sampling and scale affect SADs and developing statistical tools for describing and comparing SADs. We are optimistic that SADs can provide significant insights into basic and applied ecological science.  相似文献

1 Integrated management of crop pests requires the identification of the appropriate spatial scale at which colonization processes occurs. We assessed, by coupling demographic and genetic methods, the relative contribution of local and transient migrants of the grain aphid Sitobion avenae to wheat field colonization in spring. 2 We examined, during two consecutive years, the daily colonization of wheat by aphid migrants and compared this with daily aphid flight monitored by a local 12.2‐m suction trap. The genetic profiles of aphids landing on crops were compared with those of both flying aphids caught by the suction trap and local populations from arable crops and hedgerows. 3 In the first year, we observed: (i) a strong correlation between aphids colonizing the crop and those moving within the crop and a close genetic similarity between aphids from these samples and (ii) a high level of genetic differentiation between these aphids and populations from local cereals and field margins. In the second year, the number of migrants recorded on the wheat was three‐fold higher than in the previous year, and less correlated with that recorded by the suction trap. This was associated with a lack of genetic differentiation between all samples. 4 This variation in the colonization processes resulted mainly in an abrupt increase in abundance of genotypes from local over‐wintering sites in 2004. This suggests that, despite the long range dispersal potential of the grain aphid, outbreak risks could be mainly determined at a local scale, encouraging the design of relatively small management units.  相似文献

Interpretation of the enigmatic soft‐bodied yunnanozoans from the Lower Cambrian (Stage 3, Series 2) Chengjiang Biota has remained controversial for decades because of their strange body plan and their variable taphonomic alteration. Proposed affinities have ranged from stem bilaterian to stem vertebrate. A study of over seven hundred slabs, many newly collected, from five sections at two localities demonstrates that yunnanozoans have sclerotized dorsal and axial segments, a body cavity, a unique feeding region and a coiled alimentary canal. Although two genera have been previously discriminated, all specimens have the same number of filamentous arches (seven pairs) and of ventral circular structures (four pairs); this indicates that Haikouella Chen, Huang and Li, 1999 is a junior synonym of Yunnanozoon Hou, Ramsköld and Bergström, 1991. Our analysis reveals new details of the putative pharyngeal pores of Yunnanozoon lividum, and although there are similarities with the gill slits of deuterostomes, the question of their homology remains moot. The filamentous arches and their supporting rods were probably sclerotized and represent a specialized feeding structure. Yunnanozoans exhibit characters that are open to a number of alternative interpretations of putative homology, encompassing comparisons with deuterostomes and protostomes (including members of the Platyzoa). Given their unique features, and the current lack of secure homologies, the affinities of yunnanozoans should continue to be considered in a wider bilaterian context.  相似文献

Conflicts between forest management and nature protection This study gives an overview of the change of the German flora over time with special emphasis on species growing in forest habitats. As a consequence of land use plant species numbers have continually increased. Land management has also maintained some endangered plant species. The plant diversity of managed forests is higher than that of unmanaged forests. Not a single obligate forest species has gone extinct over the past 250 years of forest management, the time span of formal records. For organisms with special habitat requirements alternative protection measures other than non‐management are needed to maintain these habitats.  相似文献
