Solanum elaeagnifolium Cav. fruits contain high concentrations of steroidal saponins. Treatment of 3-day-old clover seedlings with aqueous fruit extracts modified Ca2+ uptake without significantly altering K+ and H2 PO4 − uptake. The extracts increased Ca2+ uptake in the concentration range of 0.2 to 20 m M Ca2+ . Uptake curves could be represented by two phases. In the lower phase (0.2-1.0 m M Ca2+ ), this change could be related to an increase in Vmax . Pretreatment of seedlings with saponin extracts significantly reduced ATP-dependent Ca2+ uptake and Ca2+ -dependent ATPase activity in a fraction isolated from root homogenates by centrifugation at 1500 g for 15 min. Saponins purified from S. eleagnifolium extracts by thin-layer chromatography modified in vitro the Ca2+ -ATPase activity of this fraction, indicating that the steroid may act directly on Ca2+ transport across membranes. 相似文献

Leukemic stem cells (LSCs) reside within bone marrow niches that maintain their relatively quiescent state and convey resistance to conventional treatment. Many of the microenvironmental signals converge on RAC GTPases. Although it has become clear that RAC proteins fulfill important roles in the hematopoietic compartment, little has been revealed about the downstream effectors and molecular mechanisms. We observed that in BCR-ABL-transduced human hematopoietic stem/progenitor cells (HSPCs) depletion of RAC2 but not RAC1 induced a marked and immediate decrease in proliferation, progenitor frequency, cobblestone formation and replating capacity, indicative for reduced self-renewal. Cell cycle analyses showed reduced cell cycle activity in RAC2-depleted BCR-ABL leukemic cobblestones coinciding with an increased apoptosis. Moreover, a decrease in mitochondrial membrane potential was observed upon RAC2 downregulation, paralleled by severe mitochondrial ultrastructural malformations as determined by automated electron microscopy. Proteome analysis revealed that RAC2 specifically interacted with a set of mitochondrial proteins including mitochondrial transport proteins SAM50 and Metaxin 1, and interactions were confirmed in independent co-immunoprecipitation studies. Downregulation of SAM50 also impaired the proliferation and replating capacity of BCR-ABL-expressing cells, again associated with a decreased mitochondrial membrane potential. Cinclotaenia sp., described originally by Georgiev & Genov (1985) from the dipper Cinclus cinclus (L.) in Bulgaria, has recently been identified from the same host in the Carpathian Mountains in the Slovak Republic. This tapeworm is considered to be a new species, which is named C. georgievi n. sp. It is characterised by: a scolex armed with 23-27 (predominantly 24-26) hooks in two rows; hooks 30.5-36 microm long, with a blade 10-13.5 microm long and resembling in shape the diorchoid hooks of hymenolepidids; irregularly alternating genital pores with simple genital atria; a slightly conical cirrus armed by small spines of up to 3 microm in length; 24-51 testes posterior to a bi-alate, branched ovary; a gravid uterus filled with egg packets; and eggs with filaments. C. georgievi n. sp. differs from the closely-related C. tarnogradskii (Dinnik, 1927) in the slightly higher number of rostellar hooks, which have longer blades, and a larger cirrus. 相似文献

Contribution of three regions (phosphate-binding, 50’s and 90’s loops) of Anabaena apoflavodoxin to FMN binding and reduction potential was studied. Thr12 and Glu16 did not influence FMN redox properties, but Thr12 played a role in FMN binding. Replacement of Trp57 with Glu, Lys or Arg moderately shifted Eox/sq and Esq/hq and altered the energetic of the FMN redox states binding profile. Our data indicate that the side chain of position 57 does not modulate Eox/sq by aromatic stacking or solvent exclusion, but rather by influencing the relative strength of the H-bond between the N(5) of the flavin and the Asn58-Ile59 bond. A correlation was observed between the isoalloxazine increase in solvent accessibility and less negative Esq/hq. Moreover, Esq/hq became less negative as positively charged residues were added near to the isoalloxazine. Ile59 and Ile92 were simultaneously mutated to Ala or Glu. These mutations impaired FMN binding, while shifting Esq/hq to less negative values and Eox/sq to more negative. These effects are discussed on the bases of the X-ray structures of some of the Fld mutants, suggesting that in Anabaena Fld the structural control of both electron transfer steps is much more subtle than in other Flds. 相似文献

Production of official statistics frequently requires expert judgement to evaluate and reconcile data of unknown and varying quality from multiple and potentially conflicting sources. Moreover, exceptional events may be difficult to incorporate in modelled estimates. Computational logic provides a methodology and tools for incorporating analyst''s judgement, integrating multiple data sources and modelling methods, ensuring transparency and replicability, and making documentation computationally accessible. Representations using computational logic can be implemented in a variety of computer-based languages for automated production. Computational logic complements standard mathematical and statistical techniques and extends the flexibility of mathematical and statistical modelling. A basic overview of computational logic is presented and its application to official statistics is illustrated with the WHO & UNICEF estimates of national immunization coverage. 相似文献

This study aimed to determine whether patients with aseptic and bacterial meningitis presented alterations in oxidative stress
parameters of cerebrospinal fluid (CSF). A total of 30 patients were used in the research. The CSF oxidative stress status
has been evaluated through many parameters, such as lipid peroxidation through thiobarbituric acid reactive substances (TBARS)
and antioxidant defense systems such as superoxide dismutase (SOD), glutathione S-transferase (GST), reduced glutathione (GSH) and ascorbic acid. TBARS levels, SOD and GST activity increase in aseptic meningitis
and in bacterial meningitis. The ascorbic acid concentration increased significantly in patients with both meningitis types.
The reduced glutathione levels were reduced in CSF of patients with aseptic and bacterial meningitis. In present study we
may conclude that oxidative stress contributes at least in part to the severe neurological dysfunction found in meningitis. 相似文献
