Zusammenfassung Pnoepyga immaculata n. sp. lebt im Areal der beiden anderenPnoepyga-Artenalbiventer (Hodgson 1837) undpusilla Hodgson 1845 und wurde durch ihren markant abweichenden Territorialgesang entdeckt. Sie besitzt eine kurze silberhelle Strophe aus kurzen, in gleichmäßigen Abständen gereihten Pfiff-Elementen, die in der Frequenz leicht abfällt. Neben der Stimme liegen die wesentlichen diagnostischen Merkmale nach jetzigem Kenntnisstand gegenüber sympatrischenalbiventer in wenig größerem Schnabel, der Fleckenlosigkeit von Oberseite, Kopf und Flügeldecken, gegenüberpusilla in den größeren Körperabmessungen, gegenüber sympatrischenalbiventer undpusilla im schwachen grünlichen Schimmer der Oberseite und in der länglichen Schuppung der Unterseite. Es bestehen keine Proportionsunterschiede zwischenimmaculata undalbiventer/pusilla einerseits wie auch nicht zwischen diesen beiden andererseits. Bis jetzt wurden 4 Belegstücke (SammlungenKoelz, Diesselhorst, Martens) und eine durch Bandaufnahme gesicherte Beobachtung zwischen Dhaulagiri in Mittelwest-Nepal bis nahe der Ost-Grenze Nepals bekannt.P. immaculata lebt zur Brutzeit in enger Nachbarschaft mitP. albiventer undP. pusilla, ist jedoch vonalbiventer vertikal getrennt bei nachgewiesenem Kontakt, vonpusilla durch Bevorzugung trockeneren Waldunterwuchses abseits von Bächen. Schon jetzt mußimmaculata als gefährdete Art eingestuft werden, da Waldvegetation in der zur Brutzeit bevorzugten Höhenstufe zwischen 2100 und 3100 m in Nepal weitgehend vernichtet worden ist.
Pnoepyga immaculata n. sp., a new ground living wren-babbler from the Nepal Himalayas (Timaliidae)
Summary P. immaculata n. sp. was discovered by its voice which strongly differs from the two other sympatric species,albiventer (Hodgson 1837) andpusilla Hodgson 1845. As a territorial song, it displays a silvery strophe arranged of short, more or less regularly spaced whistling notes (fig. 7a–d), slightly descending in pitch. The strophe is about 2 s long. Besides the distinct voice, the essential diagnostic characters in comparison toP. albiventer are the slightly larger bill, plain head, upper side and wing coverts (fig. 1a–d, 2, 4a–d), in comparison topusilla the larger body size (fig. 1i–m), in comparison to sympatricalbiventer andpusilla the slightly olive tinge of the upper side (against warm dark brown in both others, fig. 1e–m), and the more longish scaly feathers of the lower side (against stout scaly appearance in both others, fig. 1 a–d). These characters hold true at least in Central and Eastern Himalayan populations of the respective species. There do not exist proportional differences in the relations of any part of the body betweenimmaculata andpusilla and not between the latter ones as well. Heretofore, 4 museum specimens are known (Koelz Coll.,Diesselhorst Coll.,Martens Coll.) from West-central (Thakkhola, type locality) and East Nepal (Ting Sang La), 3 of them from the presumed breeding grounds (2100–3100 m), one from the winter quarters in the Terai lowlands. A field observation close to the Darjeeling border in far East Nepal is verified by a tape-recording.P. immaculata embarasses the biologist and systematist because it lives in close neighbourhood of the similar speciesalbiventer andpusilla. It must have been overlooked by its apparent scarcity and overall similarity to both other species of the genus. Slight ecological differences rather correspond to the vertical distribution ofalbiventer, which populates a higher forest belt. But both have already been found on territory at close range.P. pusilla inhabits different microbiotopes, especially the close proximity of running water in the same altitudinal belt asimmaculata. When discovered,P. immaculata is to be classified already as an endangered species. Forest vegetation of its preferred altitude is already greatly reduced in the Central and Eastern Nepal Himalayas. To get a more detailed information on its general distribution and biology in order to quickly arrange protected areas and thus to guarantee the survival ofP. immaculata, should be the urgent next step.

The death rate of hybridoma cells, grown in a continuous culture, has been studied in a small air-lift loop reactor as a function of reactor height and injected gas flow rate. The first-order death-rate constant was found to be proportional to the reciprocal height and to the gas flow rate, in accordance with the hypothetical killing volume model for insect cells in bubble columns. Furthermore, the effect of the serum concentration on viable cell concentration and cell productivity has been investigated in a continuous culture. A serum component became growth limiting when the serum concentration was decreased from 2% to 1%. No effect of the serum concentration on specific cell productivity could be measured. Samples from this culture were also studied in the air-lift loop reactor to determine the effect of serum concentration on the shear sensitivity. The cells' shear sensitivity increased with decreasing serum concentration. The protective effect of serum was found to be physical as well as physiological.  相似文献

Tryptophan-dependent biosynthesis of auxins in soil   总被引:1,自引:0,他引:1  
The presence of auxins in soil may have an ecological impact affecting plant growth and development. A rapid and simple colorimetric method was used to assess California soils for their potential to produce auxins upon the addition of L-tryptophan (L-TRP). The auxin content measured by colorimetry was expressed as indole-3-acetic acid (IAA)-equivalents. A substrate (L-TRP) concentration of 5.3 g kg-1, glucose concentration of 6.7 g kg-1, no nitrogen, pH 7.0, 40°C, shaking (aeration) and 48 h incubation time were selected as standardized conditions to assay for auxin biosynthesis in soil. IAA was confirmed as a major microbial metabolite derived from L-TRP in soil by use of high performance liquid chromatography (HPLC). Under standardized conditions, L-TRP-derived auxins in 19 soils varied greatly ranging from 18.2 to 303.2 mg IAA equivalents (auxins) kg-1 soil. This study suggests that the phenotypic character of the soil microbiota has more of an influence on auxin production than the soil physicochemical properties (e.g., pH, organic C content, CEC, etc.).  相似文献

An isocratic high-performance liquid chromatographic (HPLC) method with ultraviolet detection is described for the quantification of the atypical neuroleptic clozapine and its major metabolites, N-desmethylclozapine and clozapine N-oxide, in human serum or plasma. The method included automated solid-phase extraction on C18 reversed-phase material. Clozapine and its metabolites were separated by HPLC on a C18 ODS Hypersil analytical column (5 μm particle size; 250 mm × 4.6 mm I.D.) using an acetonitrile—water (40:60, v/v) eluent buffered with 0.4% (v/v) N,N,N′,N′-tetramethylethylenediamine and acetic acid to pH 6.5. Imipramine served as internal standard. After extraction of 1 ml of serum or plasma, as little as 5 ng/ml of clozapine and 10 or 20 ng/ml of the metabolites were detectable. Linearity was found for drug concentrations between 5 and 2000 ng/ml as indicated by correlation coefficients of 0.998 to 0.985. The intra- and inter-assay coefficients of variation ranged between 1 and 20%. Interferences with other psychotropic drugs such as benzodiazepines, antidepressants or neuroleptics were negligible. In all samples, collected from schizophrenic patients who had been treated with daily oral doses of 75–400 mg of clozapine, the drug and its major metabolite, N-desmethylclozapine, could be detected, while the concentrations of clozapine N-oxide were below 20 ng/ml in three of sixteen patients. Using the method described here, data regarding relations between therapeutic or toxic effects and drug blood levels or metabolism may be collected in clinical practice to improve the therapeutic efficacy of clozapine drug treatment.  相似文献

Glutamine synthetase (GS) of Rhodopseudomonas sphaeroides is regulated by adenylylation and deadenylylation. The extent of adenylylation/deadenylylation of the enzyme in cell free extracts was influenced by inorganic phosphate (P i), -ketoglutarate, ATP and other nucleotides. While P i and -ketoglutarate stimulated deadenylylation, ATP and other nucleotides enhanced adenylylation of the GS. By using proper combinations of the effectors and incubation conditions, any desired adenylylation state of GS could be adjusted in vitro. The enzyme was purified to electrophoretic homogenity by three steps including affinity chromatography on 5-AMP-Sepharose. Adenylylated and deadenylylated enzyme showed different UV-spectra and isoelectric points. The native enzyme had a molecular weight of 600,000, deadenylylated subunits of 50,000±1,000. Electron microscopic investigations revealed a dodecameric arrangement of subunits in two hexameric planes.  相似文献

From May 1979 on, the following parameters were measured at a station in the inlet of Königshafen near List (Island of Sylt): temperature, salinity, mesozooplankton (>76 µm), chlorophyll-a, seston dry weight, oxygen and phytoplankton-nutrients (NH4-N, NO2-N, NO3-N, PO4-P, SiO3-Si). A multiple regression analysis showed the interrelationships between the parameters measured. Tidal influences on zooplankton and seston dry weight could be observed. At low tide, the amount of zooplankton (not counting the harpacticoid copepods) declines and the number of harpacticoid copepods rises as does the seston dry weight too. The chlorophyll-a content is a function of the phytoplankton-nutrients. An increase in chlorophyll-a leads to a decrease in nitrogen and silicate concentrations. Phosphate, due possibly to a sewage inlet into the Königshafen, is not a limiting factor. The availability of nutrients is influenced by temperature, salinity and the tidal cycle. The amount of oxygen is dependent on water temperature and seston dry weight. High water temperatures and a high seston content lead to a decrease in oxygen concentrations.  相似文献

DNAs from 16 species of archaebacteria including 6 novel isolates were hybridized with 16S rRNAs from 7 species representing different orders or groups of the urkingdom of archaebacteria. The yields, normalized for the number of genes per microgram of DNA, and the temperature stabilities of all hybrids were determined and related to each other. A taxonomic tree constructed from such fractional stability data reveals the same major divisions as that derived from comparative cataloging of 16S rRNA sequences. The extreme halophiles appear however as a distinct order besides the three known divisions of methanogens. The methanogens, the halophiles and Thermoplasma form one of two clearly recognizable branches of the archaebacterial urkingdom. The order represented by Sulfolobus and the related novel order Thermoproteales form the other branch. Three novel genera, Thermoproteus, Desulfurococcus and the "stiff filaments" represent three families of this order. The extremely thermophilic methanogen Methanothermus fervidus belongs to the Methanobacteriales. SN1, a methanogen from Italy, appears as another species of the genus Methanococcus. Another novel methanogen, M3, represents a genus or family of the order Methanomicrobiales.  相似文献

Phospholipid metabolism was studied in rat sciatic nerve during Wallerian degeneration induced by crush injury. Portions of crushed sciatic nerve, incubated with labeled substrates, showed significantly higher phosphatidylcholine synthesis than normal nerve, prior to any measurable alterations of phospholipid composition. Maximum synthesis occurred 3 days after crush injury, at which time the metabolism of other phospholipids was unchanged. After a rapid decrease in biosynthetic activity, a second phase of enhanced phosphatidylcholine synthesis occurred, beginning 6 days after crush injury. Increased incorporation of [33P]phosphate, [2-3H]glycerol, and [Me-14C]choline indicated stimulation of de novo synthesis of phosphatidylcholine 3 days after injury. Neither base exchange reactions nor sequential methylation of ethanolamine phospholipids contributed significantly to phosphatidylcholine synthesis. Assay of certain key enzymes under optimal conditions in subcellular fractions of sciatic nerve revealed higher activities of cholinephosphate cytidyltransferase, choline phosphotransferase, and acyl-CoA:lysophosphatidylcholine acyltransferase in injured nerve, while choline kinase activity remained unchanged. This indicates that stimulation of phosphatidylcholine synthesis occurs via the cytidine nucleotide pathway, as well as by increased acylation of lysophosphatidylcholine. Although the cause of stimulated phosphatidylcholine synthesis remains unexplained, it is possible that trace amounts of lysophospholipids or other metabolites produced by injury-enhanced phospholipase activity may be responsible.  相似文献

Abstract: The composition and metabolism of rat sciatic nerve phospholipids were studied 20 weeks after induction of chronic diabetes by intraperitoneal injection of streptozotocin (50 mg/kg). On a wet weight basis the nerves from the diabetic animals showed a 7% decrease in total phospholipid from that of controls and a relative decrease in phosphatidylinositol. Incubations of isolated sciatic nerves of diabetic rats in a medium containing [33P]orthophosphate gave decreased labeling of phosphatidylinositol and substantial changes in the labeling pattern of phosphatidylinositol phosphate and 4,5-bisphosphate from that of controls. The ratio of label in these polyphosphoinositides decreased from 2.5 for normal nerve to about 1.0 for diabetic nerve within a 2-h incubation period. These metabolic alterations were not observed in acutely diabetic animals 5 days after streptozotocin (100 mg/kg) administration. Because polyphosphoinositides may be involved in the control of membrane permeability during axonal conduction, alterations in their relative amounts or turnover rates could be related to the physiological changes of early diabetic neuropathy.  相似文献
