The development of rust after administering allopurinol, a specific inhibitor of xanthine oxidoreductase, via roots was studied at the histological level in leaves of susceptible‘Pinto 111’bean plants inoculated with Uromyces phaseoli and‘Thatcher',‘Mentana’and‘Leopardo’wheat plants challenged with Puccinia recondita. A marked reduction and delay in fungal growth was observed in allopurinol-treated plants starting between 24 h and 48 h post-inoculation, i.e. after differentiation of the first haustoria (onset of the biotrophic plant-parasite relationship). Infection hyphae often grew twisted and convoluted in treated hosts, sometimes producing small, irregularly shaped colonies. Differentiation of subepidermal stromata in fungal colonies was delayed and restricted by the treatment and uredospore yield severely reduced. Allopurinol administration also tended to increase the proportion of haustoria which became embedded in thick translucent sheaths during the late stages of infection. These results support the view that plant xanthine oxidoreductase activity is necessary for biotrophic development of rust fungi and suggest that the inhibition of this enzyme, which impairs the pathogen metabolism, may favour some natural host responses to attack such as haustorial sheath formation. 相似文献

Human activity has more than doubled the amount of nitrogen entering the global nitrogen cycle, and the boreal forest biome is a nitrogen‐limited ecosystem sensitive to nitrogen load perturbation. Although bryophyte‐associated microbes contribute significantly to boreal forest ecosystem function, particularly in carbon and nitrogen cycling, little is known about their responses to anthropogenic global change. Amplicon pyrosequencing of the ITS2 region of rDNA was used to investigate how fungal communities associated with three bryophyte species responded to increased nitrogen loads in a long‐term fertilization experiment in a boreal Picea abies forest in southern Norway. Overall, OTU richness, community composition and the relative abundance of specific ecological guilds were primarily influenced by host species identity and tissue type. Although not the primary factor affecting fungal communities, nitrogen addition did impact the abundance of specific guilds of fungi and the resulting overall community composition. Increased nitrogen loads decreased ectomycorrhizal abundance, with Amphinema, Cortinarius, Russula and Tylospora OTUs responding negatively to fertilization. Pathogen abundance increased with fertilization, particularly in the moss pathogen Eocronartium. Saprophytic fungi were both positively and negatively impacted by the nitrogen addition, indicating a complex community level response. The overshadowing of the effects of increased nitrogen loads by variation related to host and tissue type highlights the complexity of bryophyte‐associated microbial communities and the intricate nature of their responses to anthropogenic global change. 相似文献

Background
Alexithymia, or “no words for feelings”, is a personality trait which is associated with difficulties in emotion recognition and regulation. It is unknown whether this deficit is due primarily to regulation, perception, or mentalizing of emotions. In order to shed light on the core deficit, we tested our subjects on a wide range of emotional tasks. We expected the high alexithymics to underperform on all tasks.Method
Two groups of healthy individuals, high and low scoring on the cognitive component of the Bermond-Vorst Alexithymia Questionnaire, completed questionnaires of emotion regulation and performed several emotion processing tasks including a micro expression recognition task, recognition of emotional prosody and semantics in spoken sentences, an emotional and identity learning task and a conflicting beliefs and emotions task (emotional mentalizing).Results
The two groups differed on the Emotion Regulation Questionnaire, Berkeley Expressivity Questionnaire and Empathy Quotient. Specifically, the Emotion Regulation Quotient showed that alexithymic individuals used more suppressive and less reappraisal strategies. On the behavioral tasks, as expected, alexithymics performed worse on recognition of micro expressions and emotional mentalizing. Surprisingly, groups did not differ on tasks of emotional semantics and prosody and associative emotional-learning.Conclusion
Individuals scoring high on the cognitive component of alexithymia are more prone to suppressive emotion regulation strategies rather than reappraisal strategies. Regarding emotional information processing, alexithymia is associated with reduced performance on measures of early processing as well as higher order mentalizing. The grey-sided vole (Clethrionomys rufocanus) is distributed over the entire island of Hokkaido, Japan, across which it exhibits multi-annual density cycles in only parts of the island (the north-eastern part); in the remaining part of the island, only seasonal density changes occur. Using annual sampling of 189 grey-sided vole populations, we deduced the geographical structure in their second-order density dependence. Building upon our earlier suggestion, we deduce the seasonal density-dependent structure for these populations. Strong direct and delayed density dependence is found to occur during winter, whereas no density dependence is seen during the summer period. The direct density dependence during winter may be seen as a result of food being limited during that season: the delayed density dependence during the winter is consistent with vole-specialized predators (e.g. the least weasel) responding to vole densities so as to have a negative effect on the net growth rate of voles in the following year. We conclude that the observed geographical structure of the population dynamics may be properly seen as a result of the length of the summer in interaction with the differential seasonal density-dependent structure. Altogether, this indicates that the geographical pattern in multi-annual density dynamics in the grey-sided vole may be a result of seasonal forcing. 相似文献

In this paper we describe the isolation and characterization of four monoclonal antibodies (FRP5, FSP16, FWP51, and FSP77) which specifically recognize the human erbB-2 protein. All of the antibodies recognize epitopes on the extracellular domain of the receptor protein. FRP5 and FSP16 compete with one another for binding while FWP51 and FSP77 each recognize a different epitope. The effects of the antibodies on the erbB-2 receptor protein have been analyzed. Two different erbB-2-expressing cell lines, SKBR3 breast tumor cells and HC11 R111 cells, were examined. The SKBR3 cells express approximately 1 x 10(6) molecules of the erbB-2 protein/cell; HC11 R111 cells, a clone of mouse mammary epithelial cells derived by transfection of a human erbB-2 expression plasmid, contain 10-fold less erbB-2 protein than the SKBR3 cells. Treatment of the two cell lines with FRP5, FSP16, and FWP51 led to a rapid increase in the phosphotyrosine content of the erbB-2 protein. Three of the antibodies, FRP5, FSP16, and FSP77, stimulated the turnover of the erbB-2 protein. Binding of the antibodies did not stimulate DNA synthesis in HC11 R111 cells. Thus, the erbB-2-specific monoclonal antibodies behave as partial ligand agonists. The antibodies were examined for their effects upon the growth of SKBR3 and HC11 R111 cells. The growth of SKBR3 cells was inhibited by 90% following long term treatment of the cells with FSP77. 相似文献
